Democratized Discovery of Microsclerodermin F as an Immunophilin Ligand.

While immunophilins are well-recognized therapeutic targets, several members of this family of peptidyl-proline isomerases (PPIases) have yet to be subjected to ligand discovery efforts. In this study, we demonstrate a cost-effective means to identify ligands to the insufficiently investigated two-domain PPIase human Cyclophilin40 (Cyp40). Central to this effort was the use of beads, wherein a confocal nanoscanning (CONA) approach was used to rapidly probe candidates. Here, we describe how one can adapt the physical nature of microsized beads as a means to strategically reduce cost and ultimat…

Photocatalytic fluoroalkylation by ligand-to-metal charge transfer.

Trifluoromethyl (CF3) and other fluoroalkyl groups are of great significance in the fields of pharmaceutical chemistry and agricultural chemicals. Fluoroalkyl acids, especially trifluoroacetic acid (TFA) is considered the most ideal fluoroalkylation reagent due to its low cost and easy availability. However, the extremely high oxidation potential requirement of TFA limits its wide application. In recent years, since visible-light-induced fluoroalkylation through the ligand-to-metal charge transfer (LMCT) process can overcome the above limitations, it has become an effective synthetic tool for

Microbial ligand-independent regulation of lymphopoiesis by NOD1.

Aberrant differentiation of progenitor cells in the hematopoietic system is known to severely impact host immune responsiveness. Here we demonstrate that NOD1, a cytosolic innate sensor of bacterial peptidoglycan, also functions in murine hematopoietic cells as a major regulator of both the generation and differentiation of lymphoid progenitors as well as peripheral T lymphocyte homeostasis. We further show that NOD1 mediates these functions by facilitating STAT5 signaling downstream of hematopoietic cytokines. In steady-state, loss of NOD1 resulted in a modest but significant decrease in numb

Visualizing ligand bias at the Mu-opioid receptor.

Different opioid ligands can result in biased μ-opioid signaling, differentially activating signal cascades which produce analgesia, tolerance, or adverse effects. In this issue of Cell, Xu et al. used cryo-EM and computational simulations to understand how different μ-opioid receptor selective-ligands interact with key residues to produce downstream signaling.

The Natural Ligand for Metalloproteinase-A Multifaceted Drug Target.

Metalloproteinase is one of the key components of Russell viper venom and it is the root cause of edema, blood coagulation, local tissue damage, hemorrhage, and inflammation during snakebite envenoming. Hence, finding a suitable metalloproteinase inhibitor from natural source will be of great biological importance in mitigating pathological effects. In this current study, we employed computational analysis to examine the inhibition of metalloproteinase by phytochemicals present in Andrographis paniculata. Molecular docking studies revealed interaction of A. paniculata phytochemicals with the c

Immunometabolic modulation of retinal inflammation by CD36 ligand.

In subretinal inflammation, activated mononuclear phagocytes (MP) play a key role in the progression of retinopathies. Little is known about the mechanism involved in the loss of photoreceptors leading to vision impairment. Studying retinal damage induced by photo-oxidative stress, we observed that cluster of differentiation 36 (CD36)-deficient mice featured less subretinal MP accumulation and attenuated photoreceptor degeneration. Moreover, treatment with a CD36-selective azapeptide ligand (MPE-001) reduced subretinal activated MP accumulation in wild type mice and preserved photoreceptor lay

[3+2] Fragmentation of a Pentaphosphido Ligand by Cyanide.

The activation of white phosphorus (P4 ) by transition-metal complexes has been studied for several decades, but the functionalization and release of the resulting (organo)phosphorus ligands has rarely been achieved. Herein we describe the formation of rare diphosphan-1-ide anions from a P5 ligand by treatment with cyanide. Cobalt diorganopentaphosphido complexes have been synthesized by a stepwise reaction sequence involving a low-valent diimine cobalt complex, white phosphorus, and diorganochlorophosphanes. The reactions of the complexes with tetraalkylammonium or potassium cyanide afford a …

A Role for CD154, the CD40 Ligand, in Granulomatous Inflammation.

Granulomatous inflammation is a distinctive form of chronic inflammation in which predominant cells include macrophages, epithelioid cells, and multinucleated giant cells. Mechanisms regulating granulomatous inflammation remain ill-understood. CD154, the ligand of CD40, is a key mediator of inflammation. CD154 confers a proinflammatory phenotype to macrophages and controls several macrophagic functions. Here, we studied the contribution of CD154 in a mouse model of toxic liver injury with carbon tetrachloride and a model of absorbable suture graft. In both models, granulomas are triggered in r

A 1D Coordination Polymer of UF5 with HCN as a Ligand.

