Twins and the paradox of dental-age estimations: a caution for researchers and clinicians.

The biological age difference among twins is frequently an issue in studies of genetic influence on various dental features, particularly dental development. The timing of dental development is a crucial issue also for many clinicians and researchers. The aim of this study was therefore to verify within groups of twins how dental development differs, by applying Demirjian's method, Mincer's charts of development of third molars and two of Cameriere's methods for dental age estimation, which are among the most popular methods both in the clinical and the forensic scenario. The sample consisted

Patterns of asymmetry in primary tooth emergence of Australian twins.

AIMS: This study is part of a larger investigation of genetic and environmental influences on primary tooth emergence in Australian twins. Our aims were to describe patterns of emergence asymmetry, including directional and fluctuating components (DA, FA), and to test for a genetic basis to observed asymmetry. METHODS: The study sample consisted of 131 twin pairs. Using one randomly-selected twin from each pair, dental asymmetry was examined by analysing the number of days between emergence of antimeres (Delta), with dates of emergence provided through parental recording. Scatterplots were us

Uses of calcium hydroxide: an online study guide.

The Editorial Board of the Journal of Endodontics has developed a literature-based study guide of topical areas related to endodontics. This study guide is intended to give the reader a focused review of the essential endodontic literature and does not cite all possible articles related to each topic. Although citing all articles would be comprehensive, it would defeat the idea of a study guide. This section presents uses of calcium hydroxide related to apexogenesis, apexification, direct pulp cap, indirect pulp cap, internal resorption that has perforated laterally, intracanal medication, con

Odontogenic ameloblast-associated protein nature of the amyloid found in calcifying epithelial odontogenic tumors and unerupted tooth follicles.

We have previously reported that the amyloid found in three patients with calcifying epithelial odontogenic tumors (CEOT) was composed of N-terminal fragments of a putative 153-residue protein specified by a gene designated FLJ20513 now known to represent exons 5 through 10 of the odontogenic ameloblast-associated protein (ODAM) locus that encodes a 279-residue polypeptide. Confirmation of the amyloidogenic potential of ODAM has resulted from analyses of four other cases where we found, in addition, a 74-residue segment specified by exon 4. Through preparation of ODAM-related synthetic peptide

Aberrant root formation: review of root genesis and three case reports.

The mechanism of root formation and tooth eruption is a complex process which is not fully understood. Prior to a tooth emerging into the oral cavity, root genesis is initiated by derivatives of the enamel organ. The dental follicle mediates an eruption pathway allowing for movement of the developing tooth in a coronal direction. As the tooth moves towards the oral cavity, root formation occurs passively in the resulting space. Failure of the enamel organ and dental follicle to properly coordinate may result in complications in the eruption process. This clinical report presents 3 cases of iso

Molecular basis of non-syndromic tooth agenesis: mutations of MSX1 and PAX9 reflect their role in patterning human dentition.

OBJECTIVE: Although the platelet destruction shows a primary role in the thrombocytopenia of idiopathic thrombocytopenic purpura (ITP), it has been demonstrated that impaired platelet production may also contribute to the severity of thrombocytopenia in ITP. The present study examined megakaryocyte apoptosis in bone marrow aspirates of children with acute and chronic ITP and investigated the role of megakaryocyte apoptosis in ITP pathophysiology. METHODS: Thirteen children diagnosed with acute ITP and eight children diagnosed with chronic ITP comprised the study group. Ten children, who were

The developing enamel matrix: nature and function.

The hydroxyapatite crystals of mature enamel are unusually large, uniform and regularly disposed within the tissue, implying that their development is a highly controlled process. The organic matrix of developing enamel is presumed to play an important role in the modulation of mineral deposition and growth during tooth morphogenesis but the precise functions of individual matrix proteins remain unclear. The aim of this review was to survey the current knowledge of enamel matrix proteins with a view to suggesting possible functions. The organic matrix is highly heterogeneous, comprising protei

Accelerated eruption of rat lower incisor. Relationship between impeded and unimpeded eruption rates, rate of attrition, tooth length, and production of dentin and enamel.

