Development of an enzyme-linked phage receptor-binding protein assay (ELPRA) based on a novel biorecognition molecule- receptor-binding protein Gp130 of Pseudomonas aeruginosa bacteriophage Henu5.

Pseudomonas aeruginosa is a Gram-negative bacterium associated with life-threatening healthcare-associated infections (HAIs), including burn … ytic bacteriophages (or phages) could specifically attach to and lyse host bacterial cells. Phages' host specificity is typically determined by their receptor-binding proteins (RBPs), which recognize and adsorb phages to particular bacterial host receptors. This makes RBPs promising biorecognition molecules in bacterial detection. This study identified a novel RBP …

Modified Bacteriophage Tail Fiber Proteins for Labeling, Immobilization, Capture, and Detection of Bacteria.

A critical component of bacterial detection assays is choosing a suitable affinity molecule that retains sensitivity and specificity for the target pathogen over a wide range of in situ applications. Bacteriophages (phages) are bacterial viruses that bind and infect their host cells with unmatched specificity. Phage host range is often determined by their long tail fibers (LTFs) that mediate adsorption of the virus particle to potential bacterial host cells, by binding to specific cell surface receptors. The inherent specificity of the LTFs for distinct bacterial species makes them ideal candi

Escherichia coli bacteriophage vB_EcoS_MS2 effectively prevents biofilm formation but shows limited activity against mature biofilms.

Pathogenic Escherichia coli biofilms pose a significant challenge in veterinary and clinical settings due to their high antibiotic resistance. This study characterizes the E. coli bacteriophage vB_EcoS_MS2 as a potential targeted biocontrol agent against these pathogens. Host range determination revealed high specificity, with vB_EcoS_MS2 lysing only select E. coli strains and demonstrating superior replication efficiency (Efficiency of Plating: 112.57%) on strain C4 compared to its propagation host, MS2. While nutrient limitation strongly induced biofilm formation in in vitro assays, phage vB

Isolation and Characterization of a New Bacteriophage: Host Range Assessment and Prospects of Use in the Identification of Moraxella spp.

Bacteria of the genus Moraxella are significant pathogens in both human clinical practice and veterinary medicine. A novel bacteriophage, designated vB_MboM_ZALNahr, was isolated from environmental sources (wastewater from the Moscow Region) and morphologically classified as a myovirus lytically active against Moraxella spp. The phage exhibits a broad spectrum of lytic activity, affecting 76% of the tested Moraxella isolates of various origins. Genome sequencing revealed integrase genes, indicating the temperate nature of the phage and limiting its therapeutic potential. Phylogenetic analysis

Infection Dynamics and Coexistence of Two Novel Arctic Phytoplankton Viruses.

Marine algal viruses exhibit a high level of diversity, and closely related viruses targeting the same algal host species can stably coexist. Here we report an example of a single virus-host system concealing hidden complexity. We discovered two double stranded (ds) DNA viruses infecting the Arctic picophytoplankter Micromonas polaris coexisting in culture for over a decade. Genomic sequencing of the lysate originally characterized as MpoV-44T revealed that it comprises two distinct prasinoviruses with ~203-204 kb genomes (MpoV-44T.A and MpoV-44T.B), of which conserved regions only accounted f

Understanding geographic and host range expansion of stored product insects to improve quarantine and pest management programs.

Many species of stored product insects have been spread by commerce, a few recently enough to have a written record. The bean weevil, Acanthoscelides obtectus (Say), Mediterranean flour moth, Ephestia kuehniella (Zeller), and Angoumois grain moth, Sitotroga cerealella (Olivier), have some of the most complete early records. The larger black flour beetle, Cynaeus angustus (LeConte) may have the most complete record of both geographic and host range expansion. The introduction of heated buildings and storage sites has allowed tropical species to establish in temperate climates. The adoption of c

Appelmans Protocol for in vitro Klebsiella pneumoniae phage host range expansion leads to induction of the novel temperate linear plasmid prophage vB_KpnS-KpLi5.

Adjuvant therapy with bacteriophage (phage) cocktails in combination with antibiotics is a therapeutic approach currently considered for treatment of infections with encapsulated, biofilm forming, and multidrug-resistant Klebsiella pneumoniae (Kp). Klebsiella phage are highly selective in targeting a bacterial capsule type. Considering the numerous Kp capsule types and other host restriction factors, phage treatment could be facilitated when generating phages with a broad host range. A modified 'Appelmans protocol' was used to create phages with an extended host range via in vitro forced DNA r

Reprogramming Targeted-Antibacterial-Plasmids (TAPs) to achieve broad-host range antibacterial activity.

The emergence and spread of antimicrobial resistance results in antibiotic inefficiency against multidrug resistant bacterial strains. Alternative treatment to antibiotics must be investigated to fight bacterial infections and limit this global public health problem. We recently developed an innovative strategy based on mobilizable Targeted-Antibacterial-Plasmids (TAPs) that deliver CRISPR/Cas systems with strain-specific antibacterial activity, using the F plasmid conjugation machinery for transfer into the targeted strains. These TAPs were shown to specifically kill a variety of Enterobacter

Host Range and Phenology of Sugarcane Aphid (Hemiptera: Aphididae) and Natural Enemy Community in Sorghum in Haiti.

