Platelet-activating factor (PAF) was found to induce histamine release from human basophils in mixed leukocytes in the presence of cytochalasin B. The reaction was rapid and dependent on temperature. The release was lower at a high concentration of PAF and was inhibited by EDTA, suggesting a noncytolytic mechanism. CV-3988, a PAF antagonist, inhibited the reaction dose-dependently. PAF acted as an IgE-independent stimulus, but PAF-induced histamine release from leukocytes of allergic asthmatics, who had elevated serum IgE levels, was significantly higher than that from leukocytes of controls.

Platelets induce generation of procoagulant tissue factor activity (TFa) by mononuclear leukocytes, and also enhance the TFa induced by endotoxin. Our present investigation demonstrated that arachidonic acid, which by itself had no effect on mononuclear TFa, greatly enhanced platelet-induced TFa. BACKGROUND: Platelet-activating factor (PAF) seems to be implicated in systemic lupus erythematosus (SLE) patients with associated renal diseases.
AIMS: In this study, we ensured the role of PAF in SLE patients without renal complications.
METHODS: Blood PAF and acetylhydrolase activity, plasma soluble phospholipase A(2), and the presence of antibodies against PAF were investigated in 17 SLE patients without active nephritis and in 17 healthy controls.
