Effects of a Functional Food Mixture on Patients Undergoing Hemodialysis: Preliminary Results Highlighting the Modulation of Uremic Toxins Derived From Microbiota and Antioxidant Markers.

Combining functional foods may mitigate oxidative stress and gut dysbiosis complications in patients with chronic kidney disease (CKD). This study evaluated the impact of a mixture of turmeric and propolis on redox balance markers and microbiota-derived uremic toxins in patients undergoing hemodialysis (HD). Patients received 10 mL of a compound containing Curcuma longa L. (130 mg curcuminoids/day) and green propolis extract (110 mg/day of green propolis [EPP-AF]), along with ginger and lemon juice, or a placebo, once a day for 8 weeks. Antioxidant capacity was analyzed using the oxygen radica

Tomato MYB21 Acts in Ovules to Mediate Jasmonate-Regulated Fertility.

The function of the plant hormone jasmonic acid (JA) in the development of tomato (Solanum lycopersicum) flowers was analyzed with a mutant defective in JA perception (jasmonate-insensitive1-1, jai1-1). In contrast with Arabidopsis (Arabidopsis thaliana) JA-insensitive plants, which are male sterile, the tomato jai1-1 mutant is female sterile, with major defects in female development. To identify putative JA-dependent regulatory components, we performed transcriptomics on ovules from flowers at three developmental stages from wild type and jai1-1 mutants. One of the strongly downregulated gene

Pivotal role of PAI-1 in a murine model of hepatic vein thrombosis.

Hepatic veno-occlusive disease (VOD) is a common complication of high-dose chemotherapy associated with bone marrow transplantation. While the pathogenesis of VOD is uncertain, plasminogen activator inhibitor-1 (PAI-1) has emerged as a diagnostic marker and predictor of VOD in humans. In this study, we investigated the role of PAI-1 in a murine model of VOD produced by long-term nitric oxide synthase inhibition using L-NAME. After 6 weeks, wild-type (WT) mice developed extensive fibrinoid hepatic venous thrombi and biochemical evidence of hepatic injury and dysfunction. In contrast, PAI-1-defi

Glycolysis selectively shapes the presynaptic action potential waveform.

Mitochondria are major suppliers of cellular energy in neurons; however, utilization of energy from glycolysis vs. mitochondrial oxidative phosphorylation (OxPhos) in the presynaptic compartment during neurotransmission is largely unknown. Using presynaptic and postsynaptic recordings from the mouse calyx of Held, we examined the effect of acute selective pharmacological inhibition of glycolysis or mitochondrial OxPhos on multiple mechanisms regulating presynaptic function. Inhibition of glycolysis via glucose depletion and iodoacetic acid (1 mM) treatment, but not mitochondrial OxPhos, rapidl

Synaptic activity modifies the levels of Dorsal and Cactus at the neuromuscular junction of Drosophila.

The Drosophila Rel transcription factor Dorsal and its inhibitor Cactus participate in a signal transduction pathway involved in several biologic processes, including embryonic pattern formation, immunity, and muscle development. In contrast with embryonic muscle, where Dorsal is reportedly absent, this protein and Cactus accumulates in the neuromuscular junctions in the muscle of both larvae and adults. The phenotype of homozygous dorsal mutant larvae suggested that Dorsal and Cactus maybe necessary for normal function and maintenance of the neuromuscular system. Here we investigate if these

SPATULA regulates CLAVATA1 and GCN5 genetic interaction during gynoecium development in Arabidopsis thaliana.

GENERAL CONTROL NONDEREPRESSIBLE 5 (GCN5) is a histone acetyltransferase in Arabidopsis thaliana that plays a key role in various biological processes, including the development of flowers and inflorescences. We have shown that GCN5 genetically interacts with the CLAVATA (CLV) signalling pathway, potentially affecting gynoecium and inflorescence development by modulating auxin and cytokinin dynamics and regulating WUSCHEL (WUS) expression. The bHLH transcription factor SPATULA (SPT) is also a crucial regulator of gynoecium formation. In clv1gcn5 double mutants, notable phenotypic changes inclu

Phosphorus application enhances cadmium tolerance in Salix caprea via physiological and anatomical adjustments.

