Onion skin extract as a protective agent against oxidative stress in Saccharomyces cerevisiae induced by cadmium.

Onion skin extract is a source of bioactive compounds demonstrating a high antioxidant activity. The purpose of this research was to investigate the effect of onion skin extract at the level of oxidative stress markers in Saccharomyces cerevisiae exposed to cadmium. The generation of reactive oxygen species (ROS), antioxidant enzymes activity, total content of glutathione, and the level of lipid peroxidation in yeast cells were determined. Research showed that onion skin extract protected the yeast cells from the occurrence of oxidative stress induced by cadmium. Cells treated with onion skin …

Producing alcohol and salt stress tolerant strain of Saccharomyces cerevisiae by heterologous expression of pprI gene.

INTRODUCTION: Ethanol is considered a comparatively clean biofuel, and its large scale production has been a long time concern. Saccharomyces cerevisiae has proven to be the suitable microorganism for large scale ethanol production, but production of other alcohols like butanol and using lignocellulosic substrates is restricted due to lacking tolerance toward toxicity of alcohols, and compounds released from substrates. This study aimed to produce a tolerant strain by using pprI gene of Deinococcus radiodurans. MATERIAL AND METHOD: pprI gene was introduced into Saccharomyces cerevisiae. To ev…

Overexpression of PDR16 confers resistance to complex sphingolipid biosynthesis inhibitor aureobasidin A in yeast Saccharomyces cerevisiae.

Sphingolipids are essential for normal cell growth of yeast Saccharomyces cerevisiae. Aureobasidin A (AbA), an antifungal drug, inhibits Aur1, an enzyme catalyzing the synthesis of inositol phosphorylceramide, and induces a strong growth defect in yeast. In this study, we screened for multicopy suppressor genes that confer resistance to AbA, and identified PDR16. In addition, it was found that PDR17, a paralog of PDR16, also functions as a multicopy suppressor. Pdr16 and Pdr17 belong to a family of phosphatidylinositol transfer proteins; however, cells overexpressing the other members of the f…

A novel toxicity mechanism of CdSe nanoparticles to Saccharomyces cerevisiae: enhancement of vacuolar membrane permeabilization (VMP).

Cadmium selenide (CdSe) nanoparticles are implemented in a wide range of applications, but their potential risk to the ecosystem, especially to the organisms essential for the maintenance of ecosystem homeostasis, such as fungal populations, plants and bacteria, remains to be elucidated. In this study, we investigated their toxicity to one of the most important fungal model organisms, Saccharomyces cerevisiae. Growth inhibition assays revealed that the synthesized CdSe nanoparticles with the sizes of 20-30 nm had strong inhibitory effect on yeast growth (IC50=80 ppm). This toxicity was not att…

Clinical significance and prevalence of anti-Saccharomyces cerevisiae antibody in Chinese patients with primary biliary cirrhosis.

Clinical significance of anti-Saccharomyces cerevisiae antibody (ASCA) and its prevalence in Chinese primary biliary cirrhosis (PBC) patients have not been characterized and therefore needs to be defined. Enzyme-linked immunosorbent assay was used to test ASCA in sera from 198 PBC patients, 85 patients with other liver diseases (OLD) and 35 health controls (HC). Indirect immunofluorescence was used to detect anti-mitochondrial antibodies (AMA) in PBC. Results showed that the frequency of ASCA in PBC, 29.8 %, was higher than other disease groups. And ASCA occurred more frequently in PBC patient…

A comparative study of Saccharomyces cerevisiae sensitivity against eight yeast species sensitivities to a range of toxicants.

Fungi are widespread and very important in many ecosystems but the extensive use of pesticides can adversely affect beneficial fungi. The yeast, Saccharomyces cerevisiae has been proposed for the toxicological assessment of the effects of environmental pollutants on non-target fungi. To assess whether S. cerevisiae is a good representative of the immense ecological and phylogenetic diversity of yeasts, we compare the sensitivity of eight other yeasts from diverse phylogenetic taxa to a range of toxicants and environmental samples. Sensitivity was assessed using both the growth inhibition and a…

Apoptosis as a mechanism for removal of mutated cells of Saccharomyces cerevisiae: the role of Grx2 under cadmium exposure.

Cadmium is a strong mutagen that acts by inhibiting DNA mismatch repair, while its toxic effect seems to be related to an indirect oxidative stress that involves glutathione (GSH) mobilization. Among the roles of GSH is the protection of proteins against oxidative damage, by forming reversible mixed disulfides with cysteine residues, a process known as protein glutathionylation and catalyzed by glutaredoxins (Grx). In this current study, Saccharomyces cerevisiae cells deficient in GRX2, growing in 80 muM CdSO(4), showed high mitochondrial mutagenic rate, determined by frequency of mutants that…

Synthesis of FUDP-N-acetylglucosamine and FUDP-glucose in Saccharomyces cerevisiae cells treated with 5-fluorouracil.

Saccharomyces cerevisiae cells (strain W303-1A) treated with 5-fluorouracil and grown in 2% (fermentative conditions) or in 0.1% glucose (oxidative conditions) accumulated two types of 5-fluoro-UDP-sugars (FUDP-sugars): FUDP-N-acetylglucosamine and FUDP-glucose. No difference was observed in both conditions of culture. The viability of yeast cells on treatment with 5-fluorouracil was also followed. Both FUDP-sugars were partially purified by column chromatography (on Hypersil ODS and Mono Q columns) and characterized by: (i) treatment with alkaline phosphatase (EC 3.1.3.1), snake venom phospho…

Mechanisms of copper loading on the Schizosaccharomyces pombe copper amine oxidase 1 expressed in Saccharomyces cerevisiae.

