The N-terminal region of HASPIN regulates phosphorylation of AURKA and meiotic progression in spermatocytes.

Protein phosphorylation is an important post-translational modification that plays a critical regulatory role in meiosis. HASPIN, a kinase highly conserved from yeast to mammals, is required for male fertility. In this study, we found that the intrinsically disordered N-terminal domain of HASPIN is also required for this function. Mice with deletion of N-terminal amino acids (aa) 1-243 of HASPIN exhibited reduced testicular size, sperm count, and fertility. Using immunoprecipitation-mass spectrometry and phosphoproteomics analysis, we found that HASPIN could interact with AURKA and regulate it

Lysine acetylation in the spargana of Spirometra mansoni: Insights into glycolysis and EF-hand domain proteins.

Protein post-translational modifications (PTMs) refer to the covalent attachment of chemical groups to polypeptide chains after protein synthesis, which expands protein chemical diversity and enhances functional versatility. Common PTM types include phosphorylation, glycosylation, and lysine acetylation (Kac), a major, reversible PTM regulated by lysine acetyltransferases (KATs) and lysine deacetylases (KDACs). Kac cross-regulates with other modifications to govern chromatin transcription and epigenetic programs, and occurs in histones, nucleoproteins, and cytoplasmic regulatory factors. In th

The multifaceted roles of the transcription factor SP1 in the pathogenesis of Alzheimer's disease: From molecular regulation to therapeutic targets.

UNLABELLED: Alzheimer's disease (AD) is driven by interrelated pathologies, including the accumulation of amyloid β (Aβ), tau pathology, chronic neuroinflammation, and oxidative stress (OS). These pathological processes collectively lead to progressive neurodegeneration. The transcription factor specificity protein 1 (SP1) functions as a key transcriptional regulator in AD pathogenesis. By binding to GC-box elements, SP1 promotes Aβ production and tau hyperphosphorylation. It also activates microglia, perpetuating neuroinflammation; disrupts oxidative balance; and ultimately induces neuronal d

Potential Role of SUMO and SUMOylation in the Pathogenesis of Diabetes Mellitus.

Diabetes mellitus is a group of metabolic disorders characterized by hyperglycemia and associated with multiple organ systems complications. The incidence and prevalence of diabetes are increasing in an epidemic proportion worldwide. In addition to environmental factors, some epigenetic and post-translational modifications have critical roles in the pathogenesis of diabetes and its complications. Reversible covalent modification such as SUMOylation by SUMO (Small Ubiquitin-like Modifier) has emerged as a new mechanism that affects the dynamic regulation of proteins. In this review, we initiall

SUMOylation in Skeletal Development, Homeostasis, and Disease.

The modification of proteins by small ubiquitin-related modifier (SUMO) molecules, SUMOylation, is a key post-translational modification involved in a variety of biological processes, such as chromosome organization, DNA replication and repair, transcription, nuclear transport, and cell signaling transduction. In recent years, emerging evidence has shown that SUMOylation regulates the development and homeostasis of the skeletal system, with its dysregulation causing skeletal diseases, suggesting that SUMOylation pathways may serve as a promising therapeutic target. In this review, we summarize

UBL3 Modification Reduces Nuclear POLE Levels to Potentiate Antitumor Immunity in Aero-upper Digestive Squamous Cell Carcinomas.

While immune checkpoint inhibitors (ICIs) have revolutionized treatment of the patients with aero-upper digestive squamous cell carcinomas (SCCs), including esophageal squamous cell carcinoma (ESCC) and lung squamous cell carcinoma (LUSC), only a subset of patients benefit from ICIs, therefore entailing exploration of the mechanisms to regulate treatment responsiveness. POLE, as a subunit of DNA polymerase ε, possesses 3'-5' proofreading activity, and its mutations have been associated with favourable response to ICIs in several solid tumors. However, post-translational modification (PTM) of P

Human γS-Crystallin Resists Unfolding Despite Extensive Chemical Modification from Exposure to Ionizing Radiation.

Ionizing radiation has dramatic effects on living organisms, causing damage to proteins, DNA, and other cellular components. γ radiation produces reactive oxygen species (ROS) that damage biological macromolecules. Protein modification due to interactions with hydroxyl radical is one of the most common deleterious effects of radiation. The human eye lens is particularly vulnerable to the effects of ionizing radiation, as it is metabolically inactive and its proteins are not recycled after early development. Therefore, radiation damage accumulates and eventually can lead to cataract formation.

RNA modifications in cancer.

RNA modifications are emerging as critical cancer regulators that influence tumorigenesis and progression. Key modifications, such as N6-methyladenosine (m6A) and 5-methylcytosine (m5C), are implicated in various cellular processes. These modifications are regulated by proteins that write, erase, and read RNA and modulate RNA stability, splicing, translation, and degradation. Recent studies have highlighted their roles in metabolic reprogramming, signaling pathways, and cell cycle control, which are essential for tumor proliferation and survival. Despite these scientific advances, the precise

N6-Methyladenosine RNA Modification in Normal and Malignant Hematopoiesis.

Over 170 nucleotide variants have been discovered in messenger RNAs (mRNAs) and non-coding RNAs so far. However, only a few of them, including N6-methyladenosine (m6A), 5-methylcytidine (m5C), and N1-methyladenosine (m1A), could be mapped in the transcriptome. These RNA modifications appear to be dynamically regulated, with writer, eraser, and reader proteins being identified for each modification. As a result, there is a growing interest in studying their biological impacts on normal bioprocesses and tumorigenesis over the past few years. As the most abundant internal modification in eukaryot

The role and mechanism of post-translational modifications (PTMs) in immune-related eye diseases.

