Neuropeptide Y-immunoreactive primary afferents in the dental pulp and periodontal ligament following nerve injury to the inferior alveolar nerve in the rat.

The distribution of neuropeptide Y (NPY)-immunoreactive (IR) primary afferents in the dental pulp and periodontal ligament of the rat mandible were examined following combined chronic constriction injury (CCI) of the inferior alveolar nerve (IAN) and sympathectomy of the superior cervical ganglion (SCG). NPY-IR nerve fibers were observed around the blood vessels in the trigeminal ganglion, dental pulp and periodontal ligament in normal animals. Following combined CCI of the IAN and sympathectomy of SCG (SCGx), perivascular NPY-IR nerve fibers originating from SCG disappeared completely, but ma

Dentin phosphoprotein gene locus is not associated with dentinogenesis imperfecta types II and III.

Dentinogenesis imperfecta (DGI) is an autosomal dominant inherited dental disease which affects dentin production and mineralization. Genetic linkage studies have been performed on several multigeneration informative kindreds. These studies determined linkage between DGI type II and III and group-specific component (vitamin D-binding protein). This gene locus has been localized to the long arm of human chromosome 4 in the region 4q11-q21. Although this disease has been mapped to chromosome 4, the defective gene product is yet to be determined. Biochemical studies have suggested abnormal levels

Human root caries: histopathology of arrested lesions.

The histopathology of arrested root caries lesions was examined in extracted human teeth. The main structural characteristics of arrested lesions were the completely mineralized surface area and the formation of a distinct sclerosis of the dentinal tubules. Intertubular dentin was, with the exception of the dentinal tubules, fully mineralized up to the surface. Dentinal tubules near the surface were either filled with ghosts of microorganisms or with crystals of different shapes. Sclerosis of the dentinal tubules was characterized by the presence of three different patterns of intratubular min

In vitro effects of calcitonin and/or parathyroid hormone on odontogenesis of mouse embryonic molars.

Mandibular first molars of mouse embryos were cultivated for examination of the effects of calcitonin (CT) and/or parathyroid hormone (PTH) on the odontogenesis of the molars, and for determination of whether and how CT, which is a PTH antagonist, has an influence on the effect of PTH on odontogenesis. On the second day, the inner enamel epithelium in the control group had already differentiated into pre-ameloblasts. Typical odontoblasts had secreted a layer of predentin. On the fourth day of culture, the pre-ameloblasts achieved terminal differentiation into secretory ameloblasts, and enamel …

Reaction of the human dental pulp to silver amalgam restorations. The effect of enclosing amalgam of initially high plasticity in intermediate depth or deep cavities.

From earlier studies using amalgam of known physical and sealing properties it was concluded that the initially high mercury content was the main reason for pulpal changes and that clinical leakage had been less important. The purpose of the present investigation was to study how a possible sealing insufficiency may contribute to the pulpal changes beneath restorations of silver amalgam. The influence of bacteria at the tooth/filling interface upon pulpal changes was also studied. The material consisted of 21 contralateral pairs of premolars. Intermediate depth cavities were prepared in 13 pai

Histological comparison of pulpal inflammation in primary teeth with occlusal or proximal caries.

OBJECTIVE: A number of clinical and histological studies have investigated caries-related changes in the primary tooth pulp, but the effect of caries site, as a clinical variable, has not been previously considered. This study sought to compare inflammatory changes within the pulp of primary molars according to the location of the caries lesion (occlusal or proximal). METHODS: Eighty-three primary molars were extracted under general anaesthesia for caries and/or orthodontic reasons, and were split immediately after removal and fixed in 10% formalin. Teeth were then decalcified, sectioned, and

Preserving the vital pulp in operative dentistry: 4. Factors influencing successful pulp capping.

The sequence of factors that mediate pulp inflammation and necrosis are unclear, and controversy surrounds the effects of different pulp capping materials on exposed pulps. Clinicians have few quantitative studies that rank the in vivo pulp capping effects of commonly used restorative materials.

Dentin sialoprotein acts as an angiogenic factor through association with the membrane receptor endoglin.

Dentin sialophosphoprotein (DSPP) is highly expressed by odontoblasts, the cell type responsible for dentin formation. DSPP therefore has been extensively studied as a regulator of dentinogenesis. Besides defective dentinogenesis in teeth, Dspp-deficient mice also display reduced blood vessels in the transition zone of femurs. However, the exact role and underlying mechanisms of DSPP in the process of blood vessel formation remain enigmatic. Here, we show that dentin sialoprotein (DSP), the NH2-terminal cleavage product of DSPP, promotes the migration and capillary-like structure formation of …

"Dental Pulp-On-Chip" Microfluidic Devices for Emulating Human Dental Pulp Tissue Physiology and Pathology.

The dental pulp is a highly vascularized and innervated connective tissue located at the central part of the tooth and composed of a diverse array of cell types, including fibroblasts, multipotent mesenchymal stem cell, odontoblasts, and immune cells. Traumatic injuries and carious lesions can lead to dental pulp pathologies that often require replacement of damaged tissue with inert materials. Despite significant progress in recent years, cell-based dental pulp regenerative therapies remain distant from clinical applications. The lack of platforms capable of accurately modeling the human dent…

A labeling study of dentin formation rates during crown and root growth of porcine mandibular first molars.

We used fluorochrome labeling to study spatiotemporal variation of dentin apposition (DAR) and extension (DER) rates during crown and root f … t, which occurred earlier in the domestic pigs. We consider the more rapid recruitment of secretory odontoblasts in domestic pigs (reflected by higher DER) a side effect of selection for rapid body growth during pig domestication.

Schwann cell precursors in health and disease.

Schwann cell precursors (SCPs) are frequently regarded as neural crest-derived cells (NCDCs) found in contact with axons during nerve format … fin cells) and non-neural-like (mesenchymal stromal cells, able e.g., to generate dentine-producing odontoblasts) cell types. Until now no reports of endoderm-like derivatives were reported so far. Interestingly, in the Schwann cell lineage only early SCPs are likely able to differentiate into melanocytes and bone marrow mesenchymal stromal cells. We have also …

Nucleus-targeted Dmp1 transgene fails to rescue dental defects in Dmp1 null mice.

Dentin matrix protein 1 (DMP1) is essential to odontogenesis. Its mutations in human subjects lead to dental problems such as dental deformi … nd. Although immunohistochemistry demonstrated that (NLS)DMP1 was localized in the nuclei of the preodontoblasts and odontoblasts, the histological, morphological and biochemical analyses showed that it failed to rescue the dental and periodontal defects as well as the delayed tooth eruption in Dmp1 null mice. These data suggest that the full-length DMP1 plays …

SM50 repeat-polypeptides self-assemble into discrete matrix subunits and promote appositional calcium carbonate crystal growth during sea urchin tooth biomineralization.

The two major proteins involved in vertebrate enamel formation and echinoderm sea urchin tooth biomineralization, amelogenin and SM50, are both characterized by elongated polyproline repeat domains in the center of the macromolecule. To determine the role of polyproline repeat polypeptides in basal deuterostome biomineralization, we have mapped the localization of SM50 as it relates to crystal growth, conducted self-assembly studies of SM50 repeat polypeptides, and examined their effect on calcium carbonate and apatite crystal growth.
