Sepsis is a life-threatening syndrome driven by a dysregulated host response to infection, characterized by excessive inflammation, coagulation imbalance, and endothelial dysfunction, often progressing to multiple organ dysfunction and death. Small-molecule agents have emerged as promising therapeutic candidates by targeting key pathogenic pathways, including microbial clearance, immunomodulation, restoration of coagulation homeostasis, endothelial protection, and metabolic support. We summarize recent progress in small-molecule therapies for sepsis, focusing on their molecular targets, struct

Cancer therapy has entered an era dominated by molecularly targeted agents and immune checkpoint inhibitors (ICIs), yet therapeutic resistance and patient heterogeneity remain major barriers to durable benefit. Cuproptosis, a recently identified form of copper-dependent regulated cell death linked to mitochondrial metabolism, has emerged as a potentially relevant therapeutic concept. Distinct from apoptosis and ferroptosis, cuproptosis is triggered by the interaction of copper with lipoylated enzymes in the tricarboxylic acid (TCA) cycle, leading to proteotoxic stress and metabolic collapse. This review highlights the emergence of CALR as a promising therapeutic target in CALR-mutated myeloproliferative neoplasms. While current JAK inhibitors alleviate symptoms and reduce thrombosis risk, they lack clonal specificity, have a limited impact on the natural history of disease, and are limited by toxicity and resistance. In contrast, CALR mutations generate a unique, disease-specific neoepitope that drives disease pathogenesis through aberrant MPL activation and is accessible on the cell surface, making it an ideal immunologic and antibody target. Preclinical and early clinical advanc

Dry eye disease, as an ocular surface disorder, affects millions of people globally and has consequently become one of the most prevalent conditions encountered in ophthalmic clinics. The pathogenesis of dry eye involves several key factors, including tear film instability, hyperosmolarity, chronic ocular surface inflammation, and neurosensory abnormalities. The ocular surface expresses a diverse array of ion channels, which play essential physiological roles in maintaining tear film stability, regulating osmotic pressure, balancing ion transport, and modulating inflammatory responses. This re

Tumor-agnostic, or histology-agnostic, cancer therapy marks a groundbreaking evolution in the realm of precision oncology. In stark contrast to conventional cancer treatments that categorize malignancies based on their tissue of origin (eg, breast, lung, renal cell, etc), tumor-agnostic therapies transcend histologic boundaries, honing in on the genetic and molecular attributes of tumors, regardless of their location. This article offers a comprehensive review of the current landscape of tissue-agnostic cancer therapies and provides clinical insights to empower surgical oncologists with a deep

There has been perhaps no greater advance in the prognosis of solid tumors in the last decade than for patients with metastatic melanoma. This is due to significant improvements in treatment based on two key components of melanoma tumor biology (1) the identification of driver mutations with therapeutic potential and (2) the mechanistic understanding of a tumor-specific immune response. With breakthrough findings in such a relatively short period of time, the treatment of patients with metastatic melanoma has become intensely personalized.

Diabetic peripheral neuropathy (DPN), a debilitating diabetic complication, has a complex pathological mechanism involving oxidative stress, mitochondrial dysfunction, and endoplasmic reticulum stress, and there are no effective disease-mitigating treatments. Current management is restricted to glycaemic control and symptomatic analgesia, both of which offer only modest benefit and carry appreciable adverse-effect profiles. Heat Shock Proteins (HSP) are stress-inducible chaperones that counteract protein misfolding and aggregation. Through suppression of apoptosis, cytoskeletal stabilisation a

Biliary tract cancer (BTC) is a rare group of malignancies that develop from the epithelial lining of the biliary tree and have a poor prognosis. Although chemotherapy is the standard of care for patients with advanced BTC in China, its clinical benefits are moderate. In recent years, the approval of targeted therapies and immunotherapies has provided new avenues for the management of advanced BTC. Nonetheless, the increasing number of personalized medicine approaches has created a challenge for clinicians choosing individualized treatment strategies based on tumor characteristics. In this art

The sodium/potassium adenosine triphosphatase (Na+/K+ ATPase, NKA) is a ubiquitous membrane protein crucial for maintaining ionic homeostasis by regulating Na+ and K+ concentrations across the plasma membrane. Emerging evidence indicates that NKA plays a multifaceted role in cardiovascular physiology and pathophysiology beyond its well-established ion-transporting functions. Dysregulation of NKA function has been increasingly linked to cardiovascular diseases such as heart failure, hypertension, and arrhythmias. While the fundamental biological roles of NKA are well-documented, its precise con

