Magnetic Tissue Engineering for Voice Rehabilitation - First Steps in a Promising Field.

BACKGROUND/AIM: The voice is one of the most important instruments of communication between humans. It is the product of intact and well-working vocal folds. A defect of these structures causes dysphonia, associated with a clear reduction of quality of life. Tissue engineering of the vocal folds utilizing magnetic cell levitation after nanoparticle loading might be a technique to overcome this challenging problem. MATERIALS AND METHODS: Vocal fold fibroblasts (VFFs) were isolated from rabbit larynges and cultured. For magnetization, cells were incubated with superparamagnetic iron oxide nanop…

Novel Poly(3-hydroxybutyrate-g-vinyl alcohol) Polyurethane Scaffold for Tissue Engineering.

The design of new synthetic grafted poly(3-hydroxybutyrate) as composite 3D-scaffolds is a convenient alternative for tissue engineering applications. The chemically modified poly(3-hydroxybutyrate) is receiving increasing attention for use as biomimetic copolymers for cell growth. As of yet, these copolymers cannot be used efficiently because of the lack of good mechanical properties. Here, we address this challenge, preparing a composite-scaffold of grafted poly(3-hydroxybutyrate) polyurethane for the first time. However, it is unclear if the composite structure and morphology can also offer…

Tissue engineering of the temporomandibular joint disc: current status and future trends.

INTRODUCTION: Temporomandibular joint disorders are extremely prevalent and there is no ideal treatment clinically for the moment. For severe cases, a discectomy often need to be performed, which will further result in the development of osteoarthritis. In the past thirty years, tissue engineering has provided a promising approach for the effective remedy of severe TMJ disease through the creation of viable, effective, and biological functional implants. METHODS: Although TMJ disc tissue engineering is still in early stage, unremitting efforts and some achievements have been made over the pas…

A novel bioprinting method and system for forming hybrid tissue engineering constructs.

Three dimensional (3D) bioprinting is a promising approach to form tissue engineering constructs (TECs) via positioning biomaterials, growth factors, and cells with controlled spatial distribution due to its layer-by-layer manufacturing nature. Hybrid TECs composed of relatively rigid porous scaffolds for structural and mechanical integrity and soft hydrogels for cell- and growth factor-loading have a tremendous potential to tissue regeneration under mechanical loading. However, despite excessive progress in the field, the current 3D bioprinting techniques and systems fall short in integration…

Biomaterials and Tissue Engineering Strategies for Conjunctival Reconstruction and Dry Eye Treatment.

The ocular surface is a component of the anterior segment of the eye and is covered by the tear film. Together, they protect the vital external components of the eye from the environment. Injuries, surgical trauma, and autoimmune diseases can damage this system, and in severe cases, tissue engineering strategies are necessary to ensure proper wound healing and recovery. Dry eye is another major concern and a complicated disease affecting the ocular surface. More effective and innovative therapies are required for better outcomes in treating dry eye. This review focuses on the regenerative medi…

Tissue engineering of tumor stromal microenvironment with application to cancer cell invasion.

3D organotypic cultures of epithelial cells on a matrix embedded with mesenchymal cells are widely used to study epithelial cell differentiation and invasion. Rat tail type I collagen and/or matrix derived from Engelbreth-Holm-Swarm mouse sarcoma cells have been traditionally employed as the substrates to model the matrix or stromal microenvironment into which mesenchymal cells (usually fibroblasts) are populated. Although experiments using such matrices are very informative, it can be argued that due to an overriding presence of a single protein (such as in type I Collagen) or a high content

Collagen intermingled chitosan-tripolyphosphate nano/micro fibrous scaffolds for tissue-engineering application.

Scaffolds comprising a nano- and micro-fibrous architecture are promising for tissue engineering, where nanofibers act as connecting network among microfibers and provide a 3D structural environment and mechanical stability for facilitating cell attachment, proliferation and migration. In this study, a novel structure was developed with polymeric micro and nano combined fibrous architecture, which aims to mimic the native extracellular matrix for tissue regeneration. Chitosan-tripolyphosphate (TPP) microfibers were prepared by wet spinning method, where collagen solution was allowed to self-as…

Embryonic wound healing: a primer for engineering novel therapies for tissue repair.

Scar is the default tissue repair used by the body in response to most injuries-a response that occurs in wounds ranging in seriousness from minor skin cuts to complete severance of the spinal cord. By contrast, before the third trimester of pregnancy embryonic mammals tend to heal without scarring due to a variety of mechanisms and factors that are uniquely in operation during development in utero. The goal of tissue engineering is to develop safe and clinically effective biological substitutes that restore, maintain, or improve tissue function in patients. This review provides a comparative …

Tissue engineering craniofacial defects with adult stem cells? Are we ready yet?