The present investigation studies the effects of persistently cutting one lower rat incisor out of occlusion. Within four days, the rate of eruption of the cut (unimpeded) incisor increased to 216% and that of the uncut (impeded) contralateral to 136% of the baseline rate. While the former remained high, the latter decreased gradually to about 90% within three weeks. The rate of attrition of the impeded incisor increased to 233% of the baseline rate within two days, then fell abruptly, and remained at a slightly lower level than the rate of eruption. Accordingly, the length of the erupted part

Morphology and cytidine-5'-monophosphatase cytochemistry of odontogenic epithelium at the enamel-free areas of mouse molars.

This epithelium in developing first and second molars of 1-15 days old balb/c mice was examined by light and electron microscopy. The lysosomal content of these epithelial cells was assessed using CMPase as a marker. At the early developmental stages (4-7 days), the epithelial cells at the enamel-free areas had morphological features of secretory cells. A distinct afibrillar layer was identified covering the enamel-free dentine surfaces. This layer was morphologically different from both the underlying dentine and the enamel at its periphery. At a later stage (9 days), the epithelial cells had

Developmental defects of enamel and dentin in individuals born preterm: a scoping review of evidence and mechanisms.

BACKGROUND: Preterm birth (<37 weeks of gestation) occurs during critical stages of odontogenesis and is associated with developmental defects in dental hard tissues. Enamel hypoplasia, enamel hypomineralization, and structural dentin alterations may compromise esthetics, function, and long-term oral health outcomes. However, the nature and extent of these defects and their underlying mechanisms have not been comprehensively investigated. OBJECTIVE: To map and evaluate the available evidence regarding developmental defects of enamel and dentin in individuals born preterm, identify proposed pa…

OCT4-driven PER1 activation promotes odontogenic differentiation in senescent SCAP spheres.

Mesenchymal stem cells (MSCs) hold significant promise for applications in regenerative medicine, yet their therapeutic potential is often limited by replicative senescence. Identifying effective strategies to reverse replicative senescence in MSCs and elucidating the underlying molecular mechanisms are essential steps in advancing their clinical use. Here, this study demonstrated that the pluripotency regulator octamer-binding transcription factor 4 (OCT4) promoted odontogenic differentiation by activating period circadian regulator 1 (PER1) in replicative senescent stem cells from apical pap

Frequency and associated factors of segmental root developmental arrest in endodontically treated immature permanent teeth with apical periodontitis: a retrospective cohort study.

BACKGROUND: Segmental root developmental arrest (SRDA) is an uncommon developmental pattern in immature permanent teeth with apical periodontitis, in which a separated apical segment continues to develop independently of the main root. This retrospective study evaluated the frequency, clinical features, and factors associated with SRDA after regenerative endodontic procedures (REPs) or apexification. METHODS: A total of 103 immature permanent teeth with apical periodontitis treated with REPs or apexification were retrospectively reviewed. Clinical records and radiographs were assessed for SRD

Mutanobactin-D, a Streptococcus mutans Non-Ribosomal Cyclic Lipopeptide, Induces Osteogenic/Odontogenic Differentiation of Human Dental Pulp Stem Cells and Human Bone Marrow Stem Cells.

Bacterium-triggered carious lesions implicate dental hard tissue destruction and the simultaneous initiation of regenerative events comprising dental stem cell activation. Streptococcus mutans (S. mutans) is a prominent pathogen of the oral cavity and the principal cause of caries. S. mutans generates complex products involved in interbacterial interactions, including Mutanobactin-D (Mub-D), which belongs to a group of non-ribosomal cyclic lipopeptides. In the present study, we aimed to analyse the potential role of the synthetic Mub-D peptide in cell populations involved in tissue regenerativ
