Profiling of Single Cells on a Droplet Microfluidics-SERS Platform for Multiplexed Analysis of Oxidative and Inflammatory Stress.

Breast cancer is a highly heterogeneous disease, with subtypes such as triple-negative (TNBC) and HER2-positive (HER2+) exhibiting distinct … amplification. Single-cell measurements reveal pronounced oxidative and inflammatory activation in MDA-MB-231 cells, whereas SK-BR-3 cells display attenuated responses, highlighting subtype-specific heterogeneity. In addition, cisplatin treatment further modulated the stress-response patterns induced by electrical stimulation, indicating distinct subtype-dependen …

Mitochondrial Pro-Apoptotic Properties of Sinularia compressa from Persian Gulf against Breast Cancer Cells and Its Chemical Composition.

Locals in the Persian Gulf islands traditionally use Sinularia compressa to treat cancer. Therefore, this study deals with the cytotoxic activity of the soft coral Sinularia compressa chloroform extract (SCE), its pro-apoptotic activity, and the determination of its secondary metabolites. Cytotoxicity was done against MCF-7 and MDA-MB-231 and MCF‑10A cells. Apoptosis induction was checked by flow cytometry. The DCFDA and JC-1 probes were used to assess the production of reactive oxygen species (ROS) and the mitochondrial transmembrane potential. Caspase-9, Bax, and Bcl-2 proteins were determin

Calyculin A from Discodermia calyx is a dual action toxin that blocks calcium influx and inhibits protein Ser/Thr phosphatases.

Calyculin A (Caly A) is cell permeable toxin widely used in cell biology research as an inhibitor of type 1 and type 2A protein Ser/Thr phosphatases of the PPP family. Here we tested effects of low concentrations of Caly A on proliferation of human cancer and non-cancer cell lines. We found that long-term 0.3 nM Caly A prevented G1 to S phase cell cycle progression in human Hs-68 fibroblasts and ARPE19 epithelial cells, but not human breast cancer MDA-MB-468, MDA-MB-231 and MCF7 cells. These conditions produced no change in cyclin D1 levels or in the phosphorylation of endogenous proteins. How

A pneumolysin mutant (ΔA146 ply) induces pyroptosis in triple-negative and HER2-positive breast cancer cells via the Caspase-1/GSDME pathway: a potential antibody-drug conjugate payload.

BACKGROUND: The success of antibody-drug conjugates (ADCs) relies on potent cytotoxic payloads. Discovery of novel toxins that induce immunogenic cell death (such as pyroptosis) holds promise for overcoming tumor heterogeneity and enhancing anti-tumor immunity. Pneumolysin (Ply) is a pore-forming toxin from Streptococcus pneumoniae, but its wild-type form is overly toxic. Here we investigate the potential of a detoxified mutant, ΔA146 ply, to induce pyroptosis in triple‑negative (MDA‑MB‑231) and human epidermal growth factor receptor 2 (HER2)‑positive (MDA‑MB‑453) breast cancer cells, using a

Investigation of novel nimesulide derivatives against breast cancer.

This study focused on the synthesis of novel nimesulide semicarbazone derivatives and the evaluation of their cytotoxic potential against lu … ompound 5e. Western blot results showed a dose-dependent decrease in p-ERK levels in both MCF-7 and MDA-MB-231 cells, confirming MAPK pathway inhibition. These findings support that nimesulide-based semicarbazones, particularly compound 5e, exhibit potent antiproliferative and pro-apoptotic activity via MAPK pathway modulation, offering a promising avenue for the …

The proteomics analysis of mitochondria-enriched fractions reveals alteration of key mitochondrial pathways in breast cancer cell lines.

Breast cancer, the leading cause of death in women worldwide, shows significant heterogeneity that makes this disease extremely difficult to treat. Many reports point to metabolic shifts, mainly those carried out into mitochondria, as key processes governing the behavior and heterogeneity of several types of breast cancer. In this study, we performed label-free proteomics analysis on mitochondria-enriched fractions from T47D and MDA-MB-231 breast cancer cell lines, which have distinct molecular classifications, using bioinformatics analyses to identify differentially expressed proteins compare

ErbB-2 blockade and prenyltransferase inhibition alter epidermal growth factor and epidermal growth factor receptor trafficking and enhance (111)In-DTPA-hEGF Auger electron radiation therapy.

