Lipidomic Profiling of Milk Enables Identification of Novel Biomarkers for Subclinical Ketosis in Dairy Cows.

The current clinical test for ketosis in dairy cows, which is based on measuring blood β-hydroxybutyrate (BHB) concentrations, is an invasive procedure that impacts herd welfare. Alternative biomarkers, especially non-invasive ones, are urgently needed for subclinical ketosis (SCK) diagnosis. To address this, we examined the milk lipidomic profiles of healthy and SCK cows from a research dairy farm over two consecutive years. A total of 125 polar lipid species were quantified in milk samples using a targeted liquid chromatography-mass spectrometry (LC-MS) approach. Chemometric analysis identif

Lipidomic Evidence for Cannabidiol-Induced Remodeling of Cellular Lipid Composition in SH-SY5Y Cells.

Cannabidiol (CBD) is increasingly discussed in relation to neurodegenerative disorders, including Alzheimer's disease (AD), yet its effects on cellular lipid profiles remain poorly understood. Using targeted lipidomics, we investigated whether CBD alters the lipidome of SH-SY5Y wildtype (wt) cells and whether the principal phosphatidylcholine response is also observed in cells expressing amyloid precursor protein with the Swedish mutation (APPswe). CBD induced a broad, class-specific lipid response. Total diacyl and ether-linked phosphatidylcholines increased. Within these classes, many specie…

Metabolic, microbial, and pharmacological stimuli elicit distinct lipidomic and cytokine responses in the human placenta.

The placenta integrates metabolic and immune functions essential for fetal development, and disruptions in lipid metabolism and inflammatory signaling have been implicated in pregnancy complications such as gestational diabetes mellitus (GDM), infection-associated inflammation, and preterm birth. To examine how distinct clinically relevant stressors shape these processes, we investigated placental explant responses to three representative exposures. These included high glucose (metabolic stress relevant to diabetes/GDM), lipopolysaccharide (LPS; microbial-inflammatory stress relevant to infect

Anabolic androgenic steroids induce distinct monoaminergic and lipidomic alterations in the male rat brain.

The illicit use of anabolic androgenic steroids (AAS) to enhance physical strength, performance, and appearance is associated with severe physical and psychological adverse effects. Long-term AAS use has been associated with altered behavior and cognitive impairments. Nevertheless, the neurobiological effects of AAS use remain underexplored, especially in terms of brain neurotransmitter and lipid profiles. This study applied advanced Fourier-transform ion cyclotron resonance matrix-assisted laser desorption/ionization mass spectrometry imaging (FTICR-MALDI-MSI) to investigate the effects of th

Serum lipidomic alterations associated with post-stroke cognitive impairment: an exploratory prospective study.

BACKGROUND: Post-stroke cognitive impairment (PSCI) is a common complication of stroke that adversely affects long-term functional outcomes and quality of life among survivors. However, reliable biomarkers for early risk prediction remain limited. Dysregulation of lipid metabolism has been implicated in ischemic stroke and may contribute to PSCI development. OBJECTIVE: This study investigated early serum lipidomic alterations associated with PSCI in patients with acute ischemic stroke and evaluated whether integrating lipidomic features with clinical indicators improves predictive performance

Lipidomic and Metabolomic Profiling of Breast Muscle Between Squabs and Adult White King Pigeons.

This study systematically investigated age-related variations in meat quality traits, lipidomic profiles, and metabolomic profiles of breast muscle from White King pigeons, comparing 28-day-old squabs (D28) and 12-month-old adult pigeons (M12). Six biological replicates were used per group (n = 6). Physicochemical analyses and ultra-high performance liquid chromatography-tandem mass spectrometry were employed to characterize these differences. Results showed significant disparities in meat quality between the two age groups: D28 pigeons exhibited significantly higher lightness (L*), redness (a

Causal Relationships Among Lifestyle, Lipidome, and Polycystic Ovary Syndrome: A Mendelian Randomization Study.

BACKGROUND: Lifestyle interventions often influence lipid levels in patients with polycystic ovary syndrome (PCOS), thereby modulating disease progression. However, whether lipid levels play a mediating role remains unclear. OBJECTIVE: To investigate causal associations between lifestyle, lipidome, and PCOS, as well as mediating role of lipidome. METHODS: This study employed a bidirectional two-sample Mendelian randomization (MR) analysis, with lifestyle and lipidome as exposures and PCOS as outcome. A two-step MR approach was used to assess mediating role of lipidome. Inverse-variance weigh

Multiomics insights into functional constipation: Exploring microbiome, metabolome, and lipidome independent of transit time.