Over three-quarters of all craniofacial defects observed in the US per year are cleft palates. Usually involving significant bony defects in both the hard palate and alveolar process of the maxilla, repair of these defects is typically performed surgically using autologous bone grafts taken from appropriate sites (i.e., iliac crest). However, surgical intervention is not without its complications. As such, the reconstructive surgeon has turned to the scientist and engineer for help. In this review, the application of the field of tissue engineering to craniofacial defects (e.g., cleft palates)…

[Current status of researches in the development of blood tissue engineering product].

The term "blood substitutes" includes plasma substitutes and blood cell substitutes in the broad sense, but in its narrow sense, it means red blood cell (RBC) substitutes, platelet substitutes and white blood cell (WBC) substitutes. The RBC substitutes includes perfluorocarbon, hemoglobin-based and encapsuled substitutes. The hemoglobin-based substitutes which was widely researched in the world includes human hemoglobin-based, animal hemoglobin-based and gene recombined hemoglobin based substitutes. The function and immunology of WBC is very complicated, so it is rarely used in clinic. Nowaday

Porous poly (DL-lactic acid) modified chitosan-gelatin scaffolds for tissue engineering.

Chitosan-gelatin (Cs-Gel) scaffolds are modified with poly (DL-lactic acid) (PDLLA) dichloromethane solution of different concentrations (0.1, 0.5, and 1.0%) and immersed in water after the evaporation of the solvent. The swollen scaffolds are freeze dried. The concentration of PDLLA has significant effects on both the physicomechanical properties and the cytocompatibility. Data reveal that only the 0.1% concentration could increase the tensile strength fourfold in comparison with the pristine Cs-Gel scaffold, while maintaining the human fibroblast adhesion, migration, and proliferation just l

Biodegradable composite scaffolds with an interconnected spherical network for bone tissue engineering.

Tissue engineering scaffolds are highly engineered structures that accommodate cells, facilitate their expression, and resorb to facilitate regeneration of tissue. A new technique for producing controlled pore shape and pore size interconnectivity offers promise for application as a tissue engineering scaffold. Salt particles were spheroidized in a flame and sintered to provide an interconnecting salt template. The salt template was filled with a carbonated fluorapatite powder and a polylactic polymer to produce a composite scaffold. It was found that a higher pore space is possible with the u…

Tissue engineering of a human sized and shaped auricle using a mold.

OBJECTIVES: The creation of a tissue-engineered auricle was initially successful in an immunocompromised nude mouse model. Subsequently, an immunocompetent porcine model successfully generated a helical construct. We wished to evaluate the novel technique of using a mold to create a complete, anatomically refined auricle in a large animal model. METHODS: Mixtures of autogenous chondrocytes and biodegradable polymers were used inside a perforated, auricle shaped hollow gold mold. Three biodegradable polymers (calcium alginate, pluronic F-127, and polyglycolic acid) were used to retain the seed

Autologous cell transplantation and cardiac tissue engineering: potential applications in heart failure.

The promising concept of cell transplantation and cardiac tissue engineering has been developed in the last few years and focused on strategies attempting to replace dysfunctional, necrotic, and/or apoptotic cardiomyocytes with new cells of mesodermal origin. Transplantation of autologous cells minimizes the risk of neoplasia and avoids immune rejection associated with allogenic or xenogenic cells and recent data hold enormous hopes for short term clinical practices. Tissue engineering represents another promising approach that makes possible the creation of new functional tissues to replace t…

Structure and function of the temporomandibular joint disc: implications for tissue engineering.

The temporomandibular joint (TMJ) disc is a little understood structure that, unfortunately, exhibits a plethora of pathologic disorders. Tissue engineering approaches may be warranted to address TMJ disc pathophysiology, but first a clear understanding of structure-function relationships needs to be developed, especially as they relate to the regenerative potential of the tissue. In this review, we correlate the biochemical content of the TMJ disc to its mechanical behavior and discuss what this correlation infers for tissue engineering studies of the TMJ disc. The disc of the TMJ exhibits a …

Advanced cell-instructive bio-inks: Engineering multiscale niches for functional tissue formation.

Three-dimensional bioprinting has transformed tissue fabrication by enabling the precise spatial organization of living cells and biomaterials into biomimetic tissue constructs. However, advances in manufacturing have outpaced the development of bio-inks capable of actively directing tissue formation. Most existing bio-inks remain optimized for printability, rheological behaviour, and structural fidelity, while providing limited control over the biological processes that govern tissue morphogenesis, functional maturation, and regeneration. This review examines the emerging paradigm of cell-ins
