Assessing the combined toxicity of conventional and newer insecticides on the cotton mealybug Phenacoccus solenopsis.

Reduced susceptibility to insecticides often results in failure of insect pest control and repetitive use of broad-spectrum insecticides, which could have detrimental effects on beneficial arthropods and surrounding agro-ecosystems. The cotton mealybug, Phenacoccus solenopsis Tinsley is a pest of worldwide importance that can be effectively controlled using a number of insecticides. This insect has developed resistance due to injudicious use and repeated exposure to insecticides throughout the year. The aim of the present study was to investigate the toxicity of the insecticides chlorpyrifos,

Bemisia tabaci Q carrying tomato yellow leaf curl virus strongly suppresses host plant defenses.

The concurrence of tomato yellow leaf curl virus (TYLCV) with the spread of its vector Bemisia tabaci Q rather than B in China suggests a more mutualistic relationship between TYLCV and Q. Here, we investigated the hypothesis that viruliferous B and Q have different effects on plant defenses. We found the fecundity of nonviruliferous B, nonviruliferous Q, viruliferous Q and viruliferous B was 11.080, 12.060, 10.760, and 11.220 respectively on plants previously attacked by the other biotype, however, on their respective noninfested control leaves fecundity was 12.000, 10.880, 9.760, and 8.020 r

Food assimilated by two sympatric populations of the brown planthopper Nilaparvata lugens (Delphacidae) feeding on different host plants contaminates insect DNA detected by RAPD-PCR analysis.

Contamination of insect DNA for RAPD-PCR analysis can be a problem because many primers are non-specific and DNA from parasites or gut contents may be simultaneously extracted along with that of the insect. We measured the quantity of food ingested and assimilated by two sympatric populations of brown planthopper (BPH), Nilaparvata lugens, one from rice and the other from Leersia hexandra (Poaceae), a wetland forage grass, and we also investigated whether host plant DNA contaminates that of herbivore insects in extractions of whole insects. Ingestion and assimilation of food were reduced signi

Exploitative competition between invasive herbivores benefits a native host plant.

Although biological invasions are of considerable concern to ecologists, relatively little attention has been paid to the potential for and consequences of indirect interactions between invasive species. Such interactions are generally thought to enhance invasives' spread and impact (i.e., the "invasional meltdown" hypothesis); however, exotic species might also act indirectly to slow the spread or blunt the impact of other invasives. On the east coast of the United States, the invasive hemlock woolly adelgid (Adelges tsugae, HWA) and elongate hemlock scale (Fiorinia externa, EHS) both feed on

Bemnisia tabaci feeding induces pathogenesis-related proteins in cassava (Manihot esculenta Crantz).

Cassava (Manihot esculenta Cranzts) plants fed upon by whitefly Bemisia tabaci showed increased levels of pathogenesis-related (PR) proteins, such as beta-1, 3-glucanase, peroxidase and chitinase activities, as compared to uninfested plants. The enzymes increased in specific activities from 2 to 7 fold and protein content in leaf extracts decreased in whitefly-infested plants, compared to uninfested plants. Among the three PR proteins, B. tabaci feeding induced significantly higher beta-1, 3-glucanase activities, when compared with other two PR proteins. Study also discussed the possible appli

The role of CYP4Gs involved in hydrocarbon formation in the survival, reproduction, and pyrethroid resistance of Triatoma infestans, a major Chagas disease vector.

Very long-chain normal and branched hydrocarbons are essential for insect physiology and ecology. The final metabolic step of hydrocarbon formation involves an oxidative decarbonylation carried out by cytochrome P450 from family 4, subfamily G (CYP4G). Triatoma infestans Klug (Hemiptera: Reduviidae: Triatominae) has two CYP4G genes, named CYP4G106 and CYP4G107. In this study, we have silenced in T. infestans the expression of both genes by RNA interference in order to assess the relevance of hydrocarbons to survival, reproductive physiology, and insecticide susceptibility. Gene knockdown led t…

Improving the Efficiency and Safety of Sentinel Stink Bug Eggs Using X-rays.

Sentinel eggs used to monitor field parasitism of stink bug pests (Hemiptera: Pentatomidae) can only be deployed for a few days to avoid releasing the pest in the monitored area. Using sterile eggs removes the risk of accidental pest introduction and extends deployment time. Freezing the eggs before deployment is one common method of sterilizing sentinel eggs. However, some egg parasitoid species have low or no parasitism on frozen eggs. In this study, X-ray irradiation was used to sterilize Bagrada hilaris sentinel eggs intended for monitoring parasitism by Gryon aetherium (Hymenoptera: Sceli…

Comparative Transcriptome Analysis Reveals Genetic Mechanisms of Sugarcane Aphid Resistance in Grain Sorghum.

The sugarcane aphid, Melanaphis sacchari (Zehntner) (Hemiptera: Aphididae) (SCA), has become a major pest of grain sorghum since its appearance in the USA. Several grain sorghum parental lines are moderately resistant to the SCA. However, the molecular and genetic mechanisms underlying this resistance are poorly understood, which has constrained breeding for improved resistance. RNA-Seq was used to conduct transcriptomics analysis on a moderately resistant genotype (TAM428) and a susceptible genotype (Tx2737) to elucidate the molecular mechanisms underlying resistance. Differential expression …

RNAi-Mediated Knockdown of Tektins Does Not Affect Male Fertility in Lygus hesperus.

