Astrocytic lysosome deficits reduce alpha-synuclein degradation and induce the spread of pathology.

Parkinson's Disease (PD) is a neurodegenerative disorder that results from a loss of dopaminergic neurons in the substantia nigra. A pathological hallmark of PD is proteinaceous inclusions called Lewy body aggregates, which consist primarily of misfolded neuronal alpha-synuclein (αSyn). PD pathology progression is thought to be driven by a prion-like spread of αSyn aggregates between adjacent neurons; however, the role of other cell types, such as pathology bearing astrocytes, in this process is still elusive. αSyn pathology has been observed in PD patient astrocytes, suggesting that astrocyte

Genetically encoded fluorescent reporters to visualize α-synuclein pathology in live brain.

Lewy bodies, a pathological hallmark of Parkinson's disease, are α-synuclein-enriched cytoplasmic inclusions that drive progressive neurodegeneration. A long-standing yet unmet goal has been the visualization of α-synuclein (α-Syn) inclusions in live brain and measurements of their pathological effects on individual neurons. Here, we developed genetically encoded reporters and knock-in mouse lines to achieve this goal. The reporters exhibited a 5-fold increase in fluorescence upon incorporation into α-Syn inclusions. They reliably reflected α-Syn inclusion propagation in the cortex of awake mi

[Molecular Imaging of Tau and α-Synuclein Pathologies: Technical Advances, Challenges, and Opportunities].

The differential diagnosis of neurodegenerative diseases, particularly Parkinsonian syndromes, remains a major challenge despite improved clinical criteria. Positron emission tomography enables the in vivo visualization of pathological protein aggregates and has therefore become an important tool in molecular imaging. While amyloid PET is already clinically established, tau and α-synuclein PET are increasingly gaining attention in translational research and clinical applications. This review provides a current overview of the development, validation, and clinical application of tau and α-synuc

Cholesterol differentially regulates α-synuclein binding across membrane packing regimes.

α-Synuclein (αSyn) is an intrinsically disordered protein that preferentially binds anionic membranes with lipid packing defects. Cholesterol is an abundant membrane component that regulates packing and organization within membranes, yet its effect on αSyn binding remains unclear, as prior studies report both cholesterol-mediated enhancement and suppression. Here, we investigated whether these conflicting effects reflect differences in the intrinsic packing state of the phospholipid bilayer. Using a quantitative fluorescence microscopy-based binding assay, we measured αSyn binding preferences

Polymeric lysosome-targeting chimeras for extracellular α-synuclein degradation in Parkinson's disease.

Disease progression in Parkinson's disease has been driven by extracellular α-synuclein prion-like seeding throughout the course of the disease and therefore not just by the intracellular accumulation of the protein in isolated aggregates. Current therapies utilizing PROTACs cannot address the extra-cellular effects of α-synuclein spreading in this manner. This article proposes PolyTACs (Polymeric Lysosome-Targeting Chimeras) as hybrid antibody-polymer conjugates which use neuronal exofacial thiol groups produced because of DJ-1/GSH dysregulation to capture α-synuclein pathological conformers

Characterizing Cutaneous α-Synuclein Deposition and Seeding Activity in Parkinson's Disease Subtypes.

OBJECTIVE: Cutaneous phosphorylated α-synuclein (p-syn) and α-synuclein seeding activity are promising biomarkers for Parkinson's disease (PD), but their clinical value remains uncertain due to disease heterogeneity. This study evaluates these two biomarkers in PD patients to inform phenotype-specific diagnosis and disease severity assessment. METHODS: We compared the diagnostic performance of cutaneous p-syn immunostaining and α-synuclein seed amplification assay (syn-SAA) in 108 PD patients and 60 controls. Additional analyses assessed the influence of clinical subtypes on p-syn distributio

Alpha-synuclein co-pathology in a real-world early Alzheimer's disease cohort.

BACKGROUND: Most Alzheimer's disease (AD) cases show mixed pathology, with α-synuclein (αSyn) aggregates present in a substantial proportion. The cerebrospinal fluid (CSF) α-synuclein seed amplification assay (αS-SAA) enables in vivo detection of pathogenic αSyn aggregates, but its clinical significance remains unclear. METHODS: We prospectively evaluated 108 individuals with mild cognitive impairment or mild dementia due to suspected AD undergoing lumbar puncture for anti-amyloid therapy (ATT) eligibility. CSF AD biomarkers and αS-SAA were analyzed alongside cognitive, olfactory, and rapid e

Stage-Dependent β-Synuclein Links MRI and Cognitive Decline in Alzheimer's Disease.

OBJECTIVE: Synaptic degeneration drives cognitive decline in Alzheimer's disease (AD), but synaptic biomarkers are scarce. Brain-enriched β-synuclein emerged as a synaptic damage marker. We investigated its diagnostic, prognostic, and structural correlates across the AD continuum. METHODS: In a tertiary-center cohort (n = 306), CSF β-synuclein was measured. Cognitively unimpaired (CU), AD-MCI, AD dementia (ADD), and non-AD (FTD, PD, DLB, others) groups were included. ANCOVA compared groups (age/sex adjusted); ROC assessed diagnostic performance. Multivariable regression examined 2-year MMSE d

In situ NMR and integrative proteomics reveal the interaction signature of serum α-synuclein.

Alpha-synuclein (αS) plays a central role in several neurodegenerative diseases. Although predominantly neuronal, αS is also present in peripheral fluids such as serum and cerebrospinal fluid (CSF), where it may contribute to disease propagation. However, its extracellular interactions remain poorly understood. By combining in situ NMR spectroscopy and bottom-up MS proteomics, we reveal that monomeric αS exhibits distinct interactomes in serum versus CSF. Both N- and C-terminal αS regions bind to serum components, while in CSF, αS remains largely unbound. The N- and C-terminal interactions are

Lipid Acyl Chain-Driven α-Synuclein Fibril Polymorphisms and Neuronal Pathologies.

