Damage-associated responses of the host contribute to defence against cyst nematodes but not root-knot nematodes.

When nematodes invade and subsequently migrate within plant roots, they generate cell wall fragments (in the form of oligogalacturonides; OGs) that can act as damage-associated molecular patterns and activate host defence responses. However, the molecular mechanisms mediating damage responses in plant-nematode interactions remain unexplored. Here, we characterized the role of a group of cell wall receptor proteins in Arabidopsis, designated as polygalacturonase-inhibiting proteins (PGIPs), during infection with the cyst nematode Heterodera schachtii and the root-knot nematode Meloidogyne incog

Benzothiadiazole induces disease resistance in Arabidopsis by activation of the systemic acquired resistance signal transduction pathway.

Benzothiadiazole (BTH) is a novel chemical activator of disease resistance in tobacco, wheat and other important agricultural plants. In this report, it is shown that BTH works by activating SAR in Arabidopsis thaliana. BTH-treated plants were resistant to infection by turnip crinkle virus, Pseudomonas syringae pv 'tomato' DC3000 and Peronospora parasitica. Chemical treatment induced accumulation of mRNAs from the SAR-associated genes, PR-1, PR-2 and PR-5. BTH treatment induced both PR-1 mRNA accumulation and resistance against P. parasitica in the ethylene response mutants, etr1 and ein2, and

Elucidating the immunomodulatory effects of phytoestrogens and their groundbreaking applications in transplantation medicine.

Phytoestrogens are natural compounds found in plants and plant-based foods. When ingested, they can affect the human body in the same way as estrogen produced by the body. Phytoestrogens affect the regulation, differentiation, and production of immune cells. People who consume polyphenol and flavonoid-rich foods have lower incidences of inflammation, autoimmune diseases, and cancer. In organ transplantation, immune rejection is a lifelong problem for patients. In clinical practice, acute rejection is treated with hormonal shock or immunosuppressive drugs. However, effective reversal measures f

Control of H2S synthesis by the monomer-oligomer transition of OsCBSX3 for modulating rice growth-immunity balance.

Hydrogen sulfide (H2S) is recognized as an important gaseous signaling molecule, similar to nitric oxide and carbon monoxide. However, less is known about the biosynthetic mechanism of H2S in plants and its role in plant-pathogen interactions. Here, we show that H2S induces the bursts of reactive oxygen species and upregulates the expression of defense-related genes in rice. However, excessive H2S concentrations inhibit rice growth. We found that the cystathionine β-synthase OsCBSX3 regulates rice growth and resistance to bacteria pathogens, Xanthomonas oryzae pv. oryzicola (Xoc) and X. oryzae

Differential effects of plant ontogeny and damage type on phloem and foliage monoterpenes in jack pine (Pinus banksiana).

Coniferous trees have both constitutive and inducible defences that deter or kill herbivores and pathogens. We investigated constitutive and induced monoterpene responses of jack pine (Pinus banksiana Lamb.) to a number of damage types: a fungal associate of the mountain pine beetle (Dendroctonus ponderosae Hopkins), Grosmannia clavigera (Robinson-Jeffrey & R.W. Davidson); two phytohormones, methyl jasmonate (MJ) and methyl salicylate (MS); simulated herbivory; and mechanical wounding. We only included the fungal, MJ and mechanical wounding treatments in the field experiments while all treatme

Advances in Research on the Involvement of Selenium in Regulating Plant Ecosystems.

Selenium is an essential trace element which plays an important role in human immune regulation and disease prevention. Plants absorb inorganic selenium (selenite or selenate) from the soil and convert it into various organic selenides (such as seleno amino acids, selenoproteins, and volatile selenides) via the sulfur metabolic pathway. These organic selenides are important sources of dietary selenium supplementation for humans. Organoselenides can promote plant growth, improve nutritional quality, and play an important regulatory function in plant ecosystems. The release of selenium-containin

Antiallergic effects of Lactobacillus pentosus strain S-PT84 mediated by modulation of Th1/Th2 immunobalance and induction of IL-10 production.