Cadmium (Cd) contamination has pronounced negative effects on plant physiological processes, while phosphorus (P) as an essential macronutrient might mitigate Cd toxicity by modulating plant adaptivity. This study employed Salix caprea as a model to assess the effects of P on physiological and anatomical characteristics under Cd stress. The results demonstrated that plant growth, e.g., height, basal diameter and biomass, photosynthetic pigment content and net photosynthetic rate were all significantly inhibited by Cd stress. However, adequate P could alleviate these inhibitory effects in contr

Microbiota-derived 3-IAA influences chemotherapy efficacy in pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) is expected to be the second most deadly cancer by 2040, owing to the high incidence of metastatic disease and limited responses to treatment1,2. Less than half of all patients respond to the primary treatment for PDAC, chemotherapy3,4, and genetic alterations alone cannot explain this5. Diet is an environmental factor that can influence the response to therapies, but its role in PDAC is unclear. Here, using shotgun metagenomic sequencing and metabolomic screening, we show that the microbiota-derived tryptophan metabolite indole-3-acetic acid (3-IAA) is

Cyanogenic pseudomonads influence multitrophic interactions in the rhizosphere.

In the rhizosphere, plant roots cope with both pathogenic and beneficial bacterial interactions. The exometabolite production in certain bacterial species may regulate root growth and other root-microbe interactions in the rhizosphere. Here, we elucidated the role of cyanide production in pseudomonad virulence affecting plant root growth and other rhizospheric processes. Exposure of Arabidopsis thaliana Col-0 seedlings to both direct (with KCN) and indirect forms of cyanide from different pseudomonad strains caused significant inhibition of primary root growth. Further, we report that this gro

In the ventral tegmental area picrotoxin blocks FGIN 1-27-induced increases in sexual behavior of rats and hamsters.

RATIONALE AND OBJECTIVES: There are two types of benzodiazepine receptors, mitochondrial benzodiazepine receptors (MBRs), and gamma-aminobutyric acid (GABA(A))/benzodiazepine receptor complexes (GBRs). MBR activation increases neurosteroidogenesis. Ventral tegmental area (VTA) infusions of the MBR agonist, FGIN 1-27, increase midbrain levels of the progesterone metabolite 5alpha-pregnan-3alpha-ol-20-one (3alpha,5alpha-THP) and lordosis of rats and hamsters. Activation of GBRs leads to membrane hyperpolarization. In the VTA, infusions of GBR agonists enhance 3alpha,5alpha-THP-facilitated lordos

Genome-wide identification and transcriptional profiling analysis of auxin response-related gene families in cucumber.

BACKGROUND: Auxin signaling has a vital function in the regulation of plant growth and development, both which are known to be mediated by auxin-responsive genes. So far, significant progress has been made toward the identification and characterization of auxin-response genes in several model plants, while no systematic analysis for these families was reported in cucumber (Cucumis sativus L.), a reference species for Cucurbitaceae crops. The comprehensive analyses will help design experiments for functional validation of their precise roles in plant development and stress responses. RESULTS:

Involvement of calcium-dependent protein kinase in rice (Oryza sativa L.) lamina inclination caused by brassinolide.

Promotive effect of brassinolide (BL) on green lamina inclination was concentration-dependent when excised rice (Oryza sativa L.) lamina was floated on BL solution under continuous light conditions. Protein kinase inhibitor staurosporine and Ca2+ channel blocker LaCl3 could completely, while Ca2+ chelator EGTA could partially inhibit the lamina inclination caused by BL. Two protein kinases with apparent molecular masses of 45 and 54 kDa were detected using an in-gel kinase assay with histone III-S as a substrate. In particular, the changes in 45 kDa protein kinase activity correlated with lami

Salt-tolerant plant growth-promoting Bacillus pumilus strain JPVS11 to enhance plant growth attributes of rice and improve soil health under salinity stress.

