Assessing human variability in kinetics for exposures to multiple environmental chemicals: a physiologically based pharmacokinetic modeling case study with dichloromethane, benzene, toluene, ethylbenzene, and m-xylene.

The objective of this study was to compare the magnitude of interindividual variability in internal dose for inhalation exposure to single versus multiple chemicals. Physiologically based pharmacokinetic models for adults (AD), neonates (NEO), toddlers (TODD), and pregnant women (PW) were used to simulate inhalation exposure to "low" (RfC-like) or "high" (AEGL-like) air concentrations of benzene (Bz) or dichloromethane (DCM), along with various levels of toluene alone or toluene with ethylbenzene and xylene. Monte Carlo simulations were performed and distributions of relevant internal dose met

Historical perspectives on cadmium toxicology.

The first health effects of cadmium (Cd) were reported already in 1858. Respiratory and gastrointestinal symptoms occurred among persons using Cd-containing polishing agent. The first experimental toxicological studies are from 1919. Bone effects and proteinuria in humans were reported in the 1940's. After World War II, a bone disease with fractures and severe pain, the itai-itai disease, a form of Cd-induced renal osteomalacia, was identified in Japan. Subsequently, the toxicokinetics and toxicodynamics of Cd were described including its binding to the protein metallothionein. International w

Metallothionein and Other Factors Influencing Cadmium-Induced Kidney Dysfunction: Review and Commentary.

Cadmium is widely recognized as an important environmental toxicant that may give rise to kidney dysfunction, bone disease, and cancer in humans and animals. Kidney dysfunction occurs at very low exposures and is often considered as the most sensitive or critical effect. Cadmium exposures of concern occur in many countries. In low- and middle-income countries with small-scale mining, excessive exposure to cadmium and other metals occurs in occupational and environmental settings. This is of particular importance in view of the growing demand for metals in global climate change mitigation. Sinc

Genetic differences in the aryl hydrocarbon receptor and CYP1A2 affect sensitivity to developmental polychlorinated biphenyl exposure in mice: relevance to studies of human neurological disorders.

Polychlorinated biphenyls (PCBs) are persistent organic pollutants that remain a human health concern with newly discovered sources of contamination and ongoing bioaccumulation and biomagnification. Children exposed during early brain development are at highest risk of neurological deficits, but highly exposed adults reportedly have an increased risk of Parkinson's disease. Our previous studies found allelic differences in the aryl hydrocarbon receptor and cytochrome P450 1A2 (CYP1A2) affect sensitivity to developmental PCB exposure, resulting in cognitive deficits and motor dysfunction. Simultaneous detection of the tetrachloroethylene metabolites S-(1,2,2-trichlorovinyl) glutathione, S-(1,2,2-trichlorovinyl)-L-cysteine, and N-acetyl-S-(1,2,2-trichlorovinyl)-L-cysteine in multiple mouse tissues via ultra-high performance liquid chromatography electrospray ionization tandem mass spectrometry.

Tetrachloroethylene (perchloroethylene; PERC) is a high-production volume chemical and ubiquitous environmental contaminant that is hazardous to human health. Toxicity attributed to PERC is mediated through oxidative and glutathione (GSH) conjugation metabolites. The conjugation of PERC by glutathione-s-transferase to generate S-(1,2,2-trichlorovinyl) glutathione (TCVG), which is subsequently metabolized to form S-(1,2,2-trichlorovinyl)-L-cysteine (TCVC) is of special importance to human health. Specifically, TCVC may be metabolized to N-acetyl-S-(1,2,2-trichlorovinyl)-L-cysteine (NAcTCVC) whi

Tissue distribution, metabolism and hepatic tissue injury in Chinese lizards (Eremias argus) after a single oral administration of lambda-cyhalothrin.

