WDR62 Regulates Early Neural and Glial Progenitor Specification of Human Pluripotent Stem Cells.

Mutations in WD40-repeat protein 62 (WDR62) are commonly associated with primary microcephaly and other developmental cortical malformations. We used human pluripotent stem cells (hPSC) to examine WDR62 function during human neural differentiation and model early stages of human corticogenesis. Neurospheres lacking WDR62 expression showed decreased expression of intermediate progenitor marker, TBR2, and also glial marker, S100β. In contrast, inhibition of c-Jun N-terminal kinase (JNK) signalling during hPSC neural differentiation induced upregulation of WDR62 with a corresponding increase in n

Impaired response inhibition and excess cortical thickness as candidate endophenotypes for trichotillomania.

Trichotillomania is characterized by repetitive pulling out of one's own hair. Impaired response inhibition has been identified in patients with trichotillomania, along with gray matter density changes in distributed neural regions including frontal cortex. The objective of this study was to evaluate impaired response inhibition and abnormal cortical morphology as candidate endophenotypes for the disorder. Subjects with trichotillomania (N = 12), unaffected first-degree relatives of these patients (N = 10), and healthy controls (N = 14), completed the Stop Signal Task (SST), a measure of respo

High-definition transcranial direct current stimulation over right dorsolateral prefrontal cortex differentially modulates inhibitory mechanisms for speech vs. limb movement.

Evidence suggests that planning and execution of speech and limb movement are subserved by common neural substrates. However, less is known about whether they are supported by a common inhibitory mechanism. P3 event-related potentials (ERPs) is a neural signature of motor inhibition, which are found to be generated by several brain regions including the right dorsolateral prefrontal cortex (rDLPFC). However, the relative contribution of rDLPFC to the P3 response associated with speech versus limb inhibition remains elusive. We investigated the contribution of rDLPFC to the P3 underlying speech

Go/No Go task performance predicts cortical thickness in the caudal inferior frontal gyrus in young adults with and without ADHD.

Response inhibition deficits are widely believed to be at the core of Attention-Deficit Hyperactivity Disorder (ADHD). Several studies have examined neural architectural correlates of ADHD, but research directly examining structural correlates of response inhibition is lacking. Here we examine the relationship between response inhibition as measured by a Go/No Go task, and cortical surface area and thickness of the caudal inferior frontal gyrus (cIFG), a region implicated in functional imaging studies of response inhibition, in a sample of 114 young adults with and without ADHD diagnosed initi

Age-Related Changes in E/I Balance Estimated With Hurst Exponent and Links With Developmental Skills in Preschool Children.

Cortical excitation-inhibition (E/I) balance regulates sensitive periods of plasticity and is critical for brain development. Yet, neuroimaging studies characterizing E/I dynamics in early childhood are rare. The Hurst exponent (H), derived from resting-state functional magnetic resonance imaging (rs-fMRI), provides a noninvasive proxy for E/I balance. We investigated age-related changes in H using longitudinal rs-fMRI from 55 typically developing children aged 1.5-5 years (78 sessions during natural sleep). Linear mixed-effects models revealed significant age-related increases in H, signifyin

Inhibitory control-related neural pathways of early adversity to internalizing problems in preadolescence.

OBJECTIVE: Exposure to early life adversity has long-reaching effects on the psychopathology of adolescents. However, some individuals exposed to early life stressors, such as threat and deprivation, do not later present with symptoms. Previous research suggests that neural mechanisms may play a role in differentiating pathways from early life threat or deprivation to internalizing symptoms. METHOD: This study utilizes neuroimaging to examine the relationship between early adversity experiences and internalizing symptoms later on. Data from the ABCD® study were used to investigate whether inh

Frequency-Dependent Inhibition during Deep Brain Stimulation of Thalamic Ventral Intermediate Nuclei.

Deep brain stimulation (DBS) of the thalamic ventral intermediate nucleus (Vim) has been a standard therapy for essential tremor. It has been shown that high-frequency (≥100 Hz) DBS suppresses Vim firing and tremor activity; however, the underlying mechanisms are not fully understood. Here, we investigate whether neuronal suppression during high-frequency DBS occurs at cellular levels or is influenced by network-level effects. Using in vivo recordings of Vim neurons, from people of unknown sex, during different DBS frequencies, we detected a positive-going evoked-field potential, quasi-evoked

Midbrain dopamine loss drives parvalbumin interneuron vulnerability through tissue plasminogen activator-linked perineuronal-net breakdown and hippocampal disinhibition.

