Operational 24-h PM2.5 forecasting using ensemble methods and open-access satellite data: application to Dakar, Senegal.

Accurate short-term air quality forecasting is urgently needed in African megacities. Chronic PM2.5 pollution threatens public health in these cities. At the same time, ground-based monitoring infrastructure remains sparse. This study develops and evaluates an operational 24-h PM2.5 forecasting system for the low-income housing (HLM) district of Dakar, Senegal. The site represents a high-exposure urban-industrial environment. Daily PM2.5 concentrations from 2019 to 2024 (mean = 274.71 µg/m3) were predicted using four ensemble machine learning (ML) models: Random Forest (RF), Extra Trees Regres…

Perceiving temporal rhythms in Sahelian soundscapes of Senegal: A pilot interdisciplinary study bridging psychoacoustics, ecology, and anthropologya).

At the intersection of ecological, psychoacoustic, and anthropological perspectives, this study examines how listeners living in Senegal's Sahelian region perceive seasonal and diurnal patterns in local soundscapes. Field recordings representing rainy and dry seasons, as well as different times of day, were first characterized using ecoacoustic analyses that revealed differences in spectro-temporal modulations. The recordings were then presented to two groups: Senegalese pastoralists (Peul herders) living in the Ferlo (n = 9) and a French control group with no prior exposure to the region (n =…

Emergence of Plasmodium falciparum pfdhps A581G mutation in Southern Senegal under Seasonal Malaria Chemoprevention pressure, 2020-2023.

BACKGROUND: Malaria prevention in Senegal includes the use of Seasonal Malaria Chemoprevention with sulfadoxine-pyrimethamine plus amodiaquine. Prolonged use of sulfadoxine-pyrimethamine, however, can select for mutations in the Plasmodium falciparum mutations in pfdhfr and pfdhps genes. METHODS: We analyse dried-blood-spot samples collected during therapeutic-efficacy studies in two SMC districts (Kolda, Kédougou) between 2020 and 2023. Using targeted amplicon deep sequencing we determine the prevalence of pfdhfr and pfdhps mutations by year and site. RESULTS: The pfdhfr triple mutant genot…

High Detection Frequency of Enteric Pathogens: Insight from Wastewater-Based Epidemiology (WBE) Surveillance Approach in Dakar, Senegal.

Despite the importance of wastewater environmental monitoring in disease prevention and response strategies, its use remains poorly documented in Senegal. In addition, there is more onsite sanitation than sewer networks in Dakar, and open drains channel for rainwater are also used as clandestine wastewater discharge into the sea. This study aimed to assess the presence of specific pathogens in wastewater, faecal sludge, and bathing water (the sea). Samples were taken at treatment plants, an open drain, and in the receiving environment (the sea) from June to December 2023. Total nucleic acid wa…

Genetic and Molecular Characterization of Avian Influenza A(H9N2) Viruses from Live Bird Markets (LBM) in Senegal.

Despite extensive experience with influenza surveillance in humans in Senegal, there is limited knowledge about the actual situation and genetic diversity of avian influenza viruses (AIVs) circulating in the country, hindering control measures and pandemic risk assessment. Therefore, as part of the "One Health" approach to influenza surveillance, we conducted active AIV surveillance in two live bird markets (LBMs) in Dakar to better understand the dynamics and diversity of influenza viruses in Senegal, obtain genetic profiles of circulating AIVs, and assess the risk of emergence of novel strai…

Stakeholder engagement to co-design implementation strategies for integrating depression management into HIV care services in Senegal.

BACKGROUND: Depression is highly prevalent in people living with HIV (PWH), affecting their daily life and HIV outcomes. Following a successful pilot study to treat depression in PWH with Group Interpersonal Therapy, we examined its implementation potential. Despite a strong willingness for its adoption routine practice, formal integration of mental health services into HIV care remained challenging. Using Implementation Mapping, we aimed to select and specify a set of implementation strategies to integrate depression services into Senegalese HIV care. METHODS: For each step of depression ser…

Common Molecular Detection of the Neglected Human Malaria Parasite Among Febrile Patients in Southern Regions in Senegal.