Lambda-cyhalothrin (LCT) is a widely used pyrethroid with neurotoxicity. However, little is known about the toxicokinetics of LCT in reptiles. In this study, the absorption, distribution, metabolism and excretion of LCT in Chinese lizards (Eremias Argus) were determined following a single dose (10 mg kg-1) treatment. In the liver, brain, gonads and skin, LCT levels peaked within several hours and then decreased rapidly. However, the concentration of LCT gradually increased in the fat tissue. More than 90% of the LCT dose was excreted in the faeces. One LCT metabolite, 3-phenoxybenzoic acid (PB

Comprehensive analysis of 5-aminolevulinic acid dehydrogenase (ALAD) variants and renal cell carcinoma risk among individuals exposed to lead.

BACKGROUND: Epidemiologic studies are reporting associations between lead exposure and human cancers. A polymorphism in the 5-aminolevulinic acid dehydratase (ALAD) gene affects lead toxicokinetics and may modify the adverse effects of lead. METHODS: The objective of this study was to evaluate single-nucleotide polymorphisms (SNPs) tagging the ALAD region among renal cancer cases and controls to determine whether genetic variation alters the relationship between lead and renal cancer. Occupational exposure to lead and risk of cancer was examined in a case-control study of renal cell carcinoma

[Ultrafine particles and effects on the body: review of the literature].

The International laws and the technology developments led to a situation where the current levels of environmental pollution are below those that existed at the beginning of the century: however, these pollution levels produce harmful effects to health linked to an increase in morbidity and mortality. Over the years the pollution has changed: following the transformation of heating, motor innovation and emissions reducing, has been a reduction in air concentration of some conventional pollutants (sulfur dioxide, carbon monoxide, benzene), while there wasn't a significant reduction of particul

A simple and highly sensitive spectrophotometric method for the determination of cyanide in equine blood.

An epidemiological association among black cherry trees (Prunus serotina), eastern tent caterpillars (Malacosoma americana), and the spring 2001 episode of mare reproductive loss syndrome in central Kentucky focused attention on the potential role of environmental cyanogens in the causes of this syndrome. To evaluate the role of cyanide (CN (-)) in this syndrome, a simple, rapid, and highly sensitive method for determination of low parts per billion concentrations of CN (-) in equine blood and other biological fluids was developed. The analytical method is an adaptation of methods commonly in

Effects of simultaneous low-level dietary supplementation with inorganic and organic selenium on whole-body, blood, and organ levels of toxic metals in mice.

Classical experiments have demonstrated that Se compounds protect against the toxicity of several toxic metals in acute experiments with simultaneous parenteral administration of high doses of Se and the toxic metal. Blood and organ levels of the toxic metals were increased, conceivably due to formation of inert Se complexes. Less is known about effects of long-term Se status on the toxicokinetics of toxic metals. Possible Se interactions in toxic metal biokinetics should therefore be studied at Se levels ranging from those just sufficient to avoid Se deficiency and up to those believed to be

T4-mediated rescue of aortic malformations in hypothyroid rats indicates maternal thyroid status can affect great vessel development.

Pexacerfont is a corticotrophin-releasing factor subtype 1 receptor (CRF-1) antagonist developed for potential treatment of anxiety and stress-related disorders. In male rats, pexacerfont caused hepatic enzyme induction leading to increased thyroxine (T4) clearance. When administered to pregnant rats on gestation day 6 to 15, pexacerfont at 300 mg/kg/day (30× mean AUC in humans at 100 mg/day) produced similar effects on thyroid homeostasis with serum T4 and thyroid-stimulating hormone levels that were 0.3-0.5× and 3.3-3.7× of controls, respectively. At this dose, fetuses of pexacerfont-treated

Physics-informed neural ordinary differential equations for PBPK-based internal dose prediction in environmental health risk assessment.

Reliable prediction of internal doses, especially systemic blood concentrations, is crucial for exposure science and environmental health risk assessment. While traditional physiologically based pharmacokinetic (PBPK) models provide mechanistic insights, their fixed parameters often fail to capture nonlinear dynamics at high concentrations, as encountered in acute contamination scenarios. This study introduces a hybrid PBPK-constrained neural ODE framework that combines neural ordinary differential equations with PBPK modeling. By integrating a minimal two-neuron network correction at the live

Protein-Affinity-Guided Identification of Bioaccumulative Silanol Quaternary Ammonium Compounds in Indoor Environments and Human Serum.

