Antioxidant, Anti-Cholinesterase, and Neuroprotective Properties of Morus alba and Morus nigra Extracts.

The Morus genus comprises several tree species whose fruits are used in human nutrition, while the leaves and roots are used in traditional … of galantamine, which was used as standard. Both extracts reversed, in a dose-dependent manner, the scopolamine-induced cognitive impairment and behavioural alterations in scopolamine-treated zebrafish (Danio rerio) as evaluated by the Y-maze test, novel tank diving test, and novel object recognition test.

ENALAPRIL EFFECT ON THE STATE OF NITROGEN OXIDE SYSTEM AND PROOXIDANT-ANTIOXIDANT BALANCE IN BRAIN UNDER CONDITIONS OF BLOCKADE OF CENTRAL CHOLINERGIC SYSTEM.

One of the most important medical-social issues today is the search for new pathogenic directions in pharmacological prevention and treatment of neurodegenerative diseases of the central nervous system. Objective: to investigate enalapril effect on the indices of the nitrogen oxide system and prooxidant-antioxidant balance in the cerebral cortex and hippocampus under conditions of experimental cerebral neurodegeneration. The state of the nitrogen oxide system and prooxidant-antioxidant balance is investigated in the experiment on male rats with scopolamine-induced neurodegeneration (1 mg/kg, 2…

Effect of purification followed by solubilization of receptor material on quantitative receptor assays for anticholinergic drugs.

In order to optimize quantitative receptor assays for anticholinergics, the different receptor preparations resulting from the purification and the solubilization of the P2 pellet from the calf striatum were evaluated. The dissociation constants for two chemically different anticholinergics, the tertiary amine scopolamine and the quaternary amine oxyphenonium, were calculated from inhibition studies of 3H-NMS binding in buffer and plasma. The Kd values for both anticholinergics were similar for all the membrane-bound receptor preparations (unpurified and the purified P2 pellet) either in buffe…

Molecular cloning of hyoscyamine 6 beta-hydroxylase, a 2-oxoglutarate-dependent dioxygenase, from cultured roots of Hyoscyamus niger.

Roots of several solanaceous plants produce anticholinergic alkaloids, hyoscyamine and scopolamine. Hyoscyamine 6 beta-hydroxylase, a 2-oxoglutarate-dependent dioxygenase (EC 1.14.11.11), catalyzes hydroxylation of hyoscyamine in the biosynthetic pathway leading to scopolamine. We report here on the isolation of cDNA clones encoding the hydroxylase from a cDNA library made from mRNA of the cultured roots of Hyoscyamus niger. The library was screened with three synthetic oligonucleotides that encode amino acid sequences of internal peptide fragments of the purified hydroxylase. Nucleotide seque…

The role of control activity levels in the reported strain differences to the behavioral effects of drugs in mice.

The effect of d-amphetamine (3-100 mumoles/kg), scopolamine (0.3-100 mumoles/kg) and morphine (3-1000 mumoles/kg) were studied on the spontaneous motor activity (SMA) of four strains of mice: CF-1, DBA/2, C57BL/6 and CD-1. All three drugs increased the SMA of the CF-1, C57BL/6, and CD-1 strains at low to moderate doses and decreased SMA at higher doses. In the DBA/2 strain, d-amphetamine and scopolamine increased SMA at low doses and decreased SMA at high doses; only decreases in SMA were observed with morphine. When the drug effect was expressed relative to control levels of SMA, large appare…

Chlordiazepoxide, an anxiolytic benzodiazepine, impairs place navigation in rats.

There are separate proposals that the hippocampus is involved in 'spatial memory' and in the control of 'anxiety'. Despite a larger number of common effects of anxiolytic drugs and hippocampal lesions, no effect of anxiolytic drugs has yet been reported in those spatial tasks which are particularly sensitive to the effects of hippocampal lesions. The present study addresses this issue. Separate groups of rats were treated, i.p., with 5 mg/kg chlordiazepoxide (an anxiolytic benzodiazepine), 1 mg/kg scopolamine (a muscarinic antagonist which has previously been shown to impair spatial learning) …

Motion sickness in cats: a symptom rating scale used in laboratory and flight tests.