Midbrain dopaminergic degeneration is an early feature of Alzheimer's Disease (AD), dementia with Lewy bodies (DLB), and AD-Parkinson's disease overlap (AD-PD). However, its direct contribution to the failure of hippocampal inhibitory-circuits, a pathological feature shared across these conditions, remains unresolved. Parvalbumin-positive interneurons (PV-INs) regulate hippocampal excitation-inhibition balance and are directly modulated by dopamine (DA). These neurons are protected by perineuronal nets (PNNs), extracellular-matrix structures supporting fast GABAergic signaling and neuronal res

Elevated body mass index in youth is associated with neural disinhibition and internetwork functional dysconnectivity: A magnetoencephalography study.

OBJECTIVE: The developing brain has a vulnerability to the negative impact of obesity and excessive consumption of hyperpalatable foods. Animal models suggest changes in the excitation:inhibition balance of the brain in obesogenic diets, including disinhibition and hyperexcitability, and altered connectivity between brain regions. METHODS: We investigated the neurophysiological effects of overweight and obesity in 32youth (8-19 years), using a naturalistic viewing protocol with magnetoencephalography (MEG). Subjects were recruited into two groups based on normalised / z-scored body mass index

Selective Optogenetic Inhibition of Slow Conducting Fibers at the Level of the Sciatic Nerve Trunk in the Mouse.

OBJECTIVE: Peripheral nerve stimulation is a drug-free alternative for chronic pain management, suppressing nociception through gating mechanisms in the spine. However, excitation through electrical stimulation does not easily discriminate between sensory and motor fibers or their functional subtypes, and can cause off-target effects. Targeted optogenetic inhibition may be a more selective method to suppress nociceptive activity directly while leaving neighboring fibers unaffected. MATERIALS AND METHODS: In this study, the chloride-specific light-gated ion pump halorhodopsin (enhanced Natrono

Cell type-specific and frequency-dependent centrifugal modulation in olfactory bulb output neurons in vivo.

Mitral/tufted cells (M/TCs) form complex local circuits with interneurons in the olfactory bulb and are powerfully inhibited by these interneurons. The horizontal limb of the diagonal band of Broca (HDB), the only GABAergic/inhibitory source of centrifugal circuit with the olfactory bulb, is known to target olfactory bulb interneurons, and we have shown targeting also to olfactory bulb glutamatergic neurons in vitro. However, the net efficacy of these circuits under different patterns of activation in vivo and the relative balance between the various targeted intact local and centrifugal circu

Evidence of altered corticomotor inhibition in older adults with a history of repetitive neurotrauma. A transcranial magnetic stimulation study.

International concern continues regarding the association between the long-term neurophysiologic changes from repetitive neurotrauma associated with contact and collision sports. This study describes corticomotor changes in retired contact/collision sport athletes and controls, between the ages of 30 and 70 years. Retired athletes (n = 152; 49.1 ± 8.5 years) and controls (n = 72; 47.8 ± 9.5 years) were assessed using single and paired-pulse transcranial magnetic stimulation (TMS) for active motor threshold (aMT), motor evoked potential and cortical silent period duration (expressed as MEP:cSP

Toxoplasma infection induces microglia-neuron contact and the loss of perisomatic inhibitory synapses.

Infection and inflammation within the brain induces changes in neuronal connectivity and function. The intracellular protozoan parasite, Toxoplasma gondii, is one pathogen that infects the brain and can cause encephalitis and seizures. Persistent infection by this parasite is also associated with behavioral alterations and an increased risk for developing psychiatric illness, including schizophrenia. Current evidence from studies in humans and mouse models suggest that both seizures and schizophrenia result from a loss or dysfunction of inhibitory synapses. In line with this, we recently repor

Application of long-interval paired-pulse transcranial magnetic stimulation to motion-sensitive visual cortex does not lead to changes in motion discrimination.