Immunoglobulin light chain (AL) amyloidosis is an incurable disease caused by the accumulation and sedimentation of unstable free light chains produced by monoclonal plasma cells. The key to treatment is to achieve a deep hematologic remission in order to improve organ function or reverse organ dysfunction. Conventional treatment has not been able to fully meet the treatment needs of patients with AL, while therapies targeting malignant plasma cells or amyloid have potentially improved treatment outcomes. This study provides an overview of the latest reports on targeted therapies for AL amyloi

Cataracts, characterized by the opacification of the eye lens, remain a leading cause of reversible blindness globally. Age and diabetes are key risk factors, and with the increasing aging and diabetic population, the global burden of cataracts is projected to rise significantly. Current treatment is predominantly surgical; however, pharmacological strategies could offer a non-invasive alternative with the potential to delay, prevent, or even reverse cataract progression. Recent research has enhanced our understanding of cataractogenesis, emphasizing oxidative stress as a key underlying mechan

Calcific aortic valve disease (CAVD) is a common heart valve disease among elderly individuals, and despite its prevalence, the causal mechanisms behind CAVD development remain poorly understood. The current standard treatment for symptomatic aortic stenosis is surgical or transcatheter aortic valve replacement (AVR). Nevertheless, surgical interventions come with disadvantages such as lifelong anticoagulation therapy in patients with mechanical valves and high rates of reoperation due to valve degeneration in adults or somatic growth in pediatric patients. To date, no pharmacological therapy

Interleukin (IL)-16 is expressed mostly by the cells of the human immune system. Upon cell activation IL-16 is cleaved, forming two functional proteins, one regulating cell cycle and the other acting as chemoattractant for the cells carrying CD4 or CD9. Increased levels of IL-16 are found in the circulation and at the sites of inflammation, infection and cancer. Polymorphisms in the IL-16 gene have been coupled to several of these conditions and high IL-16 has been suggested as a disease biomarker. Using unbiased proteomic approach we and others independently identified IL-16 as a biomarker of

This article focuses on ovarian cancer (OC), the most lethal gynecological malignancy, whose risk is influenced by multiple factors, including genetic background, age, reproductive history, parity, obesity status, and smoking habits. Mitochondria, as the core organelles in eukaryotic cells, play a pivotal role in the initiation and progression of OC. In recent years, growing evidence has revealed a close relationship between mitochondrial dysfunction and the accelerated proliferation, enhanced invasiveness, metastasis ability, and therapy resistance of OC. This paper provides an in-depth analy

Breast cancer is the most frequent gynecological malignancy and the main cause of cancer death in the female population worldwide. One of the most significant challenges in its clinical management is the molecular heterogeneity of malignant breast tumors, which is reflected in the current molecular classification of these entities. In each of these tumor molecular subtypes, distinct genetic alterations are involved, and several intracellular signaling pathways contribute to defining their biological identity and clinical response. This literature review summarized the main classic and emerging

Hemophagocytic lymphohistiocytosis (HLH) is a severe and life-threatening hyperinflammatory disorder characterized by dysregulated immune activation, primarily driven by excessive stimulation of cytotoxic lymphocytes (CTLs) and macrophages. This uncontrolled immune response leads to cytokine-induced tissue damage and multiorgan dysfunction. HLH presents a significant clinical challenge due to its rapid progression and high mortality rate. In adult patients, current first-line treatment strategies, adapted from pediatric protocols such as HLH-94 and HLH-2004, achieve complete disease resolution

Sepsis-induced cardiac dysfunction, usually termed sepsis-induced cardiomyopathy or septic cardiomyopathy(SCM), is developed in approximately 70 % of the patients with sepsis, making it is a major concern for sepsis patients. However, the pathogenesis of SCM remain incompletely understood. Ferroptosis, a newly identified mechanism of regulated cell death, characterized by a decline in antioxidant capacity, iron accumulation, and lipid peroxidation(LPO), is involved in sepsis and SCM. Moreover, ferroptosis inhibitors confer a novel therapeutic regimen in SCM. In this Review, we first summarizes