BACKGROUND: Many types of fermented food are consumed in Japan. Although some are produced by plant-origin lactic-acid bacteria (LAB) fermentation, the physiological functions of such bacteria remain unclear. We therefore isolated LAB of plant origin from Kyoto pickles and determined the immunological activity of heat-killed preparations of plant-origin LAB. METHODS: The Lactobacillus pentosus strain S-PT84 was selected from among 16 LAB of plant origin as the strongest interleukin (IL)-12-inducing strain. IL-12- and IL-10-inducing activities were determined with macrophages from BALB/c mice.

Multiple BnaSOBIR1-Associated Receptor-like Proteins Contribute to Enhanced Resistance to Sclerotinia sclerotiorum in Brassica napus L. (Oilseed Rape).

As a major fungal pathogen of Brassica napus, Sclerotinia sclerotiorum causes significant yield reductions worldwide. Receptor-like proteins (RLPs) are essential components of plant immunity, but the functions of many RLPs in B. napus still remain unclear. In this study, three BnaSOBIR1-interacting leucine-rich repeat RLPs (LRR-RLPs), named BnaRLP-G13-2, BnaRLP-G13-3, and BnaRLP-G13-4, were identified by yeast two-hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays. Promoter analysis revealed diverse cis-acting elements involved in stress and phytohormone responses. Overexp…

Cysteine-rich receptor-like kinases OsCRK19 and OsCRK22 interact with OsRbohI to modulate ROS production and disease resistance in rice.

Reactive oxygen species (ROS) burst is a hallmark response of plant innate immunity. ROS production must be under tight control since inappropriate accumulation is toxic. In rice, OsRbohI is the key NADPH oxidase responsible for ROS generation. However, the upstream kinases regulating OsRbohI activity remain unknown. Here, we identified two cysteine-rich receptor-like kinases, OsCRK19 and OsCRK22, as regulators of OsRbohI. OsCRK19 and OsCRK22 bind to N- both termini of OsRbohI. OsCRK19/22-OsRbohI interactions are constitutive and not responding to PAMP elicitation. OsCRK19 and OsCRK22 phosphor

The roles of ginseng and ginsenosides for immune regulation: Relation to immunometabolic responses.

Ginseng, a traditional medicinal plant with a long history of clinical application has garnered increasing attention due to its broad immunomodulatory properties. Its principal bioactive component ginsenosides regulate diverse immune activity through complex molecular and cellular mechanisms. Despite growing evidence supporting the role of ginseng in modulating innate and adaptive immunity, current studies remain largely focused on individual immune cell types or isolated signaling pathways. A unifying framework that integrates ginseng-mediated immunometabolic regulation with coordinated immun

Severe infectious diseases of childhood as monogenic inborn errors of immunity.

This paper reviews the developments that have occurred in the field of human genetics of infectious diseases from the second half of the 20th century onward. In particular, it stresses and explains the importance of the recently described monogenic inborn errors of immunity underlying resistance or susceptibility to specific infections. The monogenic component of the genetic theory provides a plausible explanation for the occurrence of severe infectious diseases during primary infection. Over the last 20 y, increasing numbers of life-threatening infectious diseases striking otherwise healthy c

The Blumeria graminis Effector BgtH12 Suppresses Wheat Immunity and Acts as a Virulence Factor.

The obligate biotrophic fungus Blumeria graminis f. sp. tritici (Bgt), the causal agent of wheat powdery mildew, secretes effector proteins to suppress plant immunity and establish infection. Here, we characterize BgtH12, a conserved small secreted protein that is highly expressed during early infection (peaking at 48 hpi). Functional analysis using host induced gene silencing (HIGS) with two independent fragments achieved 54-82% knockdown of BgtH12 transcripts, resulting in significantly reduced fungal sporulation, hyphal length, and branching. Stable transgenic RNAi lines confirmed these fin…

Prevention of bubonic and pneumonic plague using plant-derived vaccines.

