Predicted no effect concentration derivation as a significant source of variability in environmental hazard assessments of chemicals in aquatic systems: an international analysis.

Environmental hazard assessments for chemicals are carried out to define an environmentally "safe" level at which, theoretically, the chemical will not negatively affect any exposed biota. Despite this common goal, the methodologies in use are very diverse across different countries and jurisdictions. This becomes particularly obvious when international scientists work together on documents with global scope, e.g., in the World Health Organization (WHO) International Program on Chemical Safety. In this article, we present a study that describes the extent of such variability and analyze the re

Early stellate cell activation and veno-occlusive-disease (VOD)-like hepatotoxicity in dogs treated with AR-H047108, an imidazopyridine proton pump inhibitor.

Dogs treated with AR-H047108, an imidazopyridine potassium competitive acid blocker (P-CAB), developed clinical signs of hepatic dysfunction as well as morphologically manifest hepatotoxicity in repeat-dose toxicity studies. An investigative one-month study was performed, with interim euthanasia after one and two weeks. A detailed histopathological and immunohistochemical characterization of the liver lesions was conducted, including markers for fibrosis, Kupffer cell activation, apoptosis, and endothelial injury. In addition, hepatic retinoid and procollagen 1alpha2 mRNA levels in livers of d

Single and combined chronic toxicity of polystyrene nanoplastics (PSNP) and clothianidin on collembolans and enchytraeids.

This study assessed the chronic toxicity of polystyrene nanoplastics (PSNP) and clothianidin (CLO), individually and as a binary mixture, on enchytraeids Enchytraeus crypticus and collembolans Folsomia candida. The assays were performed according to ISO protocols, using a tropical soil (Entisol) as the test substrate. The mixture assays were performed using equitoxic concentration, standardized according to the preliminary EC50 values for each contaminant (1 Toxic Unit = reproductive EC50 average), and the toxicological interactions were assessed through the Abbott model. Individually, CLO was

In Vitro Toxicity Assessment of Tire-related Chemicals in Cells from Diverse Fish Species.

Urban stormwater runoff introduces tire‑related contaminants such as 6PPD‑quinone (6PPD‑Q), which has been shown to exhibit acute toxicity in some salmonid species, although the underlying toxic mechanisms remain incompletely understood. To expand in vitro toxicological assessments, we evaluated the cytotoxicity of 3,4‑dichloroaniline and 6PPD‑quinone using six fish‑derived cell lines from salmonids and phylogenetically distant species, based on nominal concentrations at the time of addition. Among these cell lines, CSE‑119 cells, which are derived from coho salmon, exhibited the highest sensi

Growth and chlorophyll-a responses of marine microalgae to biodegradable polypropylene microplastic leachates: an initial toxicity assessment.

Biodegradable plastics are increasingly promoted as alternatives to conventional plastics; however, their degradation may release leachates containing potentially harmful compounds. Complementary field observations on microplastic occurrence along Indonesian coasts underscore the environmental relevance of this study. In marine environments, such leachates may affect algal communities. This study evaluated the toxicity of leachates derived from biodegradable polypropylene (PP) microplastics on three marine microalgal species (Chlorella sp., Tetraselmis sp., and Navicula sp.). Non-clinical toxicology of therapeutic vaccines: Regulatory considerations and species selection: A comprehensive review.

Vaccines, a proven medical innovation, have reduced mortality from infectious diseases for billions worldwide, when used prophylactically. Vaccine technology is also used to treat disease, including cancer and autoimmune disease, offering effective therapies with limited side effects. Vaccines have a good safety record and most adverse events are transient and minor. Severe adverse events occur but are extremely rare. Safety testing of vaccines is essential and require a toxicology assessment. However, therapeutic vaccines lack specific regulatory guideline and definitions vary globally, which

Evaluating Therapeutic and Chemical Toxicity Using Organ-Cultured Porcine Corneas and Epithelial Wound Healing.

Due to its anatomical and physiological similarities to the human eye, the porcine eye serves as a robust model for biomedical research and ocular toxicity assessment. An air/liquid corneal culture system using porcine eyes was developed, and ex vivo epithelial wound healing was utilized as a critical parameter for these studies. Fresh pig corneas were processed for organ culture, with or without epithelial wounding. The corneas were cultured in a humidified 5% CO2 incubator at 37 °C in MEM, with or without testing agents. Corneal permeability and wound healing rates were measured, and epithel

A hidden artefact: how surfactants can distort the results of springtail reproduction tests.

