Therapeutic application of histone deacetylase inhibitors for central nervous system disorders.

Histone deacetylases (HDACs)--enzymes that affect the acetylation status of histones and other important cellular proteins--have been recognized as potentially useful therapeutic targets for a broad range of human disorders. Pharmacological manipulations using small-molecule HDAC inhibitors--which may restore transcriptional balance to neurons, modulate cytoskeletal function, affect immune responses and enhance protein degradation pathways--have been beneficial in various experimental models of brain diseases. Although mounting data predict a therapeutic benefit for HDAC-based therapy, drug di…

Histone acetylation and flagellin are essential for Legionella pneumophila-induced cytokine expression.

Legionella pneumophila causes severe pneumonia. Acetylation of histones is thought to be an important regulator of gene transcription, but its impact on L. pneumophila-induced expression of proinflammatory cytokines is unknown. L. pneumophila strain 130b induced the expression of the important chemoattractant IL-8 and genome-wide histone modifications in human lung epithelial A549 cells. We analyzed the IL-8-promoter and found that histone H4 was acetylated and H3 was phosphorylated at Ser(10) and acetylated at Lys(14), followed by transcription factor NF-kappaB. Recruitment of RNA polymerase …

The histone deacetylase HDAC4 connects neural activity to muscle transcriptional reprogramming.

Neural activity actively regulates muscle gene expression. This regulation is crucial for specifying muscle functionality and synaptic protein expression. How neural activity is relayed into nuclei and connected to the muscle transcriptional machinery, however, is not known. Here we identify the histone deacetylase HDAC4 as the critical linker connecting neural activity to muscle transcription. We found that HDAC4 is normally concentrated at the neuromuscular junction (NMJ), where nerve innervates muscle. Remarkably, reduced neural input by surgical denervation or neuromuscular diseases dissoc

Involvement of histone H1.2 in apoptosis induced by DNA double-strand breaks.

It is poorly understood how apoptotic signals arising from DNA damage are transmitted to mitochondria, which release apoptogenic factors into the cytoplasm that activate downstream destruction programs. Here, we identify histone H1.2 as a cytochrome c-releasing factor that appears in the cytoplasm after exposure to X-ray irradiation. While all nuclear histone H1 forms are released into the cytoplasm in a p53-dependent manner after irradiation, only H1.2, but not other H1 forms, induced cytochrome c release from isolated mitochondria in a Bak-dependent manner. Reducing H1.2 expression enhanced

MECP2 truncating mutations cause histone H4 hyperacetylation in Rett syndrome.

Rett syndrome (RTT) is a mostly sporadic disorder of developmental regression, with loss of speech and purposeful hand use, microcephaly and … n antibody against the C-terminal region of MeCP2. Western blots with antibodies against acetylated histones H3 and H4 revealed that H4, but not H3, was hyperacetylated. By using antibodies against individual acetylated lysine residues, the observed H4 hyperacetylation was attributed to increased acetylation of lysine 16. Therefore, expression of endogenou …

The protective capacities of histone H1 against experimental murine cutaneous leishmaniasis.

In a murine model of experimental cutaneous leishmaniasis, we investigated the protection elicited by injection of histone H1 isolated from parasites by perchloric extraction, of a H1 recombinant protein produced in E. coli, and of H1 long and short synthetic peptides, against infection by L. major. Partial protection was achieved in most of the animals as shown by reduction in lesion size, upon immunization with histone H1 or its peptides, provided that the region 1-60 was present in the molecule. These observations argue in favor of a thorough examination of the possibility of including hist

Characterization of anti-histone antibodies in patients with type 1 autoimmune hepatitis.

