A "multi-omics" analysis of blood-brain barrier and synaptic dysfunction in APOE4 mice.

Apolipoprotein E4 (APOE4), the main susceptibility gene for Alzheimer's disease, leads to blood-brain barrier (BBB) breakdown in humans and mice. Remarkably, BBB dysfunction predicts cognitive decline and precedes synaptic deficits in APOE4 human carriers. How APOE4 affects BBB and synaptic function at a molecular level, however, remains elusive. Using single-nucleus RNA-sequencing and phosphoproteome and proteome analysis, we show that APOE4 compared with APOE3 leads to an early disruption of the BBB transcriptome in 2-3-mo-old APOE4 knock-in mice, followed by dysregulation in protein signali

Extracellular Vesicles in Amyotrophic Lateral Sclerosis.

Amyotrophic Lateral Sclerosis is a progressive neurodegenerative disease and is the most common adult motor neuron disease. The disease pathogenesis is complex with the perturbation of multiple pathways proposed, including mitochondrial dysfunction, RNA processing, glutamate excitotoxicity, endoplasmic reticulum stress, protein homeostasis and endosomal transport/extracellular vesicle (EV) secretion. EVs are nanoscopic membrane-bound particles that are released from cells, involved in the intercellular communication of proteins, lipids and genetic material, and there is increasing evidence of

Fused in sarcoma (FUS) interacts with the cytolinker protein plectin: implications for FUS subcellular localization and function.

Fused in sarcoma (FUS) is a multifunctional protein involved in transcriptional control, pre-mRNA processing, RNA transport and translation. The domain structure of FUS reflects its functions in gene regulation and its ability to interact with other proteins, RNA and DNA. By use of a recombinant fragment of FUS in pull-down experiments followed by mass spectrometry analysis we have identified a novel interaction between the FUS N-terminal and the cytolinker plectin. An in situ proximity ligation assay confirmed that FUS-plectin interactions take place in the cytoplasm of cells. Furthermore, pl…

Inhibition of uridine transport in cultured mammalian cells by theophylline.

Theophylline (theobromine, caffeine) reversibly inhibits the incorporation of labeled RNA precursors both in confluent 37 RC and in exponentially growing HeLa cells. As measured in 37 RC after 2 h labeling, 20 mM theophylline reduces the incorporation of [3H]UTP and [14C]uridine into acid-precipitable material to 5% and 9% of the control, respectively. This reduction is paralleled by a comparably lowered incorporation of the same precursors into the acid-soluble pool. The initial rate of incorporation into total cell material is similarly affected by theophylline, the inhibition being of a sim

Advances of exosomes-based applications in diagnostic biomarkers for dental disease and dental regeneration.

Exosomes are produced by all the cells and exist in all body fluids. They have been regarded as potentially promising to diagnostic biomarkers and therapeutic bioactive mediators since they transport DNA, RNA and protein information from cell to cell. Herein, we summarized the recent research about exosomes from gingival crevicular fluid, saliva and serum used as diagnostic markers in periodontitis and dental caries. Moreover, we highlighted the mechanisms of exosomes in dental pulp regeneration and periodontal regeneration, as well as the technological innovation of exosome delivery methods i

Genetics of motor neuron disorders: new insights into pathogenic mechanisms.

The past few years have seen the identification of dozens of genes with causal roles in motor neuron diseases (MNDs), particularly for amyotrophic lateral sclerosis and hereditary spastic paraplegia. Although many additional MND genes remain to be identified, the accumulated genetic evidence has already provided new insights into MND pathogenesis, which adds to the well-established involvement of superoxide dismutase 1 (SOD1) mutations. The pathways that have been recently implicated include those that affect RNA processing, axonal transport and mitochondrial function. The functional classes o

Emerging role of plant-derived extracellular vesicles in human health and disease.

Plant-derived extracellular vesicles (PDEVs) are emerging plant-derived vesicle preparations that have attracted considerable interest as potential natural nanocarriers for therapeutic delivery owing to their ability to transport bioactive lipids, proteins, nucleic acids, and plant-derived metabolites. Increasing evidence suggests that specific PDEV preparations can modulate intercellular communication, influence immune responses, and facilitate the delivery of bioactive cargo in specific preclinical models. Recent studies have demonstrated their potential applications in inflammatory bowel di

Gene Expression Analysis of Laser-Captured Purkinje Cells in the Essential Tremor Cerebellum.

Essential tremor (ET) is a common, progressive neurological disease characterized by an 8-12-Hz kinetic tremor. Despite its high prevalence, the patho-mechanisms of tremor in ET are not fully known. Through comprehensive studies in postmortem brains, we identified major morphological changes in the ET cerebellum that reflect cellular damage in Purkinje cells (PCs), suggesting that PC damage is central to ET pathogenesis. We previously performed a transcriptome analysis in ET cerebellar cortex, identifying candidate genes and several dysregulated pathways. To directly target PCs, we purified RN

Gene expression and viral prodution in latently infected, resting CD4+ T cells in viremic versus aviremic HIV-infected individuals.

