A randomized controlled trial of an emollient with ceramide and filaggrin-associated amino acids for the primary prevention of atopic dermatitis in high-risk infants.

BACKGROUND: Skin barrier dysfunction may precede infantile development of clinical atopic dermatitis (AD). Early-life emollient therapy to enhance barrier function may prevent or modify AD development in high-risk infants. OBJECTIVES: (a) To determine whether daily full-body application of an emollient with ceramide and amino acids (study emollient) can reduce the cumulative AD incidence compared to standard skin care at 1 year of age. (b) To evaluate the study emollient's effect on skin barrier function, natural moisturizing factor and the microbiome using non-invasive biophysical and bioche

Corynebacterium accolens Releases Antipneumococcal Free Fatty Acids from Human Nostril and Skin Surface Triacylglycerols.

UNLABELLED: Bacterial interspecies interactions play clinically important roles in shaping microbial community composition. We observed that Corynebacterium spp. are overrepresented in children free of Streptococcus pneumoniae (pneumococcus), a common pediatric nasal colonizer and an important infectious agent. Corynebacterium accolens, a benign lipid-requiring species, inhibits pneumococcal growth during in vitro cocultivation on medium supplemented with human skin surface triacylglycerols (TAGs) that are likely present in the nostrils. This inhibition depends on LipS1, a TAG lipase necessary

Emollients "Plus" are Beneficial in Both the Short and Long Term in Mild Atopic Dermatitis.

INTRODUCTION: Atopic dermatitis (AD) is a chronic relapsing disease with a pathophysiology including skin barrier damage, microbiome disbalance and inflammation. Classically, emollients maintaining a healthy microbiome are recommended as the basis of any AD severity management. OBJECTIVE: To assess the benefit of a light balm containing vitamin E, tocopherol and glycerine and enriched with Aqua posae filiformis and microresyl (Emollient+) in subjects with mild AD over a period of 168 days. MATERIALS AND METHODS: For this open-label study, subjects above 3 years of age with mild and stable AD

Clinical evaluation of paraprobiotic-associated Bifidobacterium lactis CCT 7858 anti-dandruff shampoo efficacy: A randomized placebo-controlled clinical trial.

OBJECTIVE: The microbiome plays an important role in a wide variety of skin disorders. Hence, dysbiosis in the skin and/or gut microbiome is associated with an altered immune response, promoting the development of skin diseases, such as atopic dermatitis, psoriasis, acne vulgaris and dandruff. Studies have shown that paraprobiotics may be promising for the treatment of skin disorders through microbiota modulation and immunomodulation. So, the objective is to develop an anti-dandruff formulation using a paraprobiotic (Neoimuno) as active ingredient. METHODS: Randomized, double-blind, placebo-c

Staphylococcus aureus Infections and Human Intestinal Microbiota.

Staphylococcus aureus (S. aureus) is a common pathogen that can cause many human diseases, such as skin infection, food poisoning, endocarditis, and sepsis. These diseases can be minor infections or life-threatening, requiring complex medical management resulting in substantial healthcare costs. Meanwhile, as the critically ignored "organ," the intestinal microbiome greatly impacts physiological health, not only in gastrointestinal diseases but also in disorders beyond the gut. However, the correlation between S. aureus infection and intestinal microbial homeostasis is largely unknown. Here, w

Skin Metagenomic Sequence Analysis of Early Candida auris Outbreaks in U.S. Nursing Homes.

Candida auris is a human fungal pathogen classified as an urgent threat to the delivery of health care due to its extensive antimicrobial resistance and the high mortality rates associated with invasive infections. Global outbreaks have occurred in health care facilities, particularly, long-term care hospitals and nursing homes. Skin is the primary site of colonization for C. auris. To accelerate research studies, we developed microbiome sequencing protocols, including amplicon and metagenomic sequencing, directly from patient samples at health care facilities with ongoing C. auris outbreaks.

Innate immunity and microbial dysbiosis in hidradenitis suppurativa - vicious cycle of chronic inflammation.

Hidradenitis Suppurativa (HS) is a chronic multifactorial inflammatory skin disease with incompletely understood mechanisms of disease pathology. HS is characterized by aberrant activation of the innate immune system, resulting in activation of pathways that aim to protect against pathogenic microorganisms, and also contribute to failure to resolve inflammation. Imbalance in innate immunity is evident in deregulation of host antimicrobial peptides (AMPs) and the complement system associated with the microbiome dysbiosis. The pathology is further complicated by ability of pathogens associated w

Unexpected guests in the tumor microenvironment: microbiome in cancer.

Although intestinal microbiome have been established as an important biomarker and regulator of cancer development and therapeutic response, less is known about the role of microbiome at other body sites in cancer. Emerging evidence has revealed that the local microbiota make up an important part of the tumor microenvironment across many types of cancer, especially in cancers arising from mucosal sites, including the lung, skin and gastrointestinal tract. The populations of bacteria that reside specifically within tumors have been found to be tumor-type specific, and mechanistic studies have d

A new approach to describe the taxonomic structure of microbiome and its application to assess the relationship between microbial niches.

BACKGROUND: Data from microbiomes from multiple niches is often collected, but methods to analyse these often ignore associations between niches. One interesting case is that of the oral microbiome. Its composition is receiving increasing attention due to reports on its associations with general health. While the oral cavity includes different niches, multi-niche microbiome data analysis is conducted using a single niche at a time and, therefore, ignores other niches that could act as confounding variables. Understanding the interaction between niches would assist interpretation of the results

The Gut-Skin and Gut-Thyroid Axis in Autoimmunity: Roles of Dysbiosis, Microbial Metabolites, Immune Dysregulation, and Diet in Psoriasis and Hashimoto's Thyroiditis.

