Pangenome analysis of Lactobacillus mulieris strains reveals distinct subspecies clusters with defined ecological adaptations.

Lactobacillus mulieris is a recently described species, reportedly isolated from human urine, vagina, and gut. Previous genomic studies of L. mulieris highlighted significant genetic diversity among its strains. To gain a deeper understanding of this genomic diversity, we conducted a comprehensive genomic comparison of 70 L. mulieris strains from diverse sources. Phylogenomic and genome relatedness analysis identified three distinct clades, each representing a potential subspecies cluster. Pangenome analysis revealed distinct gene clusters shaping the functional characteristics and unique ecol…

Functional properties and flavor of fermented germinated brown rice-chickpea beverage with Lactobacillus plantarum.

This study developed a fermented beverage from germinated brown rice and chickpea using Lactobacillus plantarum. The results showed that the 36 h-fermented product exhibited strong antioxidant capacity (DPPH: 81.16 %; ABTS: 76.37 %; FRAP: 3.44 μmol/L) and significant inhibitory effects on α-amylase (69.94 %) and α-glucosidase (70.84 %), highlighting its potential antidiabetic properties. Although simulated in vitro digestion led to a reduction in biological activities of FGCB, its substantial antioxidant and antidiabetic activities were retained. Importantly, fermentation converted undesirable…

Lactobacillus strains from sows improve swine appetite by reducing the pro-inflammatory cytokine concentrations.

Lactational feed intake (LFI) is important for the efficiency of sow production. Previous studies indicated that the gut microbiota participated in host appetite regulation. The role of the gut microbiota, especially Lactobacillus mucosae LM1 and Lactobacillus amylovorus in LFI regulation of sows was proved in the previous study. However, it is unclear whether and how L. mucosae and L. amylovorus affect feed intake. Forty-nine sows with similar backfat thickness (15.81 ± 0.31 mm), parity (3.36 ± 0.16), and initial litter size (10.08 ± 0.10) were used in the current study. They were stratified …

Therapeutic potential of heat-killed Lactobacillus reuteri against bile acid-induced male reproductive toxicity.

Exposure of testicular tissue to supraphysiological concentrations of bile acids can lead to infertility. This study aimed to investigate the protective effects of heat-killed Lactobacillus reuteri (L. reuteri) against male reproductive toxicity induced by bile duct ligation (BDL). Thirty-two male Wistar rats were randomly assigned to four groups (N = 8): control normal (CN), sham-control (SC), BDL-control (underwent BDL surgery), and BDL + heat-killed L. reuteri (received probiotic supplementation for 7 days before and 21 days after BDL). At the end of the study, reproductive parameters, incl…

Lactobacillus acidophilus promotes cognitive function recovery via regulating microglial peroxisomal function in cerebral ischemia.

Cerebral ischemia causes significant mortality and is accompanied by inflammatory reactions that exacerbate damage. While the gut microbiota is linked to clinical outcomes, specific treatments and mechanisms remain unclear. Our metagenomics linked low Lactobacillus abundance to cognitive decline in patients with cerebral ischemia. L. acidophilus supplementation alleviated post-ischemic deficits in murine middle cerebral artery occlusion and bilateral carotid artery stenosis models. The neuroprotective effects of L. acidophilus were driven by enhanced gut absorption of linoleic acid (LinA). L. …

Gut microbiome Lactobacillus acidophilus and its relationship with thyroid nodules and thyroid hormonal profile.

INTRODUCTION: Thyroid diseases are often associated with the amounts and functioning of thyroid hormones, which may have an impact on the makeup of gut microbiomes. Multiple studies have shown a correlation between gut microbiota and both Graves' disease and Hashimoto's thyroiditis. However, there is no proven link between the gut microbiota and thyroid nodules. Researchers will examine the correlation between Lactobacillus acidophilus and thyroid nodules and hormones. MATERIALS AND METHODS: This work is a prospective case-control investigation undertaken from 2021 to 2022 at endocrine clinic…

Lactobacillus gasseri associated severe soft tissue infection of the lower extremity: A case report.