BACKGROUND/AIMS: Functional constipation (FC) is a prevalent form of functional gastrointestinal disorder with an elusive etiology. Despite numerous investigations into the role of the gut microbiome in FC, no study has yet integrated microbiome, metabolome, and lipidome profiling to characterize FC. METHODS: In this study, fecal samples were collected from patients with FC both before and after laxative administration to minimize the impact of intestinal transit time and to identify the characteristics unique to patients with FC. These samples underwent comprehensive microbiome, metabolome,

Mechanism of MWCNTs induced hepatotoxicity in Cyprinus carpio: New perspective from a Lipidomic study.

Carbon nanotubes (CNTs), an extensively applied nanomaterial, can cause damage to the liver in fish. However, the underlying lipotoxic modes of CNTs exposure have not been studied. In the present study, juvenile Cyprinus carpio was exposed via water to various doses of multi-walled CNTs (MWCNTs) (0, 0.25, and 2.5 mg L-1) for a consecutive 28-day period. Our results showed elevated hepatosomatic index (HSI) and aspartate aminotransferase (AST) activities in plasma in all exposure groups, indicating that MWCNTs exposure induced liver dysfunction. Significantly increased low-density lipoprotein (

Dapagliflozin modulates plasma lipidomic profile and urinary metabolite excretion in type 2 diabetes.

BACKGROUND: Dapagliflozin (DAPA) has shown major nephroprotective effects, improving kidney metabolism and oxigenation. Lipidomics and metabolomics are powerful tools for understanding such effects, providing a comprehensive look at how SGLT2 inhibitors might change the metabolic landscape beyond their primary glucose-lowering action. We investigated changes in plasma metabolomic/lipidomic profile and urinary excretion of metabolites that could occur independent of increased diuresis. METHODS: A two-armed, parallel-design, randomized clinical trial was conducted in subjects with type 2 diabet…

Sexual dimorphism in the association of umbilical cord blood lipidome with abdominal fat in early childhood.

BACKGROUND: Although the associations between cord blood lipidome and neonatal birth weight are established, it remains uncertain whether sexual dimorphism in fetal fat accumulation extends to the relationship between cord blood lipid profiles and neonatal abdominal fat compartments. Understanding these relationships could provide insights into early sex-specific differences in lipid metabolism. METHODS: We conducted lipidomics of umbilical cord blood plasma samples (350 (46.6%) girls and 401 (53.4%) boys) from the Growing Up in Singapore Towards healthy Outcomes (GUSTO) birth cohort. Unraveling the causal link between lipidomes and unstable angina pectoris: A Mendelian randomization analysis.

Unstable angina pectoris remains a significant global health burden. Understanding the causal relationships between lipidomes and unstable angina pectoris is crucial for preventive strategies. We conducted a Mendelian randomization (MR) analysis to explore these relationships. This study utilized lipidome data from 377,277 participants and included 14,281 unstable angina pectoris cases and 364,801 controls. We applied inverse-variance weighted (IVW) methods for MR analysis, considering 179 lipid species as exposures and unstable angina pectoris as the outcome. This MR analysis investigated the

The Primacy of Adipose Tissue Gene Expression and Plasma Lipidome in Cardiometabolic Disease in Persons With HIV.

BACKGROUND: Persons with HIV (PWH) on contemporary antiretroviral therapy (ART) are at elevated risk for developing age-related cardiometabolic diseases. We hypothesized that integrative analysis of cross-tissue, multimodal data from PWH could provide insight into molecular programming that defines cardiometabolic phenotypes in this high-risk group. METHODS: We enrolled 93 PWH without diabetes who were virologically suppressed on contemporary ART and obtained measures of insulin resistance, glucose intolerance, and adiposity. We performed circulating lipidomics, proteomics, and metabolomics, …

Sex modulates the diet-induced changes to the plasma lipidome in a rat model of cardiorespiratory fitness.

OBJECTIVE: Individuals with higher intrinsic cardiorespiratory fitness (CRF) experience decreased rates of cardiometabolic disease and mortality, and high CRF is associated with increased utilization of fatty acids (FAs) for energy. Studies suggest a complex relationship between CRF, diet, and sex with health outcomes, but this interaction is understudied. We hypothesized that FA utilization differences by fitness and sex could be detected in the plasma metabolome when rats or humans were fed a high carbohydrate (HC) or high fat (HF) diet. METHODS: Male and female rats selectively bred for lo

The impact of steatotic liver disease on coronary artery disease through changes in the plasma lipidome.

Steatotic liver disease has been shown to associate with cardiovascular disease independently of other risk factors. Lipoproteins have been shown to mediate some of this relationship but there remains unexplained variance. Here we investigate the plasma lipidomic changes associated with liver steatosis and the mediating effect of these lipids on coronary artery disease (CAD). In a population of 2579 Swedish participants of ages 50 to 65 years, lipids were measured by mass spectrometry, liver fat was measured using computed tomography (CT), and CAD status was defined as the presence of coronary