The perception of visual motion is dependent on a set of occipitotemporal regions that are readily accessible to neuromodulation. The current study tested if paired-pulse Transcranial Magnetic Stimulation (ppTMS) could modulate motion perception by stimulating the occipital cortex as participants viewed near-threshold motion dot stimuli. In this sham-controlled study, fifteen subjects completed two sessions. On the first visit, resting motor threshold (RMT) was assessed, and participants performed an adaptive direction discrimination task to determine individual motion sensitivity. During the

Imbalance of laminar-specific excitatory and inhibitory circuits of the orbitofrontal cortex in autism.

BACKGROUND: The human orbitofrontal cortex (OFC) is involved in assessing the emotional significance of events and stimuli, emotion-based learning, allocation of attentional resources, and social cognition. Little is known about the structure, connectivity and excitatory/inhibitory circuit interactions underlying these diverse functions in human OFC, as well as how the circuit is disrupted in individuals with autism spectrum disorder (ASD). METHODS: We used post-mortem brain tissue from neurotypical adults and individuals with ASD. We examined the morphology and distribution of myelinated axo

Beneficial effects of estrogens in obstructive sleep apnea hypopnea syndrome.

Epidemiological studies showing the higher frequency of obstructive sleep apnea hypopnea syndrome in men, polycystic ovary syndrome (PCOS), and in post-menopausal women suggest the beneficial role of estrogen. These findings are well supported by the pre-clinical studies (ten research studies described in this review) showing that estrogen and phytoestrogens attenuate the deleterious effects of chronic intermittent hypoxia (obstructive apnea in animals) on the genioglossal muscles and on other organs (co-morbidities) in ovariectomized rodents. Moreover, clinical studies (four research studies

Binocular deprivation induces both age-dependent and age-independent forms of plasticity in parvalbumin inhibitory neuron visual response properties.

Activity of cortical inhibitory interneurons is rapidly reduced in response to monocular deprivation during the critical period for ocular dominance plasticity and in response to salient events encountered during learning. In the case of primary sensory cortex, a decrease in mean evoked firing rate of parvalbumin-positive (PV) inhibitory neurons is causally linked to a reorganization of excitatory networks following sensory perturbation. Converging evidence indicates that it is deprivation, and not an imbalance between open- and closed-eye inputs, that triggers rapid plasticity in PV neurons.

Does response inhibition have pre- and postdiagnostic utility in Parkinson's disease?

Parkinson's disease (Pd) is the second most prevalent degenerative neurological condition worldwide. Improving and sustaining quality of life is an important goal for Parkinson's patients. Key areas of focus to achieve this goal include earlier diagnosis and individualized treatment. In this review the authors discuss impulse control in Pd and examine how measures of impulse control from a response inhibition task may provide clinically useful information (a) within an objective test battery to aid earlier diagnosis of Pd and (b) in postdiagnostic Pd, to better identify individuals at risk of

Methodology for studying the transient effects of transcranial direct current stimulation combined with auditory residual inhibition on tinnitus.

BACKGROUND: Transcranial direct current stimulation (tDCS) and auditory residual inhibition (RI) are short lasting non-invasive techniques that can suppress tinnitus in some people. A pilot study was undertaken to evaluate methods to measure a potential additive effect of these treatments. NEW METHOD: Ten participants (mean age 59 years) with chronic tinnitus underwent four sessions of anodal tDCS of the left temporoparietal area(LTA) combined with 1min of auditory stimulation (broadband noise presented at tinnitus minimum masking level [MML]+10 dB), either before, during or immediately after

Structural remodeling of the heart and its premotor cardioinhibitory vagal neurons following T(5) spinal cord transection.

Midthoracic spinal cord injury (SCI) is associated with enhanced cardiac sympathetic activity and reduced cardiac parasympathetic activity. The enhanced cardiac sympathetic activity is associated with sympathetic structural plasticity within the stellate ganglia, spinal cord segments T1-T4, and heart. However, changes to cardiac parasympathetic centers rostral to an experimental SCI are relatively unknown. Importantly, reduced vagal activity is a predictor of high mortality. Furthermore, this autonomic dysregulation promotes progressive left ventricular (LV) structural remodeling. Accordingly,