β-Uranium(V) fluoride was reacted with liquid anhydrous hydrogen cyanide to obtain a 1D coordination polymer with the composition 1∞ [UF5 (HCN)2 ], 1∞ [UF4/1 F2/2 - (HCN)2/1 ], revealed by single-crystal X-ray structure determination. The reaction system was furthermore studied by means of vibrational and NMR spectroscopy, as well as by quantum chemical calculations. The compound presents the first described polymeric HCN Lewis adduct and the first HCN adduct of a uranium fluoride.

Ligand-Appended BBB-Targeted Nanocarriers (LABTNs).

Delivering therapeutics across the blood brain barrier (BBB) remains the rate-limiting step in brain medicine research. Three main categories of endogenous transportation at BBB that can be used for targeting brain are carrier-mediated transport, active efflux transport, and receptor-mediated transport. Various approaches using nanocarriers such as liposomes, niosomes, micelles, and nanoparticles with manifested surface modifications using either covalent or noncovalent methods to append suitable ligands are being intensively explored to achieve drug delivery to the brain. Harvested ligands in…

Tetraspecific ligand for tumor-targeted delivery of nanomaterials.

The polygenetic nature of most cancers emphasizes the necessity of cancer therapies that target multiple essential signaling pathways. However, there is a significant paucity of targeting ligands with multi-specificities for targeted delivery of biomaterials. To address this unmet need, we generated a tetraspecific targeting ligand that recognizes four different cancer biomarkers, including VEGFR2, αvβ3 integrin, EGFR, and HER2 receptors, which have been implicated in numerous malignant tumors. The tetraspecific targeting ligand was constructed by sequentially connecting four targeting ligand …

Friedel-Crafts functionalization of the cyclopentadienyl ligand in buckymetallocenes.

Acylated buckyferrocene and ruthenocene, Fe(η(5)-C60Me5)(η(5)-C5H4COR) (R = Me, Ph, and CH[double bond, length as m-dash]CHPh) and Ru(η(5)-C60Me5)(η(5)-C5H4COR) (R = Me and Ph), were obtained by Friedel-Crafts acylation of the parent buckymetallocenes with the corresponding acid chlorides and aluminum chloride in carbon disulfide at ambient temperature. The electron withdrawing and sterically hindered nature of the acyl groups were revealed by X-ray crystallography, infrared spectroscopy and electrochemical measurements. The possibility of further derivatizing the acylated products was illustr

[Inhibition of RANK ligand to treat bone metastases].

Bone is the most common metastatic site. The skeleton is also the preferential initial metastatic site in breast and prostate carcinomas. Objective complications of bone metastases are named « skeletal-related events » (SREs) and generally include the need for radiotherapy on bone, surgery to bone, pathologic fracture and spinal cord compression. Recent phase III double-blind trials have demonstrated the superiority of denosumab to zoledronic acid for delaying the time to first SRE in patients with breast or prostate cancer and bone metastases. Non-inferiority was shown in the trial including

Evolution of ligand specificity in vertebrate corticosteroid receptors.

BACKGROUND: Corticosteroid receptors include mineralocorticoid (MR) and glucocorticoid (GR) receptors. Teleost fishes have a single MR and duplicate GRs that show variable sensitivities to mineralocorticoids and glucocorticoids. How these receptors compare functionally to tetrapod MR and GR, and the evolutionary significance of maintaining two GRs, remains unclear. RESULTS: We used up to seven steroids (including aldosterone, cortisol and 11-deoxycorticosterone [DOC]) to compare the ligand specificity of the ligand binding domains of corticosteroid receptors between a mammal (Mus musculus) an

Ligand-induced and nonfusogenic dissolution of a viral membrane.

Hitherto, all enveloped viruses were thought to shed their lipid membrane during entry into cells by membrane fusion. The extracellular form of Vaccinia virus has two lipid envelopes surrounding the virus core, and consequently a single fusion event will not deliver a naked core into the cell. Here we report a previously underscribed mechanism in which the outer viral membrane is disrupted by a ligand-induced nonfusogenic reaction, followed by the fusion of the inner viral membrane with the plasma membrane and penetration of the virus core into the cytoplasm. The dissolution of the outer envel