Mirid plant bugs (Hemiptera: Miridae), including Lygus hesperus (western tarnished plant bug), are key pests of numerous agricultural crops. While management of this pest relies heavily on chemical insecticides, the evolution of resistance and environmental concerns underscore the need for new and more effective approaches. Genetic-based strategies that target male fertiliy are currently being evaluated for population suppression. However, a lack of candidate gene targets with appropriate function, specifically in non-model species like L. hesperus, has hindered progress in the development and…

Molecular characterization and evolution of a chemosensory receptor gene family in three notorious rice planthoppers, Nilaparvata lugens, Sogatella furcifera and Laodelphax striatellus, based on genome and transcriptome analyses.

BACKGROUND: The white-backed planthopper (WBPH) Sogatella furcifera, the brown planthopper (BPH) Nilaparvata lugens, and the small brown planthopper (SBPH) Laodelphax striatellus (Hemiptera: Delphacidae) are rice pests that damage rice plants by sap-sucking and by transmitting viruses. Host-seeking behavior involves chemosensory receptor genes that include odorant receptors (ORs), ionotropic receptors (IRs) and gustatory receptors (GRs). RESULTS: We used genome and transcriptome data to identify 141 ORs, 28 GRs and 25 IRs in BPH; 135 ORs, 18 GRs and 16 IRs in WBPH; and 37 ORs, 14 GRs and 6 IR…

Role of syrphid larvae and other predators in suppressing aphid infestations in organic lettuce on California's Central Coast.

Organic lettuce, Lactuca sativa L., growers on the Central Coast of California rely on conservation biological control to manage Nasonovia ribisnigri Mosley (Hemiptera: Aphididae) and other aphid pests of lettuce. In 2006, we carried out five replicated field trials to determine the importance of syrphid larvae in the suppression of N. ribisnigri and other aphids infesting organic romaine lettuce. We used Entrust, a spinosad-based insecticide approved for use on organic farms, to suppress syrphid larvae in aphid-infested romaine. Romaine treated with Entrust was unmarketable at harvest because…

Metabolome profiling reveals the resistance mechanisms of rice against brown planthopper.

Rice (Oryza sativa L.) is a crucial global cereal crop, severely threatened by the brown planthopper (BPH), which causes extensive damage through direct feeding and virus transmission. To elucidate the resistance mechanism mediated by the BPH-resistance gene Bph6, metabolic responses to BPH infestation were compared between the resistant rice line G6 and the susceptible cultivar Nipponbare using LC-MS-based metabolomics combined with quantitative real-time PCR analysis. The Bph6 gene conferred both antixenotic and antibiotic resistance to BPH. A total of 673 metabolites were identified across

Insect salivary effectors disrupt PIEZO1-centric mechanoimmunity against piercing-sucking vectors.

Hosts deploy mechanosensory systems to intercept herbivory threats, yet how plants decode insect-derived mechanical force remains unclear. Here, we demonstrate that PIEZO-type mechanosensitive ion channel component 1 (PIEZO1) serves as a mechanoimmune hub converting whitefly probing into calcium-dependent defense activation. Mechanical force during feeding activates PIEZO1-dependent Ca2+ influx, triggering CBP60g phosphorylation that amplifies both PIEZO1 transcription and salicylic acid (SA) biosynthesis, forming a self-amplifying defense loop. Furthermore, PIEZO1 positively modulates the cor

Vector-enabled metagenomics reveals the first detection of the geminivirus beet curly top Iran virus in Europe.

Geminiviruses are among the most threatening emerging insect-borne viruses and are responsible for serious outbreaks worldwide. Climate change (i.e. higher temperatures) could further exacerbate their impact on crops, highlighting the need for new diagnostic approaches to manage potentially dangerous situations. vector-enabled metagenomics (VEM) exploits the natural ability of highly mobile insects to accumulate viruses acquired from plants over time and space within an ecosystem; this approach is effective for monitoring the presence of new invasive or indigenous viruses in large areas. Wolbachia enhances ovarian development in the rice planthopper Laodelphax striatellus through elevated energy production.

The endosymbiont Wolbachia can both benefit host nutrition and manipulate host reproduction to its own advantage. However, the mechanisms of its nutritional benefits remain unclear. We show that Wolbachia enhances ovarian development in the small brown planthopper Laodelphax striatellus by boosting energy production. Wolbachia-infected females have increased fecundity, accelerated ovarian development, and prolonged oviposition. Enhanced activity of mitochondrial complex I is linked to increased ATP production and the expression of energy metabolism-related genes. We further identify that Wolba

Magnaporthe oryzae infection triggers rice resistance to brown planthopper through the influence of jasmonic acid on the flavonoid biosynthesis pathway.

In agroecosystems, plants are constantly exposed to attack from diverse herbivorous insects and microbes, and infestation with one species may change the plant defense response to other species. In our investigation of the relationships among rice plants, the brown planthopper Nilaparvata lugens (Stål) and the rice blast fungus Magnaporthe oryzae, we observed a significant increase in the resistance of rice treated with rice blast to N. lugens, as evidenced by improved plant survival rates in a small population resistance study. Subsequent transcriptome data analysis revealed that the rice bla
