The D stereoisomer of dihydroxyphenylalanine (D-DOPA) and its alpha-keto acid metabolite 3,4-dihydroxyphenylpyruvic acid (DHPPA) when infused into the striatum, significantly increased in vivo extracellular dopamine levels. Following D-DOPA administration, the cumulative increase in dopamine levels was 30% of the increase following L-DOPA: following DHPPA it was 11% that of L-DOPA. Rats with unilateral 6-hydroxydopamine-induced lesions of the substantia nigra demonstrated brisk contralateral turning following each compound. The turning, however, was delayed by 10-20 min and total turning was 4

The effect of dizocilpine (MK-801) on dopaminergic agonist-induced rotational behavior was investigated in rats with 6-hydroxydopamine lesions of the nigrostriatal pathway after chronic administration of levodopa. The rotational response to the D2 agonist quinpirole was markedly increased in levodopa-treated animals compared with rats chronically administered saline. The increase in responsiveness to quinpirole was reversed by co-administered MK-801. Conversely, the rotational response to the D1 agonist SKF 38393 was reduced following chronic treatment with levodopa. The decrease in response t

We have used the alpha-glucosidase inhibitor, castanospermine, to investigate the possible involvement of N-linked glycosylation in the process of neurite outgrowth both during the development and regeneration of sympathetic nerve fibres into the iris of the rat. The effects on nerve growth were assessed qualitatively using catecholamine histochemistry and quantitatively by measuring the uptake of [3H]noradrenaline. Castanospermine was injected during the first, second or third postnatal week or during the second to fourth week after degeneration of adrenergic nerve terminals with the neurotox

Unilateral destruction of the nigrostriatal pathway by injection of 6-hydroxydopamine (6-OHDA) into the medial forebrain bundle (MFB) produces a long-lasting (> 3 months) increase in the expression of Fos-like immunoreactivity (FLI) in medium-sized neurons (12-18 microns) of the ipsilateral striatum. In order to determine the nature of neurons which contain FLI in the 6-OHDA-denervated striatum, striatonigral and striatopallidal neurons were retrogradely labelled with the fluorescent tracer Fluoro-Gold. Nuclei displaying FLI were frequently found in striatopallidal neurons (72% overlap) but se

Substance P (SP) infused into the third cerebral ventricle blocks spontaneous ovulation in female rats, probably through catecholaminergic neurons. The studies presented in this paper were undertaken to investigate whether SP exerts its suppressing effect on ovulation in 6-hydroxydopamine (6-OHDA) pretreated female rats. After 8-12 days following 6-OHDA pretreatment female rats were infused, on the day of proestrus, with vehicle and all animals were found to ovulate or with a solution containing 5 nmol of SP and 89% of female rats were then found to ovulate. In the group pretreated with vehicl

Morphine was injected into the periaqueductal gray region of the rat and 8-phenyltheophylline, an adenosine receptor antagonist, was injected intrathecally 15 or 30 min later, to determine whether supraspinally-administered morphine activated descending mechanisms to release adenosine (or a nucleotide which is metabolized to adenosine) from the spinal cord. 8-Phenyltheophylline (10 micrograms) reversed the antinociceptive action of morphine in the hot plate but not the tail-flick test. A combination of methysergide/phentolamine (15 micrograms each) reversed the action of morphine in both tests

Several structural analogs of mazindol were tested as inhibitors of the uptake of [3H] dopamine in rat neostriatum, of [3H] norepinephrine in rat occipital cortex and of [3H] serotonin in whole rat brain. A rather wide range of potencies was observed but a number of the drugs were even more potent than mazindol as uptake inhibitors. All of the drugs studied were weak releasing agents for previously accumulated [3H]amines. Several of the drugs caused large increases in motor activity in normal mice but not in reserpinized mice. However, these same drugs were able to prevent amphetamine-induced

After cold storage of guinea-pig taenia strips for 4 days at 2 degrees C, structures with specific catecholamine fluorescence were not observed. Upon incubation in Krebs Ringer medium (37 degrees C) the fluorescence in the cold stored tissue reappeared and the density of fluorescence was comparable to that of fresh preparations. The specific fluorescence was affected following reserpine or 6-hydroxydopamine treatment; however, the rewarming process restored fluorescence only in the reserpine-treated tissue. The cold storage treatment (4 and 7 days) also decreased the tissue noradrenaline and d

Intracisternal injection of blood in the rat induces an angiographically demonstrable, biphasic cerebral vasospasm of the vertebrobasilar system, with a maximal acute spasm at 10 min and a maximal late spasm at 2 days after the subarachnoid hemorrhage (SAH). Selective lesioning of the A1 nuclei in the medulla oblongata prior to the SAH prevents the development of the late spasm, but the acute spasm develops to the same extent as in sham-lesioned animals. Lesions of the medullary A2 nuclei not only prevent the development of both acute and late spasm, but give rise to a dilatation of the verteb