We have recently found that antibodies to total histones are common in a group of American patients with type 1 autoimmune hepatitis (AIH). In an attempt to determine the profile and clinical association of anti-histone antibody (AHA), 45 Japanese AIH patients were studied for serum isotypic reactivity with individual histones (H1, H2A, H2B, H3, H4) by enzyme-linked immunosorbent assay and western blotting. The results revealed that 40% of sera had reactivities with at least one of individual histones and that the antibodies were detected in all three classes of immunoglobulins (IgG, IgM, IgA)…

Monoclonal anti-histone autoantibodies derived from murine models of lupus.

We characterized three monoclonal antibodies with histone reactivity which were derived from spleen cells obtained from unmanipulated NZB/NZW or MRL/1 mice. By using an enzyme-linked immunosorbent assay, we noted that all three antibodies reacted with chromatin histones as well as with total histones extracted from chromatin. None of the antibodies appeared to require DNA as part of the antigen. One antibody (BWH-1) recognized a determinant present in the nucleosome core H2A-H2B complex but showed little reactivity with any of the individual histones (H1, H2A, H2B, H3, or H4). In contrast, the…

Histone modifications in benzo[a]pyrene carcinogenicity and toxicity: mechanisms, implications, and prospects.

Benzo[a]pyrene (B[a]P), a widespread environmental carcinogen, exerts multifaceted toxicity beyond direct DNA damage. Recent studies highlight the profound capacity of B[a]P to disrupt epigenetic landscapes, particular histone modifications, across diverse biological systems. This review synthesizes both in vitro and in vivo experimental findings to elucidate the mechanisms and consequences of B[a]P-induced histone modifications. In vitro studies demonstrate that B[a]P and its ultimate metabolite, BPDE, significantly alter key histone marks, including phosphorylation (γH2AX), acetylation (H3K9

Influence of Histone Acetylation on Chromatin 3D Organization in Drosophila S2 Cells.

Alterations in histone acetylation are frequently detected in various cancers and neurological diseases. Histone acetylation is known to disrupt both intra- and inter-nucleosomal interactions, but its influence on higher-order chromatin organization has been studied to a lesser extent. In this study, we changed global levels of histone acetylation in Drosophila S2 cells by treatment with trichostatin A (TSA; an inhibitor of histone deacetylases) or with curcumin (an inhibitor of histone acetyltransferases and histone deacetylases). We also performed a knockdown of the main histone deacetylase

Plant histone variants: deposition mechanisms, functional diversification, and translational prospects.

Histone variants are non-allelic isoforms of canonical histones that assemble into nucleosomes via replication-independent pathways, reshaping chromatin biophysical properties and transcriptional control at specific genomic loci. Constrained by their sessile lifestyle, plants have evolved a rich set of histone variants, including lineage-specific innovations such as heterochromatin-stabilizing H2A.W, stress-inducible H1.3, gametophyte-restricted H2B.8, Arabidopsis H3.14, and Oryza-specific H4.V. This review systematically summarizes recent advances in plant histone variants from three dimensio…

eIF6 links histone acetylation with translational control of HDAC in skeletal muscle.

Skeletal muscle is crucial for glucose regulation and amino acid storage, significantly influencing overall metabolic balance. Its function is tightly regulated by complex mechanisms, with histone acetylation as a key epigenetic control point. Our previous work identified eIF6 as a key regulator of muscle energy homeostasis and demonstrated its role in modulating histone acetylation in the liver. However, whether similar epigenetic mechanisms underpin eIF6's effects in muscle remains undetermined. To investigate this, we measured H3K9 acetylation levels and HDAC activity both in vivo, using eI

Chromatin chronicles: Unlocking the therapeutic potential of histone modifiers in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) represents the primary liver cancer among adults with diverse tissue appearance, high disease severity, and negative treatment expectancy. The molecular heterogeneity, extensive invasion, and propensity for relapse of HCC present a substantial challenge for oncologists. Hepatoma cells display deregulated genomic pathways interacting with epigenetic modifications. Epigenetic changes are crucial in HCC research, serving as potential biomarkers for tumor classification, prognosis, and drug targeting. Histone PTMs and chromatin regulation control gene expression duri

Histone modifications and depression: epigenetic mechanisms, therapeutic targets, and translational outlook.

