Magnetic Susceptibility and Area in the Subthalamic Nucleus Measured by QSM for Diagnosis of Progressive Supranuclear Palsy.

BACKGROUND: Iron deposition and atrophy of the subthalamic nucleus (STN) have been reported in progressive supranuclear palsy (PSP). Quantitative susceptibility mapping (QSM), an advanced MRI technique, enables noninvasive assessment of the magnetic susceptibility of biological tissue and has been reported to have good ability to delineate STN. OBJECTIVE: The aim of this study is to evaluate the diagnostic accuracy of a model using magnetic susceptibility and area of the STN assessed by QSM to differentiate PSP from Parkinson's disease (PD). METHODS: A total of 35 patients with PSP and 28 pa

Association of Structural Forms of 17q21.31 with the Risk of Progressive Supranuclear Palsy and MAPT Sub-haplotypes.

IMPORTANCE: The chromosome 17q21.31 region, containing a 900 Kb inversion that defines H1 and H2 haplotypes, represents the strongest genetic risk locus in progressive supranuclear palsy (PSP). In addition to H1 and H2, various structural forms of 17q21.31, characterized by the copy number of α, β, and γ duplications, have been identified. However, the specific effect of each structural form on the risk of PSP has never been evaluated in a large cohort study. OBJECTIVE: To assess the association of different structural forms of 17q.21.31, defined by the copy numbers of α, β, and γ duplication

A Data-Driven Operational Definition of Square-Wave Jerks and Its Diagnostic Utility in Progressive Supranuclear Palsy.

BACKGROUND: The Movement Disorder Society diagnostic criteria for progressive supranuclear palsy (PSP) list frequent square-wave jerks (SWJ) as a suggestive oculomotor feature. A standardised operational definition of SWJ is however presently lacking. OBJECTIVE: To create a data-driven optimal SWJ definition and to assess its diagnostic utility in PSP. METHODS: We prospectively recruited PSP n = 25, Parkinson's disease (PD) n = 32, Alzheimer's disease (AD) n = 35, and controls n = 88. SWJ were recorded with a visual fixation task using video-oculography. Multiple candidate definitions of SWJ

Refining a mouse model of progressive supranuclear palsy through inoculation of human post-mortem brain-derived tau.

OBJECTIVE: A major obstacle to developing effective therapies for Progressive Supranuclear Palsy (PSP), a uniformly fatal 4R tauopathy, is the absence of an animal model that faithfully reproduces the anatomical, cytopathological, and spatiotemporal progression of disease. Inoculation-based models, using human postmortem brain material bearing disease-specific proteopathic tau seeds, hold great translational potential for modeling tauopathies. Here we conducted key studies towards the development of an inoculation-based PSP model, using human postmortem brain to target three subcortical nuclei

Fronto-medial theta instability error signal correlates with postural sway in Parkinson's disease and progressive supranuclear palsy.

BACKGROUND: Postural instability is a common feature in progressive supranuclear palsy (PSP) and idiopathic Parkinson's disease (PD). Understanding the neural mechanisms behind the loss of postural stability may aid identification of patients at an increased risk of falls. OBJECTIVES: To examine differences in postural sway between PSP and PD cohorts during dual-task assessment with electroencephalography (EEG) recording to identify neural mechanisms associated with postural control in Parkinsonism patients. METHODS: We assessed static postural stability in healthy controls (HC; n = 15), peo

Development of a dynamics-enhanced facial algorithm to discriminate progressive supranuclear palsy (PSP) from Parkinson's disease (PD).

BACKGROUND: Progressive supranuclear palsy (PSP) and Parkinson's disease (PD) share overlapping motor features, making early differential diagnosis challenging in clinical practice. Facial hypomimia is common in both conditions, but disease-specific differences in facial dynamics may provide discriminative diagnostic cues. METHODS: We recruited 62 participants (27 PSP and 35 PD) from a tertiary movement-disorders center. Facial videos were recorded during a standardized expression paradigm including smiling, anger, surprise, and rapid blinking. Dynamic geometric and velocity-based facial feat

Impact of gene-gene interactions in Progressive Supranuclear Palsy: new genetic perspectives in the Asian-Indian population.

Genes play an important role in the risk of Progressive Supranuclear Palsy (PSP). Some of the major risk genes identified for PSP include MAPT, STX6, MOBP, and EIF2AK3 in several ethnic groups. However, the interactions among these genes have not been explored in PSP. Therefore, this prospective case-control study aimed to explore the impact of gene-gene interactions in patients with PSP (n = 106) and healthy subjects (n = 109) of Indian ethnicity. Eight single nucleotide polymorphisms (SNPs) of MAPT gene (rs1467967, rs242557, rs3785883, rs2471738, rs8070723, rs7521, rs12185268, and rs62063857