BACKGROUND: In sub-Saharan Africa, Plasmodium falciparum is unequivocally responsible for almost all malaria cases and deaths. However, the long-neglected human P. vivax, P. ovale, and P. malariae parasites also emerge as relevant, though their prevalence and contribution to the burden of the disease are very poorly appreciated. This study aimed to bridge this gap and surveyed the circulation of non-falciparum malaria parasites among febrile patients in four regions in south Senegal. METHODS: Blood samples were obtained from 1990 febrile patients during the malaria transmission seasons of 202…

Circulating Hepatitis B Virus (HBV) RNA and Conventional Markers in Treatment-Naive Persons With HBV in Senegal.

BACKGROUND: Hepatitis B virus (HBV) infection affects approximately 10% of the general population in West Africa. Circulating HBV RNA may help improve the characterization of HBV disease and prognosis. We aimed to evaluate the associations between HBV RNA and conventional biomarkers of HBV replication in the Senegalese Hepatitis B Cohort Study (SEN-B). METHODS: We included all treatment-naive, human immunodeficiency virus-negative participants of SEN-B with chronic HBV infection confirmed by a quantitative hepatitis B surface antigen (HBsAg) >0.05 IU/mL. We quantified HBV RNA, HBV DNA, and HB…

Challenging the notion of Aedes aegypti as the primary chikungunya virus vector: insights from Kédougou, Southeastern Senegal.

BACKGROUND: Chikungunya fever (CHIK) caused by the mosquito-borne chikungunya virus (CHIKV) and transmitted by Aedes (Ae.) mosquitoes, remains a public health burden throughout the tropics. During the CHIK outbreak in southeastern Senegal in August 2023, an entomologic investigation was conducted to identify the vector(s) and characterize the virus strains. METHODS: Adult mosquitoes were collected indoors and outdoors from houses of confirmed CHIK cases and their immediate neighborhoods using Prokopack aspirators and double-net traps and all water containers were inspected for aquatic stages. …

Variable effects of non-falciparum species infections on malaria disease severity in high transmission regions in Senegal.

In malaria endemic countries, non-falciparum species are often mixed with Plasmodium falciparum in patients with uncomplicated malaria, and their contribution to malaria severity and death is poorly studied. This study assesses the contribution of non-falciparum species to malaria severity in three regions of Senegal with the highest malaria incidence. We analysed 617 blood samples obtained between 2015 and 2021 from confirmed malaria patients at health facilities in Kedougou, Kolda and Tambacounda in Senegal. Plasmodium species composition was determined by PCR and their distribution were anal…

Two decades of molecular surveillance in Senegal reveal rapid changes in known drug resistance mutations over time.

BACKGROUND: Drug resistance in Plasmodium falciparum is a major threat to malaria control efforts. Pathogen genomic surveillance could be invaluable for monitoring current and emerging parasite drug resistance. METHODS: Data from two decades (2000-2020) of continuous molecular surveillance of P. falciparum parasites from Senegal were retrospectively examined to assess historical changes in malaria drug resistance mutations. Several known drug resistance markers and their surrounding haplotypes were profiled using a combination of single nucleotide polymorphism (SNP) molecular surveillance and…

Ex vivo RSA and Pfkelch13 targeted-amplicon deep sequencing reveal parasites susceptibility to artemisinin in Senegal, 2017.

INTRODUCTION: Malaria control is highly dependent on the effectiveness of artemisinin-based combination therapies (ACTs), the current frontline malaria curative treatments. Unfortunately, the emergence and spread of parasites resistant to artemisinin (ART) derivatives in Southeast Asia and South America, and more recently in Rwanda and Uganda (East Africa), compromise their long-term use in Sub-Saharan Africa where most malaria deaths occur. METHODS: Here, we evaluated ex vivo susceptibility to dihydroartemisinin (DHA) from 38 P. falciparum isolates collected in 2017 in Thiès (Senegal) expres…