The cat is proposed as a model for the study of motion and space sickness. Development of a scale for rating the motion sickness severity in the cat is described. The scale is used to evaluate an antimotion sickness drug, d-amphetamine plus scopolamine, and to determine whether it is possible to predict sickness susceptibility during parabolic flight, including zero-G maneuvers, from scores obtained during ground based trials.

Moreover, SV.CR treatment also reversed scopolamine-impaired cognition (P < 0.05) in various deployed memory and learning tasks. Biochemical studies of brain homogenates of SV.CR treated animals demonstrated decreased anticholinesterase activity and lipid peroxidation levels whereas increased levels …

The pharmacology of medieval sedatives: the "Great Rest" of the Antidotarium Nicolai.

ETHNOPHARMACOLOGICAL RELEVANCE: Past practices of compound drugs from different plant ingredients enjoyed remarkable longevity over centuries yet are largely dismissed by modern science as subtherapeutic, lethal or fanciful. AIM OF THE STUDY: To examine the phytochemical content of a popular medieval opiate drug called the "Great Rest" and gauge the bioavailability and combined effects of its alkaloid compounds (morphine, codeine, hyoscyamine, scopolamine) on the human body according to modern pharmacokinetic and pharmacodynamic parameters established for these compounds. CALCULATIONS AND TH…

Modelling Alzheimer-like cognitive deficits in rats using biperiden as putative cognition impairer.

To enable the development of effective treatments for dementias such as Alzheimer's disease (AD), it is important to establish valid animal models of cognitive impairments. Scopolamine is widely used to induce cognitive deficits in animal models of AD, but also causes non-cognitive side effects. We assessed whether biperiden, a selective antagonist of M1 muscarinic receptors, which are predominantly expressed in brain areas involved in cognitive processes, causes cognitive deficits without inducing peripheral side-effects. Two different doses of biperiden (3 or 10mgkg(-1)) on the acquisition o…

Studies on tropane alkaloid extraction by volatile organic solvents: dichloromethane vs. chloroform.

In order to investigate the production of tropane alkaloids by hairy roots of Atropa baetica, transgenic for the gene h6h encoding the enzyme hyoscyamine 6beta-hydroxylase, solvent extraction with chloroform and with dichloromethane of the metabolites present in the liquid medium and in the root tissue was compared. The extraction of scopolamine from the liquid medium was equally effective with either solvent, giving maximum values of around 850 microg/flask. For the roots, three different extraction methods were employed: A, employing chloroform:methanol: (25%) ammonia (15:5:1) for initial ex…

Behavioral effects of anti-muscarinic, anti-serotonergic, and anti-NMDA treatments: hippocampal and neocortical slow wave electrophysiology predict the effects on grooming in the rat.

Previous research has shown that hippocampal and neocortical activation accompanies the postural changes occurring during self-grooming in rats but is absent or reduced during the stereotyped components of grooming, including head-washing and licking or biting. Since electrocortical activation is dependent on ascending cholinergic and serotonergic projections, we hypothesized that central muscarinic and serotonergic blockade would disrupt grooming by degrading cerebral control of changes in posture. Consistent with this, we find that systemic injections of scopolamine: (a) markedly reduce the …

Drug effects on cognitive function in mice determined by the non-matching to sample task using a 4-arm maze.

A new task for the analysis of drug effects on the cognitive function in mice was investigated using a 4-arm maze with three selectable arms … nse errors remained unchanged in the well-trained mice. Without increasing the non-reward response, scopolamine and atropine, but not methylscopolamine, decreased the correct response in a delay-dependent manner at a low dose range, while diazepam did so in a delay-independent manner. Physostigmine ameliorated scopolamine-induced impairment in performance, bu …
