Neurosteroid prolongs GABAA channel deactivation by altering kinetics of desensitized states.

Fast applications of GABA (1 mM) to nucleated and outside-out patches excised from granule neurons in cerebellar slices from developing rats evoked currents with a double exponential time course reminiscent of that of IPSCs. A neurosteroid 3alpha, 21dihydroxy-5alpha-pregnan-20-one (THDOC) remarkably increased the slow deactivation time constant and slowed down recovery from desensitization, as estimated by paired-pulse GABA applications. THDOC also reduced the amplitude of GABA currents, whereas it failed to affect the fast deactivation component and its relative contribution to peak amplitude

Progestins can have a membrane-mediated action in rat midbrain for facilitation of sexual receptivity.

In rats, progesterone (P) facilitates sexual receptivity by interacting with intracellular progestin receptors in the ventromedial hypothalamus (VMH). This experiment concerns whether P can also facilitate receptivity in rats by acting extragenomically within the ventral tegmental area (VTA). Ovariectomized rats (n = 10) with bilateral guide cannulas over the VMH and VTA were primed with 2 microg subcutaneous estradiol benzoate 44 hr prior to testing. After a pretest for sexual receptivity, animals received implants to the VMH of P, P conjugated to bovine serum albumin (P:BSA), or cholesterol

A new highly sensitive procedure--a prerequisite to evaluate the density of GABAA receptors and to study their allosteric properties in distinct areas of rat brain.

A new procedure for tissue preparation is described for the study of modulatory effects of neuroactive steroids and barbiturates on the in vitro binding of [3H]muscimol in distinct areas of rat brain. This method had been optimized for the highest amount of [3H]muscimol binding and a maximal effect of pentobarbital on radioligand binding. The following steps like tissue freezing, initial homogenization in alkaline triethanolamine buffer (pH 9.0), fractionated centrifugations and washings of the resulting membrane fractions in disodium piperazine-N-N'-bis[2-hydroxypropanesulfonic acid] buffer (

Subcellular distribution and properties of progesterone (delta4-steroid) 5alpha-reductase in rat medial basal hypothalamus.

The subcellular distribution and properties of rat hypothalamic progesterone 5 alpha-reductase, which accelerates the conversion of progesterone to 5 alpha-pregnane-3,20-dione, have been investigated by utilizing 3H-labeled substrate and a reverse isotopic dilution assay system. The enxymic activity was associated primarily with a cell debris-membranes fraction deribed from the 100 x g pellet. This fraction contained mainly membrane-like particulates and was free of nuclei. Little or no activity was associated with the purified nuclei. The hypothalamic 5 alpha-reductase was stimulated by NADPH

Indole-acetaldehyde from Rothia mucilaginosa activates the PXR/NRF2 axis to enhance alveolar macrophage phagocytosis and protect against ARDS.

BACKGROUND: Despite advances in therapeutic strategies, acute respiratory distress syndrome (ARDS) mortality remains high. Growing evidence links respiratory microbiome composition to ARDS outcomes. This investigation sought to elucidate how colonizing bacteria and their metabolites influence ARDS pathogenesis. METHODS: Bronchoalveolar lavage fluid (BALF) from patients with pulmonary infections was analyzed by metagenomic next-generation sequencing (mNGS) to identify characteristic bacteria. Bacterial culture supernatants were analyzed by untargeted metabolomics (LC-MS) to identify metabolites

Mode of action analysis for mouse liver tumor formation by MGK-264 and human relevance.

MGK-264, an insecticidal synergist, has been shown to increase the incidence of liver tumors in male and female mice. As MGK-264 is not a genotoxic compound, a series of investigative studies were conducted to elucidate the mode of action (MoA) for liver tumor production by MGK-264. Male and female CD-1 mice were given diets containing MGK-264 at 400, 3000, and 6000 ppm for 7 and/or 14 days. Treatment with MGK-264 caused activation of the constitutive androstane receptor (CAR), which resulted in liver hypertrophy, induction of cytochrome P450 CYP2B subfamily enzymes, and increased replicative

Human PXR Signaling Exacerbates Ethanol-induced Liver Injury in Females: Evidence From Mouse Models and Human Alcohol-associated Liver Disease Cohorts.