Soils are exposed to multiple substance groups, including surfactants, which directly enter soils when they are used as additives in firefighting liquids or pesticide mixtures due to their surface tension-lowering properties. The impact of chemicals on soil health is often tested with the springtail reproduction test. We tested the effects of the trisiloxane Break-Thru® S 301 on the reproduction of Folsomia candida in three soils according to Organisation for Economic Co-operaton and Development (OECD) guideline 232. Juveniles were extracted either by heat or flotation. In the latter method, r

Immunogenicity-related placental infarcts and abortions in nonhuman primate developmental and reproductive toxicity studies: Proposed mechanism and impact on study interpretation.

Previous analysis of 3 developmental toxicity studies (1 EFD, 2 ePPND) conducted in cynomolgus monkeys for biotherapeutics indicated an association between immunogenicity and abortions. This led to the hypothesis that immunogenic responses to biotherapeutics administered to pregnant nonhuman primates (NHPs) can cause altered placental hemodynamics and/or vascular pathology, resulting in placental infarcts and abortion secondary to compromised uteroplacental perfusion. Retrospective analysis of an additional 17 NHP developmental and reproductive toxicity (DART) studies was conducted for further

Bee sensitivity derived from acute contact tests biased by standardised protocols?

In an acute contact test with bees the compound of interest is dissolved in a carrier solvent (frequently acetone) and then a droplet of the solution is placed on the dorsal thorax of the bee. The volume of the droplet is standardised to 1 µL for honeybees and to 2 µL for bumblebees. In practice the same droplet volume is used for bees with very different sizes. In this research the effect of the droplet volume was evaluated with acute contact tests with dimethoate for the alfalfa leafcutter bee, the red mason bee, the honeybee and the bumblebee. The results were analysed with a ToxicoKinetic

A review of the scientific literature on experimental toxicity studies of COVID-19 vaccines, with special attention to publications in toxicology journals.

Since the reports of the first cases of COVID-19, in less than 5 years, a huge number of documents regarding that disease and the coronavirus (SARS-CoV-2), responsible for the infection, have been published. The tremendous number of scientific documents covers many topics on different issues directly related to COVID-19/SARS-CoV-2. The number of articles-including reviews-reporting adverse/side effects of the approved COVID-19 vaccines is considerable. A wide range of adverse/side effects have been reported in humans after COVID-19 vaccination: thrombotic events/thrombocytopenia, myocarditis/p

Generation, characterization, and toxicological assessment of reference ultrafine soot particles with different organic content for inhalation toxicological studies.

Ultrafine particles (UFP) are the smallest atmospheric particulate matter linked to air pollution-related diseases. The extent to which UFP's physical and chemical properties contribute to its toxicity remains unclear. It is hypothesized that UFP act as carriers for chemicals that drive biological responses. This study explores robust methods for generating reference UFP to understand these mechanisms and perform toxicological tests. Two types of combustion-related UFP with similar elemental carbon cores and physical properties but different organic loads were generated and characterized. Huma

Influence of exposure scenario on the sensitivity to caffeine.

The chorion acts as a protective barrier, restricting some chemical absorption into the embryo and the surrounding fluids. In this sense, la … rials or chemicals with low capacity to cross the chorion. Therefore, the use of hatched embryos in toxicity tests is suggested, as they allow a shorter and less expensive exposure scenario that provides similar outcome as the conventional scenario.

Acute toxicity of tire wear particles, leachates and toxicity identification evaluation of leachates to the marine copepod, Tigriopus japonicus.

Tire wear particles (TWPs) have been characterized as microplastics in recent years, and many of these TWPs will be eventually deposited in coastal areas, leading to adverse effects to marine organisms. Results of the acute toxicity test in this study showed that the 96-h LC50 values of the particles and leachate were 771.4 mg/L (95% CI = 684.4-869.6 mg/L) and 5.34 g/L (95% CI = 4.75-6.07 g/L), respectively. The chemical constituents of TWP and the leachate are very complex, and little research has been conducted to determine which of these constituents contribute to the toxicity of TWP leacha

New framework for a non-animal approach adequately assures the safety of cosmetic ingredients - A case study on caffeine.

This case study on the model substance caffeine demonstrates the viability of a 10-step read-across (RAX) framework in practice. New approach methodologies (NAM), including RAX and physiologically-based kinetic (PBK) modelling were used to assess the consumer safety of caffeine. Appropriate animal systemic toxicity data were used from the most relevant RAX analogue while assuming that no suitable animal toxicity data were available for caffeine. Based on structural similarities, three primary metabolites of the target chemical caffeine (theophylline, theobromine and paraxanthine) were selected