Major depressive disorder (MDD) is a highly prevalent and heterogeneous psychiatric disorder shaped by the interplay of genetic, environmental, and epigenetic factors. Histone modifications, particularly acetylation and methylation, have emerged as critical regulators of chromatin dynamics and gene expression in stress adaptation, neuroplasticity, and emotional regulation. This review synthesizes current evidence linking dysregulated histone deacetylases (HDACs) and histone methyltransferases (HMTs) to impaired neuroplasticity, neuroinflammation, mitochondrial dysfunction, and hypothalamic-pit

Histone readers MLLT1 and MLLT3 concentrate AID to confer locus specificity.

Activation-induced deaminase (AID) drives antibody diversification through class-switch recombination and somatic hypermutation of the immunoglobulin (Ig) genes, but its off-target mutagenic activity contributes to B cell lymphoma1-5. How AID selectively mutates Ig loci and a restricted set of other genes remains unknown. Transcription is required for AID activity, but most transcribed genes are not mutated1,5-7, and AID occupies more loci than it mutates7-9. Here we identify the super elongation complex histone readers MLLT1 and MLLT3 as determinants of selective AID activity. Combined loss o

The role of linker histone H1.2 in tumor progression, resistance, and clinical translation.

Linker histone H1 has traditionally been regarded as a structural scaffold that stabilizes higher-order chromatin organization. Recent evidence, however, indicates that H1 variants are dynamic chromatin regulators with variant-specific functions in cancer. Among them, H1.2, encoded by H1-2 (formerly HIST1H1C), has emerged as a context-dependent regulator of chromatin compaction, DNA damage signaling, transcription, stress adaptation, and cell fate. This review summarizes the structural and post-translational features of H1.2 and examines how these properties contribute to tumor progression, in

Differential transcriptomic modulation by histone deacetylase inhibitor SAHA in LUAD and LUSC.

BACKGROUND: Histone deacetylases (HDACs) are central epigenetic regulators in non-small cell lung cancer (NSCLC), yet responses to HDAC inhibitors (HDACi) vary markedly between lung adenocarcinoma (LUAD) and lung squamous carcinoma (LUSC). We asked how the pan-HDAC inhibitor suberoylanilide hydroxamic acid (SAHA, vorinostat) rewires lineage-specific transcriptional programs and whether SAHA-aligned modules of genes, rather than individual loci, capture clinically relevant vulnerabilities in each subtype. METHODS: LUAD-like NCI-H1299 (TP53del, NRASQ61K) and LUSC-like NCI-H1703 (TP53WT, PDGFRAa

Histone tail mutants: versatile tools for decoding chromatin, development, and disease.

Histone modifications have been associated with transcriptional regulation, development, and disease, yet their direct functional roles remain incompletely understood due to the redundancy and promiscuity of cognate histone-modifying enzymes. Experimental strategies based on cis- or trans-acting histone mutants, including lysine-to-methionine (K-to-M) substitutions of histone H3 and other cancer-associated oncohistone variants, enable direct interrogation of individual chromatin marks and circumvent potentially confounding effects of histone-modifying enzyme knockouts. Here, we review how hist

Histone deacetylase enzyme activity is not the universal anticancer target of HDAC inhibitors.

Histone deacetylase inhibitors (HDIs) are approved for treating hematologic cancers and are currently being evaluated in hundreds of clinical trials for various cancers and other diseases, although their mechanisms of action remain poorly understood. Here, our unbiased bioinformatics analyses found that, for most cancer types, expression levels or genetic variants of histone deacetylase (HDACs) do not consistently correlate with carcinogenesis, do not predict cancer patient survival, and do not associate with cellular responses to HDIs. Whole-genome CRISPR library screens did not identify HDAC