Yersinia pestis, the causative agent of bubonic and pneumonic plague, is an extremely virulent bacterium but there are currently no approved vaccines for protection against this organism. Plants represent an economical and safer alternative to fermentation-based expression systems for the production of therapeutic proteins. The recombinant plague vaccine candidates produced in plants are based on the two most immunogenic antigens of Y. pestis: the fraction-1 capsular antigen (F1) and the low calcium response virulent antigen (V) either in combination or as a fusion protein (F1-V). These antige

Constraints on effectiveness of cyanogenic glycosides in herbivore defense.

Cyanogenesis is the process by which hydrogen cyanide is released from endogenous cyanide containing compounds. Many cyanogenic plants release HCN in sufficient quantities to be toxic and, as a result, tend to be avoided by herbivores. However, there are many exceptions with some herbivores either immune to the cyanogenic status of the plant, or in some cases attracted to cyanogenic plants. This has led to a certain degree of scepticism regarding the role of cyanogenic glycosides as defense compounds. In this review, we examine evidence showing that differences in the effectiveness of cyanogen

Integrated multi-omics and genetic analyses reveal molecular determinants underlying Arabidopsis snap33 mutant phenotype.

The secretory pathway is essential for plant immunity, delivering diverse antimicrobial molecules into the extracellular space. Arabidopsis thaliana soluble N-ethylmaleimide-sensitive-factor attachment protein receptor SNAP33 is a key actor of this process. The snap33 mutant displays dwarfism and necrotic lesions, however the molecular determinants of its macroscopic phenotypes remain elusive. Here, we isolated several new snap33 mutants that exhibited constitutive cell death and H2O2 accumulation, further defining snap33 as an autoimmune mutant. We then carried out quantitative transcriptomic…

Rice ubiquitin-conjugating enzyme OsUbc13 negatively regulates immunity against pathogens by enhancing the activity of OsSnRK1a.

Ubc13 is required for Lys63-linked polyubiquitination and innate immune responses in mammals, but its functions in plant immunity still remain largely unknown. Here, we used molecular biological, pathological, biochemical, and genetic approaches to evaluate the roles of rice OsUbc13 in response to pathogens. The OsUbc13-RNA interference (RNAi) lines with lesion mimic phenotypes displayed a significant increase in the accumulation of flg22- and chitin-induced reactive oxygen species, and in defence-related genes expression or hormones as well as resistance to Magnaporthe oryzae and Xanthomonas …

Protective perfumes: the role of vegetative volatiles in plant defense against herbivores.

Herbivore damage to leaves and other vegetative tissues often stimulates the emission of volatile compounds, suggesting that these substances have a role in plant defense. In fact, ample evidence has accumulated in the last few years indicating that volatiles from vegetative plant parts can directly repel herbivores, such as ovipositing butterflies and host-seeking aphids. Volatiles have also been demonstrated to protect plants by attracting herbivore enemies, such as parasitic wasps, predatory arthropods and possibly even insectivorous birds. Even below ground herbivory results in the release

[Systemic resistance induced by biocontrol agents in plants and its biochemical and cytological mechanisms].

Microbial biocontrol agents (BCAs) are generally used for controlling plant diseases via antagonistic mechanisms including competition, antibiosis, parasitism, and cross-protection. Some BCAs can even promote plant growth, and provide induced systemic resistance (ISR), i. e., induce the plants to have resistance against pathogens including phytopathogenic fungi, bacteria and virus, and in some cases, pest insects and nematodes. ISR is characterized by non-specific, wide spectrum and systemic. It is phenotypically similar to the systemic acquired resistance (SAR) induced by the infection of pat

Pseudomonas chlororaphis IRHB3 suppresses Soybean mosaic virus by coordinating photosynthetic protection, ROS homeostasis and hormone-mediated immunity.

BACKGROUND: Soybean mosaic virus (SMV) causes substantial yield losses of soybean worldwide. Although Pseudomonas chlororaphis IRHB3 is known to promote soybean growth and suppress fungal diseases, however, its efficacy against SMV has remained unclear. This study evaluated IRHB3 impacts on SMV infection and explored the underlying physiological and molecular mechanism. RESULTS: Foliar application of IRHB3 significantly reduced SMV disease index, with control efficacy of 52.69%, and decreased viral accumulation in inoculated and systemic leaves. IRHB3 also alleviated SMV-induced growth inhibi