UNLABELLED: The intracellular distribution of Auger electron-emitting radiopharmaceuticals is a determinant of cytotoxicity. However, the mechanisms by which these agents are routed through the cell are ill understood. The aim of this study was to investigate how trafficking of (111)In-labeled human epidermal growth factor ((111)In-DTPA-hEGF) relates to that of the EGF receptor (EGFR) and whether coadministration of agents that modulate EGFR signaling alters the efficacy of (111)In-DTPA-hEGF. Membrane Fusion-Mediated Cytosolic Delivery of Threose Nucleic Acids via Homotypic Nanoparticles Overcomes Drug Resistance in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) remains lethal due to its aggressive molecular heterogeneity and drug resistance. We report a biomimetic nanoplatform (PLL/TNAAKT2@CM NPs) integrating biostable threose nucleic acid (TNA) with a donor-derived cell membranes (CMs) "cloak" for subtype-specific therapy. By complexing TNAAKT2 antisense oligonucleotides with poly-L-lysine (PLL) and coating them with TNBS-subtype membranes (MDA-MB-468 or MDA-MB-231), we achieve potent homotypic affinity. PLL/TNAAKT2@468CM NPs exhibited significant enhanced uptake in donor-matched basal-like 1 cells compared to he

Tumor suppressor syntaxin binding protein 1 regulates alternative splicing of DNA repair genes related to radiation sensitivity in breast cancer cells.

As the most aggressive subtype of breast cancer, triple-negative breast cancer (TNBC) is frequently treated with radiotherapy. Downregulation of syntaxin binding protein 1 (STXBP1) gene expression is notably associated with poor prognosis in patients with breast cancer and its protein expression level is associated with tumor radioresistance. However, the precise molecular mechanisms by which STXBP1 regulates breast cancer pathogenesis and radiotherapy resistance remain to be elucidated. In the present study, STXBP1 was overexpressed in MDA-MB-231 cells to investigate its effects on cell proli…

Quinolyl-Based PGM Metallarectangles: Antiproliferative Activity, DNA and BSA Protein Interactions, and a Molecular Docking Perspective.

The increased success of small metal-containing molecules as pharmaceutical agents has prompted investigations into the pharmacological activity of a different class of metal-based compounds; supramolecular coordination complexes (SCCs). Such complexes have been extensively investigated for their anticancer activity, with many displaying activities comparable or superior to available clinical chemotherapeutic drugs. Here, we evaluated a series of quinoline-containing binuclear complexes and metallarectangles for their in vitro anticancer activity in the hormone receptor positive MCF-7 and trip

Ester derivatives of salinomycin efficiently eliminate breast cancer cells via ER-stress-induced apoptosis.

The polyether ionophore salinomycin (SAL) has been found to selectively target breast cancer cells, including those with stem-like phenotype. On the other hand, SAL amides and esters obtained through derivatisation of the C1 carboxyl of the ionophore were found to exhibit anticancer properties, whilst reducing potential toxicity issues which often occur during standard chemotherapy. However, the studies on the activity and especially on the mechanisms of action of this class of semi-synthetic products against breast cancer cells are very limited. Therefore, in this work, we confirmed the anti-

Expression of estrogen receptor alpha with a Tet-off adenoviral system induces G0/G1 cell cycle arrest in SKBr3 breast cancer cells.

Endocrine therapies targeting estrogen action are pivotal for the prevention and treatment of ER-positive breast cancers. Previous studies s … sed. Given that estrogen-induced increase of p21Cip1/Waf1 and decrease of E2F1 was also observed in MDA-MB-231 cells stably transfected with ERalpha, our results suggest that a common pathway might be shared by different breast cancer cell lines whose growth is suppressed by ectopic ERalpha and estrogen.

Induced chromosomal instability in triple negative breast cancer cells promotes M2-like polarization of macrophages.

Chromosomal instability (CIN) is a defining feature of triple-negative breast cancer (TNBC) and is increasingly recognized as a driver of tumor evolution and immune escape. However, how CINHIGH tumor cells modulate innate immune cells, particularly macrophages, remains unclear. Here, we used the MPS1 inhibitor reversine to induce acute CIN in the TNBC cell line MDA-MB-231 and examined its impact on human macrophage polarization and function. Reversine-induced CIN stably increased the expression of macrophage-attracting cytokines, including CCL2, CCL5, and IL-6. In direct co-cultures, reversine