Quaternary ammonium compounds (QACs) have raised concerns due to their widespread use in disinfectants and unknown bioaccumulation behavior. However, conventional bioaccumulation assessments are costly, time-consuming, and low-throughput, limiting their utility for screening the growing array of emerging QACs. In this study, we developed a protein affinity ultrafiltration mass spectrometry (PA-UF-MS) strategy using human serum albumin (HSA) as a molecular bait to selectively isolate bioaccumulative QACs from disinfectants. We identified 12 traditional and emerging QACs, including several silan

Bridging the Gap between External and Internal Exposure: Challenges and a Machine Learning Approach Validated Using Benzene Ring-Containing Pollutants.

Establishing quantitative linkages between external and internal exposures is essential for accurate exposure and health risk assessment, yet this goal remains hindered by multiple challenges. This Perspective systematically reviews major challenges limiting the quantitative linkage across three dimensions: (i) External exposure: complex multimedia and multipathway exposure, environmental factor variability, and substantial data gaps; (ii) Internal exposure: difficulties in biological sample collection, large-scale biomonitoring, and appropriate biomarker selection; and (iii) External-internal

A fast and green UPCC-MS/MS method for analyzing the tissue distribution of polyethylene glycol polymers (7-20 subunits) in zebrafish.

The structural heterogeneity and environmental persistence of polydisperse polymers, such as polyethylene glycol (PEG), complicate accurate ecological risk assessment. Conventional analytical techniques often suffer from inadequate sensitivity or excessive solvent consumption when analyzing complex biological matrices, hindering the precise quantification of individual oligomers. Herein, we report a high-throughput, sustainable analytical framework utilizing ultra-performance convergence chromatography-tandem mass spectrometry (UPCC-MS/MS) to characterize the toxicokinetic and spatiotemporal t

Micro- and Nanoplastic Pollution and Renal Health: Human Evidence, Mechanisms, and Clinical Implications.

Micro- and nanoplastics (MNPs) are reported in blood, urine, and tissues, including the kidney, raising concern that the urinary system may be both a target and a route of elimination. This narrative review integrates analytical, toxicokinetic, experimental, epidemiological, and dialysis-related evidence on MNPs and renal health. Human studies report polymer-confirmed particles or polymer-associated signals in blood, urine, and kidney-related specimens, supporting systemic presence and compatibility with renal exposure; however, estimates are strongly method-dependent and do not establish anat

Pacific Blue Derivatives of Paclitaxel as Fluorescent Probes of OATP1B-Type Transporters.

Paclitaxel (Taxol) is a widely used anticancer agent that undergoes extensive hepatic metabolism and that causes a debilitating, dose-limiting peripheral neurotoxicity. We previously reported that the uptake of paclitaxel in hepatocytes and dorsal root ganglion neurons, the site of injury within the nervous system, is mediated by the organic anion transporting polypeptides OATP1B1 and OATP1B3 (Oatp1b2 in rodents), transporters that are highly sensitive to pharmacological inhibition. To facilitate future screens of chemical libraries to identify modulators and imaging-based drug distribution st

Critical review on disposition of chlorinated paraffins in animals and humans.

Even though the chlorinated paraffins (CPs) have been on the environmental pollution agenda throughout the last 50 years it is a class of chemicals that only now is discussed in terms of an emerging issue with extensive annual publication rates. Major reviews on CPs have been produced, but a deeper understanding of the chemical fate of CPs, including formation of metabolites in animals and humans, is still missing. Thus, the present review aims to critically compile our present knowledge on the disposition, i.e. Adsorption, Disposition, Metabolism, and Excretion (ADME) of CPs in biota and to i