Rice (Oryza sativa L.) growth and productivity has been negatively affected due to high soil salinity. However, some salt-tolerant plant growth-promoting bacteria (ST-PGPB) enhance crop growth and reduce the negative impacts of salt stress through regulation of some biochemical, physiological, and molecular features. Total thirty six ST-PGPB were isolated from sodic soil of eastern Uttar Pradesh, India, and screened for salt tolerance at different salt (NaCl) concentrations up to 2000 millimolar (mM). Out of thirty-six, thirteen strains indicated better growth and plant growth properties (PGPs

Population diversity of bacterial endophytes from jute (Corchorus olitorius) and evaluation of their potential role as bioinoculants.

Endophytes are bacterial or fungal organisms associated with plants in an obligate or facultative manner. In order to maintain a stable symbiosis, many of the endophytes produce compounds that promote plant growth and help them adapt better to the environment. This study was conducted to explore the potential of jute bacterial endophytes for their growth promotion ability in direct and indirect ways. A total of 27 different bacterial species were identified from different varieties of a jute plant (Corchorus olitorius) and different parts of the plant (leaf, root, seed, and seedling) based on

Regulation of GacA in Pseudomonas chlororaphis Strains Shows a Niche Specificity.

The GacS/GacA two-component system plays a central role in the regulation of a broad range of biological functions in many bacteria. In the biocontrol organism Pseudomonas chlororaphis, the Gac system has been shown to positively control quorum sensing, biofilm formation, and phenazine production, but has an overall negative impact on motility. These studies have been performed with strains originated from the rhizosphere predominantly. To investigate the level of conservation between the GacA regulation of biocontrol-related traits in P. chlororaphis isolates from different habitats, the stud

Pesticide tolerant and phosphorus solubilizing Pseudomonas sp. strain SGRAJ09 isolated from pesticides treated Achillea clavennae rhizosphere soil.

In this study, an attempt was made to identify an effective phosphate solubilizing bacteria from pesticide polluted field soil. Based on the formation of solubilization halo on Pikovskaya's agar, six isolates were selected and screened for pesticide tolerance and phosphate (P) solubilization ability through liquid assay. The results showed that only one strain (SGRAJ09) obtained from Achillea clavennae was found to tolerate maximum level of the pesticides tested and it was phylogenetically identified as Pseudomonas sp. It possessed a wide range of pesticide tolerance, ranging from 117 μg mL(-1

Identification of Novel Inhibitors of Auxin-Induced Ca2+ Signaling via a Plant-Based Chemical Screen.

Many signal perception mechanisms are connected to Ca2+-based second messenger signaling to modulate specific cellular responses. The well-characterized plant hormone auxin elicits a very rapid Ca2+ signal. However, the cellular targets of auxin-induced Ca2+ are largely unknown. Here, we screened a biologically annotated chemical library for inhibitors of auxin-induced Ca2+ entry in plant cell suspensions to better understand the molecular mechanism of auxin-induced Ca2+ and to explore the physiological relevance of Ca2+ in auxin signal transduction. Using this approach, we defined a set of di

Antagonistic regulation of PIF4 and HY5 activity by BBX28 regulates thermomorphogenesis.

ELONGATED HYPOCOTYL 5 (HY5) and PHYTOCHROME INTERACTING FACTOR 4 (PIF4) compete for binding to common promoter elements of growth-related genes. While HY5 suppresses these genes to inhibit elongation, under high ambient temperature, PIF4 promotes their expression. The molecular regulation of this differential binding is critical for adaptive growth. Under elevated temperature (28°C), BBX28 expression and accumulation are enhanced, and bbx28 seedlings exhibit hyposensitivity to temperature-mediated hypocotyl elongation. Our genetic analyses indicate that BBX28 and PIF4 act in the same pathway a