Acutely (by 48 h), as well as after one month survival, morphine (20 mg/kg) significantly potentiated the extent of noradrenergic degeneration in the forebrain of newborn rats caused by 50 mg/kg of 6-hydroxydopamine (6-OHDA) s.c. This was associated with increased regenerative sprouting in the cerebellum and pons-medulla as determined by measurements of norepinephrine (NE) concentration and [3H]NE uptake. This potentiating effect of morphine was not seen in either forebrain, brainstem, or cerebellum if it was administered to newborn rats with a 24-h delay following 6-OHDA. Clonidine (0.1 mg/kg

The actions of the psychotomimetic drug phencyclidine (PCP) were studied using Purkinje neurons in the cerebellum of urethane-anesthetized rats. PCP, applied by micropressure ejection through multibarreled micropipettes, depressed the spontaneous activity of these neurons as recorded by extracellular electrophysiological techniques. This depressant effect was blocked by neuroleptic drugs and lithium, both of which also block the depressant effects of norepinephrine, but not those of gamma-aminobutyric acid. PCP-elicited depressions could not be obtained in rats in which the cerebellar noradren

To examine the mechanism of the recently reported effect of an acidified intragastric test meal on insulin release and glucose homeostasis, a liver extract test meal at either pH 2 or pH 7 was instilled into the stomach of normal dogs and dogs with a chemical sympathectomy or indomethacin-induced prostaglandin deficiency, all of which had a bisected pylorus and gastric fistula. In the normal dogs the instillation of the liver meal at pH 2 elicited a significant rise in plasma glucose, glucagon and insulin levels, while in response to the meal at pH 7 only glucagon rose significantly. This was

Single, sublethal subcutaneous doses of 6-hydroxydopamine (6-OH-DA) were given to young adult male and female Marsh mice. The ability of 6/8 males to sire 15 or more days later was not abolished. Of 10 females dosed during pregnancy, 6 failed to deliver and lost substantially in body weight. Of the 4 that delivered, 2 cannabalized their young. The offspring of the 6-OH-DA-dosed fathers and mothers showed no gross anatomical defects nor lessened survival rates. Confrontation of 6-OH-DA-dosed males with control males as pairs demonstrated increased anal sniffing as compared with control-pair con

The survival and functional properties of dispersed cell implants of catecholaminergic cells obtained from the peripheral nervous system of adult rats (adrenal medulla and carotid body glomus cells) and PC12 cells from a rat pheochromocytoma cell line were examined following transplantation into the striatum of the adult rat. The host animals, all with unilateral 6-hydroxydopamine (6-OHDA) nigrostriatal lesions, were divided into 5 groups: (1) PC12 cells transplanted into Cyclosporin-A treated hosts; (2) PC12 cell grafts into hosts without Cyclosporin-A treatment; (3) grafts of adrenal medulla

Neurotensin (NT), an endogenous tridecapeptide, produces significant hypothermia after intracisternal (i.c.) or intracerebroventricular (i.c.v.) administration in microgram quantities in a variety of laboratory animals. The present study sought to clarify the mechanism of the hypothermic action by utilizing pharmacological treatments which alter the function of brain neurotransmitter systems. Pretreatment of rats with anti-muscarinic (atropine), anti-noradrenergic (propranolol, a beta-blocker; phenoxybenzamine, an alpha-blocker) or anti-opiate (naloxone) agents did not significantly alter NT-i

In the neuro-intermediate lobe (NIL) of the eel, Anguilla anguilla, a specific formaldehyde-induced fluorescence, indicating a catecholamine (CA) innervation, has been demonstrated in the neural lobe processes. Microspectrofluorimetric analyses and pharmacological treatments indicate noradrenaline or dopamine or both to be responsible for the fluorescence. The fluorescence in the NIL has displayed a definite tendency toward variation during the adaptation to a white and to a black background. The highest amounts of fluorescence were generally found in animals adapted to a black background, esp

Parkinson's disease (PD) is characterized by the progressive degeneration of midbrain dopaminergic (DA) neurons, leading to disruption of the nigrostriatal pathway and motor deficits. Here, we present an optimized protocol for generating human midbrain organoids (hMOs) suitable for homotopic transplantation, through sequential modifications. Homotopic transplantation of these optimized hMOs into the substantia nigra (SN) of 6-hydroxydopamine (6-OHDA)-lesioned PD mice resulted in significant graft survival, with a majority of grafted cells expressing the A9-specific neuronal marker GIRK2. The g

Progressive degeneration of dopamine neurons in the substantia nigra is the primary pathological abnormality in Parkinson's disease (PD). It has been demonstrated that overactivation of glutamatergic neurons leads to excessive accumulation of intracellular calcium and impairs autophagy. To assess the neuroprotective effects of ifenprodil, rats with 6-hydroxydopamine (6-OHDA)-induced injuries were administered ifenprodil either intraperitoneally (I.P.) or directly into the caudate-putamen (CPU) and substantia nigra pars reticulata (SNr). The results indicate that ifenprodil increases motor func