BACKGROUND & AIMS: Alcohol-associated liver disease (ALD), resulting from excessive alcohol consumption, manifests with greater severity in females. The pregnane X receptor (PXR), a xenobiotic nuclear receptor critical for toxin defense, has been implicated in ALD; however, the mechanisms underlying its role, particularly in sex differences, remain unclear. Due to species-specific ligand differences, PXR-humanized (hPXR) mouse models are essential for investigating PXR's contribution to sexual dimorphism in human ALD. METHODS: Male and female C57BL/6N Tac wild-type (WT) and hPXR mice were sub

Unveiling hepatic Krüppel-like factor 15 as the key regulator of cyclosporine A metabolism and adverse effects.

Cyclosporine A (CsA) is a widely used immunosuppressant for posttransplantation and autoimmune diseases, but its clinical application is limited by severe hepatorenal toxicity. Krüppel-like factor 15 (KLF15), a key regulator of xenobiotic metabolism, has been shown to modulate the metabolism of acetaminophen and rifampicin and play a role in the associated drug-induced liver toxicity. However, its role in CsA metabolism and CsA-induced hepatorenal injury remains unclear. This study aimed to investigate whether hepatic KLF15 regulates CsA metabolism and serves as a potential therapeutic target

The Three-Component Synthesis of 4-Sulfonyl-1,2,3-triazoles via a Sequential Aerobic Copper-Catalyzed Sulfonylation and Dimroth Cyclization.

4-Sulfonyl-1,2,3-triazole scaffolds possess promising bioactivities and applications as anion binders. However, these structures remain relatively unexplored and efficient synthetic procedures for their synthesis remain desirable. A practical room-temperature, aerobic copper-catalyzed three-component reaction of aromatic ketones, sodium sulfinates, and azides is reported. This procedure allows for facile access to 4-sulfonyl-1,5-disubstituted-1,2,3-triazoles in yields ranging from 34 to 89%. The reaction proceeds via a sequential aerobic copper(II)chloride-catalyzed oxidative sulfonylation and

CSA-90 Promotes Bone Formation and Mitigates Methicillin-resistant Staphylococcus aureus Infection in a Rat Open Fracture Model.

BACKGROUND: Infection of open fractures remains a significant cause of morbidity and mortality to patients worldwide. Early administration of prophylactic antibiotics is known to improve outcomes; however, increasing concern regarding antimicrobial resistance makes finding new compounds for use in such cases a pressing area for further research. CSA-90, a synthetic peptidomimetic compound, has previously demonstrated promising antimicrobial action against Staphylococcus aureus in rat open fractures. However, its efficacy against antibiotic-resistant microorganisms, its potential as a therapeut

Natural products for depression: Preclinical insights into microbiota-gut-brain axis modulation and pharmacological potential.

The microbiota-gut-brain axis (MGBA) is progressively associated with the pathophysiology of depression. This analysis consolidates existing preclinical findings on MGBA mechanisms associated with depression and assesses natural-product therapies through three key metabolite-related pathways: bile acid-responsive signaling via the farnesoid X receptor, G protein-coupled bile acid receptor 1, vitamin D receptor, and pregnane X receptor; metabolites from the kynurenine pathway and their connection to N-methyl-D-aspartate receptor signaling; and microbial indole-aryl hydrocarbon receptor signalin

High-Fat Diet Attenuates the Benefits of Increasing Phospholipid-to-Cholesterol Ratio on Lipid Metabolism in Mice: Evidence From Egg Yolk Low- and High-Density Lipoprotein Intake.

Our previous study suggested that increasing the phospholipid-to-cholesterol intake ratio might mitigate the negative effects of cholesterol intake, based on a comparison of lipid metabolism changes in mice fed chow diets containing egg yolk low-density lipoprotein (LDL) and high-density lipoprotein (HDL). This benefit might stem from the involvement of phospholipids, leading to reduced food efficiency, decreased body weight, and increased bile acid excretion. In this study, we further investigated the effects of this ratio on lipid metabolism under a high-fat diet (HFD) using egg yolk LDL and

AFB1 consolidates HBV harm to induce liver injury and carcinogenic risk by inactivating FTCD-AS1-PXR-MASP1 axis.

Aflatoxin B1 (AFB1) has been reported to synergize with hepatitis B virus (HBV) to induce development of hepatocellular carcinoma (HCC). Precise daily exposure to AFB1 and its contribution to liver injury have not been quantified and have even been disregarded due to lack of convenient detection, and the strong species specificity of HBV infection has restricted research on their synergistic harm. Hence, our objective was to investigate the molecular mechanisms by which AFB1 exacerbates HBV-related injury. We constructed tree shrew models with 400 μg HBV plasmid and 4 mg/kg AFB1 co-exposure fo