Post-translational modifications (PTMs) encompass the spectrum of chemical covalent alterations that proteins undergo after being synthesized from mRNA. These modifications typically involve the covalent bonding of chemical groups or small protein molecules to the amino acid backbones or side chains. Currently, over 650 types of PTMs have been identified, including significant ones such as phosphorylation, ubiquitination, SUMOylation, methylation, acetylation, glycosylation, among other novel varieties like lactylation. However, a systematic review of PTMs associated with immune-mediated ocula

Immune recognition of citrullinated epitopes.

Conversion of arginine into citrulline is a post-translational modification that is observed in normal physiological processes. However, abnormal citrullination can provoke autoimmunity by generating altered self-epitopes that are specifically targeted by autoantibodies and T cells. In this review we discuss the recognition of citrullinated antigens in human autoimmune diseases and the role that this modification plays in increasing antigenic diversity and circumventing tolerance mechanisms. Early published work demonstrated that citrullinated proteins are specifically targeted by autoantibodi

Hydrogen Sulfide Signaling in Plants: Emerging Roles of Protein Persulfidation.

Hydrogen sulfide (H2S) has been largely referred as a toxic gas and environmental hazard, but recent years, it has emerged as an important gas-signaling molecule with effects on multiple physiological processes in both animal and plant systems. The regulatory functions of H2S in plants are involved in important processes such as the modulation of defense responses, plant growth and development, and the regulation of senescence and maturation. The main signaling pathway involving sulfide has been proven to be through protein persulfidation (alternatively called S-sulfhydration), in which the th

Nitration in cancer signaling.

Oxidative stress arises from an imbalance between the production of reactive oxygen and nitrogen species (ROS and RNS, respectively) and the capacity of cellular antioxidant defenses to neutralize them. In cancer, this imbalance drives pathological remodeling of signaling networks that promote tumor initiation and progression. Among RNS, nitric oxide (·NO) and its highly reactive derivative peroxynitrite (ONOO-) are central mediators of redox dysregulation within the tumor microenvironment. These species induce site-specific post-translational modifications (PTMs), most notably protein tyrosin

Post-translational modifications in sepsis-induced myocardial dysfunction - Molecular mechanisms and therapeutic targets: A review.

Sepsis-induced myocardial dysfunction (SIMD) is a frequent and life-threatening complication of sepsis driven by interconnected inflammatory, oxidative, metabolic, and mitochondrial disturbances. Post-translational modifications (PTMs) have emerged as key regulators of these processes by dynamically altering protein activity, stability, localization, and signaling. This narrative review aimed to summarize the roles of major PTMs in SIMD and evaluate their potential as therapeutic targets. Relevant original studies and authoritative reviews published predominantly within the past decade were id

ALKBH8 contributes to neurological function through oxidative stress regulation.

Transfer RNA (tRNA) modification is essential for proper protein translation, as these modifications play important roles in several biological functions and disease pathophysiologies. AlkB homolog 8 (ALKBH8) is one of the nine mammalian ALKBH family molecules known to regulate selenoprotein translation through the modification of the wobble uridine (U34) in tRNA; however, its specific biological roles remain unclear. In this study, we investigated the role of ALKBH8 using Alkbh8-knockout (Albkh8-/-) mice, which were observed to have reduced 5-methoxycarbonylmethyluridine (mcm5U) and (S)-5-met

PRMT1-Mediated LDHA Methylation Drives STAT3 Lactylation to Orchestrate Intestinal Inflammation and Tumorigenesis.

Signal transducer and activator of transcription 3 (STAT3) activation is crucial in intestinal inflammation and tumorigenesis. However, its metabolic regulation is not well understood. Herein, we identified a macrophage-dependent methionine-S-adenosylmethionine (SAM)-protein arginine methyltransferase 1 (PRMT1)-lactate dehydrogenase A (LDHA)-lactate axis that controls intestinal inflammation through STAT3 regulation. Specifically, SAM promoted STAT3 Y705 phosphorylation and upregulated anti-inflammatory interleukin-10 expression in macrophages. Additionally, genetic ablation of PRMT1 in myeloi

5-mC RNA methylation: a key regulator in cancer development and treatment.

BACKGROUND: 5-methylcytosine (5-mC) RNA methylation is a prevalent post-transcriptional modification affecting both coding and non-coding RNAs. 5-mC methylation can regulate the expression level of relevant genes and biological processes by affecting RNA stability, translation efficiency, spatial structure, protein interactions, and other processes. Emerging evidence highlights its critical role in cancer biology. MAIN BODY: Abnormal changes in 5-mC methylation lead to aberrant expression of oncogenes or inactivation of suppressor genes, which in turn affects the growth and differentiation of

O-GlcNAcylation: A bridge for regulating the function of the "heart-kidney-bone axis".

The three major organ systems of heart, kidney and bone are highly correlated in physiology and pathology. They form the "heart-kidney-bone axis" together. The typical clinical manifestations of this axis include the deterioration of heart and kidney function, reflected by cardiorenal syndrome, renal bone disease, and cardiovascular calcification contained in chronic kidney disease-mineral bone disorder (CKD-MBD). However, the core molecular mechanism that regulates this axis has not been fully understood. O-linked N-acetylglucosamine (O-GlcNAc) glycosylation modification is a dynamic, reversi