BACKGROUND: Lactobacilli are gram-positive, lactic acid-producing, facultative anaerobes of the human microbiota located in the human gastrointestinal tract, genitourinary tract, and the oral cavity and are considered non-pathogenic. When certain risk factors are present, they have the potential to cause serious infections. The incidence of localized infections associated with Lactobacilli are rare and to our knowledge we present the first known case of severe soft tissue infection of the extremity linked to a Lactobacillus strain. CASE PRESENTATION: We describe the case of a 41-year-old man …

Breed-Driven Microbiome Heterogeneity Regulates Intestinal Stem Cell Proliferation via Lactobacillus-Lactate-GPR81 Signaling.

Genetically lean and obese individuals have distinct intestinal microbiota and function. However, the underlying mechanisms of the microbiome heterogeneity and its regulation on epithelial function such as intestinal stem cell (ISC) fate remain unclear. Employing pigs of genetically distinct breeds (obese Meishan and lean Yorkshire), this study reveals transcriptome-wide variations in microbial ecology of the jejunum, characterized by enrichment of active Lactobacillus species, notably the predominant Lactobacillus amylovorus (L. amylovorus), and lactate metabolism network in obese breeds. The…

Postbiotics From Lactobacillus Johnsonii Activates Gut Innate Immunity to Mitigate Alcohol-Associated Liver Disease.

Prolonged alcohol consumption disrupts the gut microbiota and the immune system, contributing to the pathogenesis of alcohol-associated liver disease (ALD). Probiotic-postbiotic intervention strategies can effectively relieve ALD by maintaining gut homeostasis. Herein, the efficacy of heat-killed Lactobacillus johnsonii (HKLJ) in mitigating alcoholic liver damage is demonstrated in mouse models of ALD. The gut-liver axis is identified as a pivotal pathway for the protective effects of L. johnsonii against ALD. Specifically, HKLJ is found to upregulate the expression of intestinal lysozymes, th…

Lactobacillus plantarum AR495 improves colonic transport hyperactivity in irritable bowel syndrome through tryptophan metabolism.

Lactobacillus plantarum AR495 is a widely used probiotic for the treatment of various digestive diseases, including irritable bowel syndrome (IBS). However, the specific mechanisms of L. plantarum AR495 in alleviating IBS remain unclear. Abnormal intestinal tryptophan metabolism can cause disordered immune responses, gastrointestinal peristalsis, digestion and sensation, which is closely related to IBS pathogenesis. The aim of this study is to explore the effects and mechanisms of L. plantarum AR495 in regulating tryptophan metabolism. Primarily, tryptophan and its related metabolites in patie…

Lactobacillus rhamnosus modulates murine neonatal gut microbiota and inflammation caused by pathogenic Escherichia coli.

BACKGROUND: Pathogenic Escherichia coli strains produce neonatal septicemia after colonizing the neonatal gut. While the probiotic Lactobacillus rhamnosus GG (LGG) effectively reduces neonatal sepsis, LGG's effects on the neonatal intestinal microbiota alterations and inflammation triggered by E. coli are incompletely understood. We hypothesized that LGG significantly modulates the specific neonatal gut microbial populations changes and the inflammatory response elicited by the enteral introduction of septicemia-producing E. coli. To test this hypothesis, newborn rats were pretreated orally wi…

Topical Treatment of Second-Degree Burn Wounds with Lactobacillus plantarum Supernatant: Phase I Trial.

BACKGROUND & OBJECTIVE: A burn wound is sterile immediately after injury, but opportunistic bacteria colonize the wound within 48 to 72 hours after the burn, causing delayed or failed burn wound healing. In addition, the presence of multidrug-resistant (MDR) pathogens doubles the treatment problems. Lactobacillus plantarum (L. plantarum) is a well-known antibacterial and healing agent that could be used topically to treat burn wounds. CASE SERIES PRESENTATION: This clinical trial study (Case Series) was performed on 20 patients with deep second-degree burns. Patients had bilateral wounds; the…