Copper amine oxidases (CAOs) are found in almost every living kingdom. Although Saccharomyces cerevisiae is one of the few yeast species that lacks an endogenous CAO, heterologous gene expression of CAOs from other organisms produces a functional enzyme. To begin to characterize their function and mechanisms of copper acquisition, two putative cao(+) genes from Schizosaccharomyces pombe were expressed in S. cerevisiae. Expression of spao1(+) resulted in the production of an active enzyme capable of catalysing the oxidative deamination of primary amines. On the other hand, expression of spao2(+…

Topological and functional study of subunit h of the F1Fo ATP synthase complex in yeast Saccharomyces cerevisiae.

Subunit h, a 92-residue-long, hydrophilic, acidic protein, is a component of the yeast mitochondrial F1Fo ATP synthase. This subunit, homologous to the mammalian factor F6, is essential for the correct assembly and/or functioning of this enzyme since yeast cells lacking it are not able to grow on nonfermentable carbon sources. Chemical cross-links between subunit h and subunit 4 have previously been shown, suggesting that subunit h is a component of the peripheral stalk of the F1Fo ATP synthase. The construction of cysteine-containing subunit h mutants and the use of bismaleimide reagents prov

Effect of UV radiation on thermotolerance, ethanol tolerance and osmotolerance of Saccharomyces cerevisiae VS1 and VS3 strains.

After a previous mass screening and enrichment programme for the isolation of thermotolerant yeasts, VS1, VS2, VS3 and VS4 strains isolated from soil samples, collected within the hot regions of Kothagudem Thermal Power Plant, AP, India, had a better thermotolerance, osmotolerance and ethanol tolerance than the other isolates. Among these isolates VS1 and VS3 were best performers. Efforts were made to further improve their osmotolerance, thermotolerance and ethanol tolerance by treating them with UV radiation. Mutants of VS1 and VS3 produced more biomass and ethanol than the parent strains at

Identification of SLF1 as a new copper homeostasis gene involved in copper sulfide mineralization in Saccharomyces cerevisiae.

In Saccharomyces cerevisiae, at least 12 genes are important for cells to propagate in medium containing elevated concentrations of copper salts (J. Welch, S. Fogel, C. Buchman, and M. Karin, EMBO J. 8:255-260, 1989). Complementation studies were carried out on a copper-sensitive mutation (cup14) from this group. A new yeast gene, designated SLF1, was identified as a multicopy suppressor of the cup14 mutation. Slf1 is important for the physiological process of copper sulfide (CuS) mineralization on the surface of cells cultured in medium containing copper salts. CuS mineralization causes the c…

Sequence determination of human papillomavirus type 6a and assembly of virus-like particles in Saccharomyces cerevisiae.

Human papillomavirus 6a (HPV6a), the most abundant HPV6 subtype, was detected in a vulvar condyloma acuminatum. The complete genome of HPV6a was cloned, and its DNA sequence was shown to be over 97% identical to the HPV6b sequence. Of the eight open reading frames (ORFs) of HPV6a, only the imputed amino acid sequence of the major capsid protein L1 was identical to the corresponding HPV6b sequence; all other HPV6a ORFs showed amino acid changes compared to the HPV6b ORFs. The HPV6a L1 or the L1 + L2 ORFs were expressed in the yeast Saccharomyces cerevisiae. Self-assembly of the L1 capsid protei…

A delay in the Saccharomyces cerevisiae cell cycle that is induced by a dicentric chromosome and dependent upon mitotic checkpoints.

Dicentric chromosomes are genetically unstable and depress the rate of cell division in Saccharomyces cerevisiae. We have characterized the effects of a conditionally dicentric chromosome on the cell division cycle by using microscopy, flow cytometry, and an assay for histone H1 kinase activity. Activating the dicentric chromosome induced a delay in the cell cycle after DNA replication and before anaphase. The delay occurred in the absence of RAD9, a gene required to arrest cell division in response to DNA damage. The rate of dicentric chromosome loss, however, was elevated in the rad9 mutant.…

[Effects of Saccharomyces boulardii yeast on trophozoites of Entamoeba histolytica in vitro and in cecal amebiasis in young rats].

Lyophilized and rehydrated Saccharomyces boulardii yeasts were administered to young rats previously inoculated into the cecum with Entamoeba histolytica. The numbers of diseased young rats and the severity of the infection, assessed from the appearance of the cecum and its content of mucus and amebae, were significantly reduced by the treatment. The lesions observed resembled those seen in the untreated controls, although healing was faster in the treated animals. Saccharomyces boulardii had no intrinsic amebicidal action in vitro.

Functional complementation of catalase-defective peroxisomes in a methylotrophic yeast by import of the catalase A from Saccharomyces cerevisiae.

A mutant of the methanol-utilizing yeast Hansenula polymorpha defective in catalase was isolated. It lacks the ability to grow on methanol as the sole source of carbon and energy due to a loss of peroxisomal function that is required for the dissimilation and assimilation of this substrate. Growth of the mutant on glucose or glycerol was not impaired. Transformation of mutant cells with the gene coding for catalase A from Saccharomyces cerevisiae [Cohen, G., Fessl, F., Traczyk, J., Rytka, J. & Ruis, H. (1985) Mol. Gen. Genet. 200, 74-79] conferred constitutive expression of catalase activity. …
