The approach involves four levels of testing that utilize both in vitro and in vivo methods. Treatment of cultured Hodgkin disease (HD) cells with neuraminidase results in decreased reactivity of monoclonal antibody VIM-D5 with its antigen, the X hapten, a fucosyl-N-acetyllactosamine. It is assumed that styrene is a prohapten metabolized in the skin by aryl hydrocarbon hydroxylase (AHH) to styrene epoxide acting as the true hapten. Diclofenac (DCF) is widely used in veterinary medicine, but its milk residues may pose risks to human health. This study designed DCF haptens using computer modeling and developed an anti-DCF monoclonal antibody (mAb) with milk samples, achieving a semi-inhibitory concentration of 0.379 ng/mL. The development of a procedure for the selection of a large number of functional antigen-binding B cells is described. Forty to 70% of the cells in the enriched population bind antigen, and the antigen-binding can be inhibited by pretreatment with either the free hapten (TNP-Lys) or anti-immunoglobulin. The enriched cells express both slgM and slgD and respond to mitogenic stimuli by proliferating and developing into antibody-forming cells. In contrast to normal spleen cells, however, the enriched cells also proliferate in response to submitogenic levels of TNP-Brucella abortus and TNP-LPS.

A novel approach for the production of [125I]amatoxin and anti-amanitin is described. The antigen and the starting material for Bolton Hunter iodination is prepared by periodic acid oxidation of the gamma-delta-dihydroxy-isoleucine side chain of alpha-amanitin followed by reductive amination. Antigenicity of terazosin was studied in the experimental animals and the following results were obtained. Terazosin was shown to be non-immunogenic in guinea-pigs and mice when immunized alone or with mixture of terazosin and protein as immunogen.
