The role of oleic acid-induced aggregation and glycation modifications in peptide release from duck myofibrillar proteins.

To investigate how oleic acid-induced protein aggregation and glycation alter peptide release and distribution from myofibrillar protein (MPs), an oleic acid-MPs oxidation model was established and subjected to in vitro gastrointestinal digestion. The results showed that under mild oxidation, the looser structure of MPs promoted advanced glycation end products (AGEs) formation without significantly reducing hydrolysis. Under moderate oxidation, MPs transitioned toward dense aggregates, with increased longer peptides. Nε-carboxymethyllysine (CML) and Nε-carboxyethyllysine (CEL) levels rose by 4

Modulation of Cod Myofibrillar Protein Gels by Egg White and Whey Proteins: From Aggregation Kinetics to Gel Mechanics.

In this study, we evaluated how varying concentrations of two exogenous proteins, egg white protein (EWP) and whey protein isolate (WPI) influence heat-induced aggregation and gelation of cod myofibrillar proteins (MPs). Addition of exogenous proteins enhanced the MP solubility and surface hydrophobicity while reducing the turbidity and total sulfhydryl levels. These physicochemical changes occurred during heating in three kinetic phases, an initial rapid phase (0-30 min), a slower transitional phase (30-60 min), and a second rapid aggregation phase (70-90 min) until equilibrium was reached. Impact of protein succinylation and myofibrillar protein-lipid co-oxidation interactions on heterocyclic amine formation of thermally processed goose meat.

This study investigated the mechanisms by which protein succinylation and protein-lipid co-oxidation affect protein structural changes, oxidative reactions, the progression of the Maillard reaction, volatile flavor formation, and heterocyclic amine (HCA) generation as evaluated by chemical model systems. The results showed that, under chemical model systems, succinylation induced partial protein unfolding and slight aggregation, reduced the α-helix content, and promoted protein structural rearrangement. Protein succinylation significantly inhibited protein-lipid co-oxidation, as evidenced by d

Pickering emulsion gels stabilised by ultrasound-treated PSE-like myofibrillar protein microgel particles for age-friendly dysphagia foods: The structural changes, stability and texture-modified gel properties.

To improve the processing suitability of pale, soft and exudative (PSE)-like myofibrillar protein (MP), ultrasound was applied to modulate heat-induced MP aggregates and prepare MP microgel particles (MMPs). The structural and interfacial properties of MMPs were compared with those of large gel particles without ultrasound treatment and native MP. The effects of different MMP concentrations on Pickering emulsion stability, gel texture and swallowing properties were evaluated. Results indicated that MMPs prepared using ultrasound treatment at 600 W for 15 min exhibited more favourable structura

Effects of NaCl partially substituted with KCl or MgCl2 on the properties, oral mastication, in vitro gastric digestion, and pepsin diffusion of myofibrillar protein gel.

The effects of NaCl partially substituted by KCl, MgCl2, and KCl/MgCl2 on the gel characteristics, oral mastication, and in vitro gastric digestion of myofibrillar protein (MP) gel were investigated. The results indicated that partial substitution of NaCl by KCl or MgCl2 did not cause obvious deterioration of gel properties within 30 % substitution levels. MgCl2 substitution significantly increased the surface hydrophobicity of MP, and formed homogeneous network structure for effective limitation of moisture loss. The MgCl2-substituted gel exhibited relatively lower viscosity and hardness for

Determination of a Critical Size Threshold for Volumetric Muscle Loss in the Mouse Quadriceps.

The goal of this study was to determine the threshold for a critically sized, nonhealing muscle defect by characterizing key components in the balance between fibrosis and regeneration as a function of injury size in the mouse quadriceps. There is currently limited understanding of what leads to a critically sized muscle defect and which muscle regenerative components are functionally impaired. With the substantial increase in preclinical VML models as testbeds for tissue engineering therapeutics, defining the critical threshold for VML injuries will be instrumental in characterizing therapeut

Muscle Changes During Atrophy.

Muscle atrophy typically is a direct effect of protein degradation induced by a diversity of pathophysiologic states such as disuse, immobilization, denervation, aging, sepsis, cachexia, glucocorticoid treatment, hereditary muscular disorders, cancer, diabetes and obesity, kidney and heart failure, and others. Muscle atrophy is defined by changes in the muscles, consisting in shrinkage of myofibers, changes in the types of fiber and myosin isoforms, and a net loss of cytoplasm, organelles and overall a protein loss. Although in the literature there are extensive studies in a range of animal mo

A continuum of myofibers in adult rabbit extraocular muscle: force, shortening velocity, and patterns of myosin heavy chain colocalization.

Extraocular muscle (EOM) myofibers do not fit the traditional fiber typing classifications normally used in noncranial skeletal muscle, in part, due to the complexity of their individual myofibers. With single skinned myofibers isolated from rectus muscles of normal adult rabbits, force and shortening velocity were determined for 220 fibers. Each fiber was examined for myosin heavy chain (MyHC) isoform composition by densitometric analysis of electrophoresis gels. Rectus muscle serial sections were examined for coexpression of eight MyHC isoforms. A continuum was seen in single myofiber shorte

Effects of growth potential and growth path on tenderness of beef longissimus muscle from bulls and steers.

The influence of growth potential or growth path on the tenderness of the longissimus muscle was investigated using 117 Angus and Angus-cross bulls and steers raised on pasture over two successive years. Growth rate for a period of 100 d from a weight of about 200 kg was used to identify the faster-growing two-thirds of cattle within the gender groups, half of which were grown fast to a slaughter weight of 530 kg at 16 to 18 mo of age (the Fast group), whereas the other half were restricted in growth (the Restricted group) so they attained a similar final weight as the slower-growing third (th

Anti-endomysial antibodies. A serologic marker of dermatitis herpetiformis.

Direct immunofluorescence (IF) studies of skin biopsies are of value in the diagnosis of most, but not all, cases of dermatitis herpetiformis (DH). Similarly, histologic studies are of help but may be questionable or completely nonspecific. Serologic studies for the presence of IgA-class anti-endomysial antibodies are very specific and are found in 70% of patients with DH and in all untreated patients with celiac disease. The titers of these antibodies are directly associated with the degree of gut disease in these patients. Thus, the presence of these antibodies even in the absence of classic

Mitochondrial abnormalities in some human muscular diseases and in experimental ischemic myopathy.

Morphologic abnormalities have been observed in two cases of human polymyositis and in three cases of ocular myopathies. Similar findings can be observed in experimental ischemic myopathy. "Ragged red" fibres, with anomalous distribution of oxidative enzymes, mitochondrial alterations, with presence of intracristal paracrystalline inclusions and degenerative myofibrillar changes are the similar features. The similarity between some of these alterations, expecially mitochondrial changes, is remarkable, but it is difficult to correlate them to the primary etiology of described human myopathies.

Ionic permeability changes as the basis of the thermal dependence of the resting potential in barnacle muscle fibres.

1. The thermal dependence of the resting potential of isolated barnacle muscle fibres was larger (1-2 mV/ degrees C) than predicted by Nernst's equation (about 0.2 mV/ degrees C). A comparative study was made of the influence on thermal dependence of parameters related to (a) passive permeability and to (b) Na extrusion.2. High [K](o) decreased the thermal dependence reversibly. [K(i)], [Na](i) and [Cl](i) were determined by chemical analysis, and Goldman's equation was fitted to data relating V to [K](o) at different temperatures, in the presence and absence of ouabain 5 x 10(-5)M.