Conformational variations in α-syn fibrils are thought to underlie the distinct clinical features of synucleinopathies, including Lewy body dementia (LBD), Parkinsons's disease (PD), and multiple system atrophy (MSA), suggesting that distinct fibril structures act as molecular fingerprints linked to disease phenotype. While the origins of these conformational variations remain unclear, increasing evidence points to membranes as key modulators of fibrils conformations. In this study, we investigated how age-related alterations in membrane composition and fluidity influence α-syn fibril formatio

Modulation of hierarchical motor sequence learning by alpha-synuclein and the adenosine A2A receptor.

Parkinson's disease (PD) is characterized pathologically by the aggregation of alpha-synuclein (α-Syn) and dopaminergic degeneration and clinically by motor and non-motor deficits, including executive dysfunction and a reduction in the speed of information processing. The neuronal mechanisms underlying executive dysfunction in PD has yet to be elucidated; consequently, there is no effective treatment. In this study, we combined a hierarchical motor sequence learning paradigm, composed of multiple layers of action organization, to simulate the planning, initiation, termination and transition of

α-Synuclein-Targeted Immunotherapies in Parkinson's Disease: In Silico, In Vitro and Clinical Perspectives.

α-synuclein (α-syn) aggregation in dopaminergic neurons is a central event in Parkinson's disease (PD) pathogenesis. Immunotherapeutic strategies targeting α-syn, including passive and active approaches, aim to inhibit aggregation, propagation, and toxicity of pathological species while promoting their clearance via immune mechanisms. This review summarizes α-syn directed immunotherapies evaluated in in silico, in vitro, and in vivo models, as well as early phase clinical trials, focusing on how epitope selection and antibody formats influence efficacy, safety, and target engagement. Locus coeruleus α-synuclein overexpression induces prodromal Parkinsonian features in mice.

The locus coeruleus is among the earliest brain regions affected by α-synuclein pathology in prodromal Parkinson's disease, contributing to non-motor symptoms, yet its early pathogenic role remains unclear. We developed a mouse model in which human α-synuclein was selectively overexpressed in noradrenergic locus coeruleus neurons using PRSx8-driven adeno-associated viral vector. Mice were analysed at 1 and 3 months post-injection using histological, behavioural, neurochemical and electrophysiological approaches. Monomeric α-synuclein was localized in the locus coeruleus cell bodies and distrib

α-synuclein positivity is associated with decline in brain microstructure in the Alzheimer's disease spectrum.

BACKGROUND: α-Synuclein co-pathology is common in Alzheimer's disease (AD) and is associated with faster cognitive decline, yet its impact on brain structural integrity remains unclear. We examined the extent to which α-synuclein pathology influences brain microstructural changes using diffusion tensor imaging in participants stratified by cognitive impairment status. METHODS: Participants from the Alzheimer’s Disease Neuroimaging Initiative with available CSF α-synuclein SAA results, diffusion MRI, and AD biomarkers were analyzed. Subjects were classified as cognitively unimpaired (CU) or cog

Loss of epigenetic adaptation to a high-fat diet in alpha-synuclein transgenic mice.

INTRODUCTION: In humans, a high-fat diet and obesity are associated with a higher risk and accelerated progression of Parkinson's disease (PD). Similarly, in animal models a high-fat diet exacerbates PD-related phenotypes, including dopaminergic neurodegeneration, and alpha-synuclein aggregation. We previously demonstrated that transgenic mice overexpressing human, mutated A30P alpha-synuclein failed to transcriptionally adapt to metabolic stress which could be a potential explanation for the high-fat diet-dependent aggravation of PD pathology. However, the underlying epigenetic mechanisms tha

Distinct Role of γ-Synuclein in the Regulation of Motor Performance and Behavioral Responses in Mice.

Background: The three Synuclein family members (α-, β-, and γ-synuclein) are presynaptic proteins that regulate synaptic vesicle trafficking and thereby influence neurotransmitter release. Synucleins belong to a class of intrinsically disordered proteins and are prone to aggregation into pathological deposits, which may impair their physiological synaptic functions. Knockout (KO) mouse lines, commonly used to model synuclein depletion in the nervous system, reveal a range of phenotypes with different motor and behavioral deficits. However, given the high sequence homology and functional interp…

Targeted α-synuclein mRNA degradation by PMO-based RNA-degrading chimeras.

α-Synucleinopathies are devastating neurodegenerative diseases characterized by pathological accumulation of a neuronal protein, α-synuclein (αSyn). Lowering soluble αSyn levels is a promising therapeutic strategy to limit aggregation and neurotoxicity, but directly targeting this protein is hindered by its intrinsically disordered structure and other factors, such as its conformational heterogeneity and intracellular drug delivery barriers. Consequently, increasing attention has been directed toward targeting the SNCA transcript, which encodes αSyn. Here, we developed phosphorodiamidate morph

Impact of N-terminal acetylation on Cu(I) coordination by alpha synuclein protein.

Amyloid aggregation of alpha-synuclein (AS) protein is associated with Parkinson's disease. Physiologically, AS plays a crucial role in the uptake, storage, and recycling of neurotransmitter vesicles. AS has three independent binding sites for Cu(II) and Cu(I) ions. N-terminal acetylation of AS impacts the highest-affinity site of Cu, encompassing the first five residues; it prevents Cu(II) coordination, enhances Cu(I) binding affinity, raises its redox potential, and extends the α-helix to the first ten residues. In this study, X-ray absorption spectroscopy and electronic structure calculatio