To determine the factors leading to outbreaks of the sugarcane aphid, Melanaphis sacchari, (Zehntner) (Hemiptera: Aphididae) in sorghum in Haiti, a survey was carried out on farms during two cropping seasons, spring and fall of 2018. A total of 45 plots in three Haitian regions were monitored from the five-leaf stage to grain ripening. Infestation with M. sacchari was significantly higher in spring than in fall, except in one location. Melanaphis sacchari populations varied significantly according to phenological stages of sorghum, with significantly higher abundance during the heading and flo

RB49-like Bacteriophages Recognize O Antigens as One of the Alternative Primary Receptors.

The power of most of the enterobacterial O antigen types to provide robust protection against direct recognition of the cell surface by bacteriophage receptor-recognition proteins (RBP) has been recently recognized. The bacteriophages infecting O antigen producing strains of E. coli employ various strategies to tackle this nonspecific protection. T-even related phages, including RB49-like viruses, often have wide host ranges, being considered good candidates for use in phage therapy. However, the mechanisms by which these phages overcome the O antigen barrier remain unknown. We demonstrate her

Expanded Diversity and Host Range of Bovine Hepacivirus-Genomic and Serological Evidence in Domestic and Wild Ruminant Species.

The hepatitis C virus (HCV)-related bovine hepacivirus (BovHepV) can cause acute as well as persistent infections in cattle. The true clinical relevance of the virus is not yet known. As reliable antibody detection methods are lacking and prevalence studies have only been conducted in cattle and few countries to date, the true distribution, genetic diversity, and host range is probably greatly underestimated. In this study, we applied several RT-PCR methods and a nano-luciferase-based immunoprecipitation system (LIPS) assay to analyze bovine serum samples from Bulgaria as well as wild ruminant

Enterohepatic Helicobacter species - clinical importance, host range, and zoonotic potential.

The genus Helicobacter defined just over 30 years ago, is a highly diverse and fast-growing group of bacteria that are able to persistently colonize a wide range of animals. The members of this genus are subdivided into two groups with different ecological niches, associated pathologies, and phylogenetic relationships: the gastric Helicobacter (GH) and the enterohepatic Helicobacter (EHH) species. Although GH have been mostly studied, EHH species have become increasingly important as emerging human pathogens and potential zoonotic agents in the last years. This group of bacteria has been assoc

Adenoviruses across the animal kingdom: a walk in the zoo.

Adenoviruses (AdVs) infect representatives of numerous species from almost every major vertebrate class, albeit their incidence shows great variability. AdVs infecting birds, reptiles, and bats are the most common and diverse, whereas only one AdV has been so far isolated both from fish and amphibians. The family Adenoviridae is divided into five genera, each corresponding to an independent evolutionary lineage that supposedly coevolved with its respective vertebrate hosts. Members of genera Mastadenovirus and Aviadenovirus seem to infect exclusively mammals and birds, respectively. The genus

Multi-host disease management: the why and the how to include wildlife.

In recent years, outbreaks caused by multi-host pathogens (MHP) have posed a serious challenge to public and animal health authorities. The frequent implication of wildlife in such disease systems and a lack of guidelines for mitigating these diseases within wild animal populations partially explain why the outbreaks are particularly challenging. To face these challenges, the French Ministry of Agriculture launched a multi-disciplinary group of experts that set out to discuss the main wildlife specific concepts in the management of MHP disease outbreaks and how to integrate wildlife in the dis

Laboratory strains of Aedes aegypti are competent to Brazilian Zika virus.

The Zika virus outbreaks are unprecedented human threat in relation to congenital malformations and neurological/autoimmune complications. Since this virus has high potential to spread in regions presenting the vectors, improvement in mosquito control is a top priority. Thus, Aedes aegypti laboratory strains will be fundamental to support studies in different research fields implicated on Zika-mosquito interactions which are the basis for the development of innovative control methods. In this sense, our aim was to determine the main infection aspects of a Brazilian Zika strain in reference Aed

The impact of host genetic diversity on virus evolution and emergence.

Accumulating evidence indicates that biodiversity has an important impact on parasite evolution and emergence. The vast majority of studies in this area have only considered the diversity of species within an environment as an overall measure of biodiversity, overlooking the role of genetic diversity within a particular host species. Although theoretical models propose that host genetic diversity in part shapes that of the infecting parasite population, and hence modulates the risk of parasite emergence, this effect has seldom been tested empirically. Using Rabies virus (RABV) as a model paras

Formula to evaluate efficacy of vaccines and systemic substances against three-host ticks.

The control of ticks with vaccines is of global interest. Experimental vaccines incorporate new technologies as soon as they are available. Historically, the main vaccine studies have focused on the one-host cattle tick Rhipicephalus microplus, and efficacy evaluations have been standardised for this tick species. On the other hand, evaluations of vaccine candidates for three-host ticks are being done somewhat arbitrarily and thus comparisons within the current literature on the efficacy of vaccines, as well as other methods of control, are difficult. We herein provide a formula for the evalua