The presence of HIV-1 in latently infected, resting CD4(+) T cells has been clearly demonstrated in infected individuals; however, the extent of viral expression and the underlying mechanisms of the persistence of HIV-1 in this viral reservoir have not been fully delineated. Here, we show that resting CD4(+) T cells from the majority of viremic patients are capable of producing cell-free HIV-1 spontaneously ex vivo. The levels of HIV-1 released by resting CD4(+) T cells were not significantly reduced in the presence of inhibitors of cellular proliferation and viral replication. However, restin

Optical Control of Therapeutic Synthetic Cells: Controlling Biomolecule Synthesis, Activation, and Communication.

Synthetic cells are emerging as promising platforms that combine cell-free biomolecule synthesis with the programmability of bottom-up engineered compartments. By decoupling biomolecule production from cellular growth and evolutionary constraints, synthetic cells enable the controlled generation of proteins, nucleic acids, peptides, and signaling molecules within defined artificial environments. However, many envisioned biomedical applications require precise regulation over biomolecule synthesis, activation, transport, release, and communication. In this Review, we examine how light can provi

Promoter Hypermethylation-Induced Silencing of FXYD1 Drives Breast Cancer Metastasis via DDX5-Mediated Wnt/β-Catenin Pathway Activation.

Epigenetic silencing of tumor suppressor genes is a hallmark of cancer progression. The FXYD (FXYD domain-containing ion transport regulator) family, classically known for ion transport regulation, has recently been implicated in oncogenesis; however, the role of its founding member, FXYD1, in breast cancer remains unclear. Here, FXYD1 is identified as a significantly downregulated gene in breast cancer tissues, and low FXYD1 expression is associated with unfavorable patient prognosis. Integrative transcriptomic, clinical, and epigenetic analyses revealed that promoter hypermethylation drives

Phosphorylation dynamics of sodium-independent organic anion transporter SLCO4A1 and its role in diseases: a review.

SLCO4A1, a membrane-localized organic anion transporter belonging to the solute carrier (SLC) family, mediates the sodium-independent transport of endogenous substrates, including steroid hormones, prostaglandins, and thyroid hormones. Despite increasing evidence supporting its physiological and pathological significance, a comprehensive review focusing on its phosphorylation landscape and phosphoregulation is currently lacking. Large-scale phosphoproteomic analyses have identified three predominant phosphorylation sites, T37, S40, and S43, within its N-terminal intrinsically disordered region

UVB enhances SLC6A15-mediated phenylalanine transport to promote melanogenesis.

BACKGROUND: Abnormal melanogenesis is a fundamental biological process underlying pigmentary disorders. While several solute carrier (SLC) transporters have been implicated in its regulation, the functions of many SLCs members remain poorly characterized. METHODS: We delineated transcriptional features of melanocytes with high melanogenic activity using single-cell and bulk RNA sequencing. Key SLC genes associated with melanogenesis were identified and prioritized using machine learning and Mendelian randomization analyses, followed by in vitro functional validation. RESULTS: Integrated mult

Transcutaneous Immunotherapy for RNAi: A Cascade-Responsive Decomposable Nanocomplex Based on Polyphenol-Mediated Framework Nucleic Acid in Psoriasis.

Skin is the first barrier against external threats, and skin immune dysfunction leads to multiple diseases. Psoriasis is an inflammatory, chronic, common, immune-related skin disease that affects more than 125 million people worldwide. RNA interference (RNAi) therapy is superior to traditional therapies, but rapid degradation and poor cell uptake are the greatest obstacles to its clinical transformation. The transdermal delivery of siRNA and controllable assembly/disassembly of nanodrug delivery systems can maximize the therapeutic effect. Tetrahedral framework nucleic acid (tFNA) is undoubted

Rab26 promotes macrophage phagocytosis through regulation of MFN2 trafficking to mitochondria.

Acute respiratory distress syndrome (ARDS) is an inflammatory disorder of the lungs caused by bacterial or viral infection. Timely phagocytosis and clearance of pathogens by macrophages are important in controlling inflammation and alleviating ARDS. However, the precise mechanism of macrophage phagocytosis remains to be explored. Here, we show that the expression of Rab26 is increased in Escherichia coli- or Pseudomonas aeruginosa-stimulated bone marrow-derived macrophages. Knocking out Rab26 reduced phagocytosis and bacterial clearance by macrophages. Rab26 interacts with mitochondrial fusion

Studies of the Efficacy of Low-Dose Apatinib Monotherapy as Third-Line Treatment in Patients with Metastatic Colorectal Cancer and Apatinib's Novel Anticancer Effect by Inhibiting Tumor-Derived Exosome Secretion.

Antiangiogenic therapy is an important treatment strategy for metastatic colorectal cancer (mCRC). We carried out a clinical study of low-dose apatinib (250 mg) monotherapy as a third-line treatment in patients with mCRC and assessed its efficacy and safety. It demonstrated that low-dose apatinib had comparable survival outcomes, significantly improved the patient quality of life, and caused tolerable adverse reactions. To further investigate the underlying mechanism of the effects of apatinib in CRC besides angiogenesis, we performed RNA-seq, and our results suggested that apatinib may have o