Background/Objectives: Psoriasis and Hashimoto's thyroiditis are chronic immune-mediated disorders affecting distinct target organs but sharing overlapping pathogenic mechanisms, including gut dysbiosis, impaired intestinal barrier function, and systemic immune dysregulation. Growing evidence highlights the gut-skin and gut-thyroid axes as important interfaces linking microbial alterations to immune-mediated inflammation. This review aims to synthesize current knowledge on gut microbiota alterations in psoriasis and Hashimoto's thyroiditis, with particular emphasis on intestinal permeability,

Pharmacomicrobiomics in inflammatory skin diseases: past, present, and the future.

This mini review article focuses on pharmacomicrobiomics, or the study of how the composition and activity of microorganisms in the body, in particular in the gut, impact drug pharmacokinetics and pharmacodynamics. This evolving field has profound implications for personalized medicine in the management of chronic inflammatory diseases. Particularly in dermatology, patient response to an expanding collection of biologic and small molecule inhibitor therapies coming out on the market remains unpredictable. The decision to start which therapy depends on physician preference, rather than based on

From niche to niche: investigating microbial communities and their specialised metabolite gene clusters in human microbiomes.

Diverse microbial communities within the human microbiome perform vital functions which influence both health and disease in hosts. Specialized metabolites produced by microbes via biosynthetic gene clusters (BGCs) drive ecological interactions and offer possibilities for therapeutic application. The biosynthetic capabilities of microorganisms present in human microbiomes are still mostly unexplored despite metagenomics advancements. The study examines the variety of microbial communities and BGC locations through metagenomic data from 1,191 samples across eight human microbiomes taken from th

Implications of an Altered Gut Microbiome in Rat Experimental Vascularized Composite Transplantation.

OBJECTIVES: Vascularized composite allotransplantation is a reconstructive option after severe injury but is fraught with complications, including transplant rejection due to major histocompatibility complex mismatch in the context of allogeneic transplant, which in turn is due to altered immuno-inflammation secondary to transplant. The immunosuppressant tacrolimus can prevent rejection. Because tacrolimus is metabolized predominantly by the gut, this immunosuppressant alters the gut microbiome in multiple ways, thereby possibly affecting immunoinflammation. MATERIALS AND METHODS: We performe

Alterations of the microbiome across body sites in systemic lupus erythematosus: A systematic review and meta-analysis.

BACKGROUND: Systemic lupus erythematosus (SLE) is a complex autoimmune disease with unclear etiology. Growing evidence suggests the microbiome plays a role in SLE pathogenesis. However, findings are inconsistent across studies due to factors like small sample sizes and geographical variations. A comprehensive meta-analysis is needed to elucidate microbiome alterations in SLE. OBJECTIVE: This study aimed to provide a systematic overview of microbiota dysbiosis across body sites in SLE through a meta-analysis of alpha diversity indices, beta diversity indices, and abundance taxa of microbiome.

Impact of shared spaces and daily routines on shared microbiome.

The diversity of microbial communities on the fingers and in the oral cavity may vary based on the regularity of shared daily routines. Participants who frequented various shared spaces, including daycare centers, offices, senior centers, universities, and mosques, provided oral and finger-swab samples. Samples from individuals who regularly shared spaces showed a high similarity in the diversity of finger microbial communities, highlighting the significant role of environmental exposure. Participants associated with shared spaces had low intra-group variation, indicating consistent microbial

The exposomal imprint on rosacea: More than skin deep.

Rosacea is a chronic, inflammatory dermatosis driven by a complex interplay of genetic, environmental and lifestyle factors, collectively known as the exposome. This review explores how intrinsic contributors such as genetic susceptibility, immune dysregulation, microbiome alterations, hormonal influences and psychosocial stress intersect with extrinsic triggers like ultraviolet radiation (UVR), air pollution, dietary factors, and climate variability to shape rosacea pathogenesis. Recent advances in single-cell transcriptomics have identified fibroblasts as key components of inflammatory and v

Mycobiota composition and diversity on the skin of the leatherback sea turtle (Dermochelys coriacea).

Due to increased understanding of the importance of high-throughput sequencing, the available literature on the microbiome of humans, non-human animals, and plants has increased in recent years. Unfortunately, research investigating fungi, as part of the microbiome, lags far behind bacterial research, and significant data gaps persist. However, the emergence of significant fungal diseases over the past few decades underscores the importance of understanding the mycobiota and their roles in host health and disease. Filling these knowledge gaps is critical for understanding and preventing diseas

Differential Immune Responses Correlate With Chytridiomycosis Severity in Italian Crested Newts.

In the midst of the current biodiversity crisis, amphibians are severely threatened by emerging diseases such as chytridiomycosis. Characterizing the mechanisms that underlie susceptibility to this disease is fundamental to improve amphibian conservation. Using a comprehensive multi-omics approach, this study investigates the impact of an exposure to the chytrid fungus Batrachochytrium salamandrivorans (Bsal) on the gene expression of Italian crested newts (Triturus carnifex) and on the bacterial symbionts that constitute their skin microbiota. Exposure to Bsal affected multiple components of

Skin Commensal Microbiota Does Not Influence the Clinical Course of Dermatophyte Infection in Persian Cats.

BACKGROUND: Dermatophytes can be isolated from both symptomatic and asymptomatic cats, yet the role of skin commensals in modulating disease expression remains unclear. This study investigates whether microbial skin communities influence the clinical presentation (symptomatic or asymptomatic) of dermatophyte infection in Persian cats. HYPOTHESIS/OBJECTIVES: We hypothesised that the fungal and bacterial skin microbiota differ between Persian cats with and without clinical signs of dermatophytosis. The objectives of this study were to compare the fungal and bacterial skin microbiota among healt
