Yersinia pestis dissemination in a host is usually studied by enumerating bacteria in the tissues of animals sacrificed at different times. This laborious methodology gives only snapshots of the infection, as the infectious process is not synchronized. In this work we used in vivo bioluminescence imaging (BLI) to follow Y. pestis dissemination during bubonic plague. We first demonstrated that Y. pestis CO92 transformed with pGEN-luxCDABE stably emitted bioluminescence in vitro and in vivo, while retaining full virulence. The light produced from live animals allowed to delineate the infected or

Nitrogen supply in the rhizosphere affects cadmium (Cd) accumulation in rice. To study bacterial nitrogen transformation effects on Cd uptake in rice, Escherichia coli MG1655 overexpressing nrfA, which encodes cytochrome c nitrite reductase involved in dissimilatory nitrite reduction to ammonium (DNRA), was constructed using a Tn5 transposon system. The nrfA genomic copy number in the engineered strain was approximately 3-fold that in wild-type, and RNA-seq analysis confirmed significantly increased nrfA transcript. Transcriptomics showed enrichment of ribosomes, oxidative phosphorylation, and

Insecticide and fungicide seed coatings have become prevalent in conventional agriculture in recent decades. From 2015-2021, the AltEn bioenergy plant (Mead, Nebraska, USA) generated ethanol from almost 100% unused/expired treated corn seeds. This use of these seeds for ethanol production resulted in the accumulation of large amounts of contaminated wastewater and solid residue, a portion of which was applied to surrounding farmland. To better understand the potential long-term environmental effects from the processing of treated seeds at this facility, five nearby stream sites were sampled in

Antibiotics are extensively used in mariculture, leading to their persistent accumulation in sediments and posing ecological and health risks. Sediments act as both major sinks and active sites for antibiotic transformation, yet the mechanisms remain poorly understood. This study examined the roles of sediment adsorption and biodegradation in removal of enrofloxacin (ENR), oxytetracycline (OTC), and sulfamethoxazole (SMZ) commonly used in mariculture. ENR and OTC were primarily removed via adsorption (88 % and 86 %), whereas SMZ removal involved both adsorption and biodegradation (54 % and 56

The overuse and misuse of antibiotics have led to their increasing release into aquatic ecosystems, promoting the selection and spread of antibiotic-resistant bacteria and potentially pathogenic microorganisms. In this study, biochars derived from sewage sludge and spent coffee grounds were produced, functionalized with iron, and coupled with H2O2 for the treatment of real municipal wastewater containing levofloxacin, doxycycline, and ampicillin. Response Surface Methodology identified sewage sludge biochar produced at 650 °C as the most effective material, and the optimized heterogeneous oxid

Virus-encoded auxiliary metabolic genes (AMGs) are important for host metabolism, such as carbon and nitrogen cycling; however, whether and how viral AMGs affect bacterial adaptation to pollution stress in long-term contaminated groundwater remain unclear, significantly limiting our understanding of phage-host interactions on the transformation of organic pollutants in groundwater. Here, by leveraging viromics sequencing for virus-like particles (VLPs) and metagenomic sequencing for host-cell, we profiled comprehensive virome datasets (recovered 16,672 viral contigs) from sites contaminated fo

BACKGROUND: Acid-tolerant enteric pathogens can evade small intestinal acid barriers, colonize and infect the intestinal tract. However, broad-spectrum antibiotics are not the best therapeutic strategy because of the disruption of intestinal flora caused by its indiscriminate antimicrobial activity against beneficial and harmful bacteria. So that is what inspired us to combine pH regulation with nanotechnology to develop a pH-triggered site-targeted antimicrobial peptide with entrapping function.
RESULTS: A pH-triggered dual biological functional self-assembled peptide (SAP) was designed acco

UNLABELLED: Biodegradation of dichloromethane (DCM) under reducing conditions is of major concern due to its widespread detection in contaminated groundwaters. Here, we report an anaerobic enrichment culture derived from a membrane bioreactor operating in an industrial wastewater treatment plant, capable of fermenting DCM and the brominated analogue dibromomethane (DBM). Comparative analysis of bacterial 16S rDNA-DGGE profiles from fresh liquid medium inoculated with single colonies picked from serial dilution-to-extinction agar vials showed that cultures degrading DCM contained a predominant

Chlamydia trachomatis is an obligate intracellular pathogen that replicates in a vacuole termed the inclusion. Many of the interactions of chlamydiae with the host cell are dependent upon bacterial protein synthesis and presumably exposure of these proteins to the cytosol. Because of the dearth of genetic tools for chlamydiae, previous studies examining secreted proteins required the use of heterologous bacterial systems. Recent advances in genetic manipulation of chlamydia now allow for transformation of the bacteria with plasmids. We describe here a shuttle vector system, pBOMB4, that permit

Fe deficiency in women contributes significantly to maternal and child morbidity in India. The intestinal bacterial flora may facilitate absorption of Fe from the caecum and proximal colon. The present study investigated the possibility that intestinal microbiota of anaemic young women may differ from that of women with normal Hb levels. The microbiota was quantified by real-time PCR in faeces of eight anaemic (Hb ≤ 100 g/l) and twenty-six normohaemic (Hb ≥ 120 g/l) women aged 18-25 years. Sequences of 16S ribosomal DNA (rDNA) specific to Bifidobacterium genus, Lactobacillus acidophilus group,

OBJECTIVE: enthesitis-related arthritis subtype of juvenile idiopathic arthritis (JIA-ERA) clinically resembles reactive arthritis (ReA). In patients with ReA the immune response is targeted at the outer membrane protein (OMP) of Salmonella typhimurium. We studied the immune response in JIA-ERA to S. typhimurium OMP.
METHODS: synovial fluid mononuclear cells (SFMC) and peripheral blood mononuclear cells (PBMC) were isolated from blood and SF of patients with JIA-ERA. Lymphocyte transformation test was done with S. typhimurium OMP and crude bacterial lysates of Yersinia enterocolitica, Shigell

Persistent oxidative inflammation complicates the treatment of multidrug-resistant (MDR) bacterial infections. Sonodynamic therapy (SDT) enables ultrasound-activated bacterial killing but does not directly alleviate infection-associated oxidative stress. Here, phenylboronic acid (PBA)-functionalized carbon dots (PS-CDs) were prepared from silkworm excrement extract and PBA-terminated poly(ethylene glycol) (PBA-PEG-PBA) through a one-pot hydrothermal process. The PS-CDs showed spectroscopic features consistent with partially retained chlorophyll-derived chromophores, while PBA-PEG-derived group

The contribution of bacterial infection to chronic low back pain and its treatment with antibiotics have generated considerable controversy in literature. If efficacious, antibiotics have the potential to transform the treatment of chronic low back pain in a significant subset of patients. Some microbiology studies of disc tissue from patients with CLBP have shown that bacteria are present, most likely due to infection, while others conclude they are absent or if found, it is due to surgical contamination. Clinical studies testing the efficacy of oral antibiotics to treat CLBP have either show

Deinococcus radiodurans exhibits remarkable survival under extreme conditions, including ionizing radiation, desiccation, and various DNA-damaging agents. It employs unique repair mechanisms, such as single-strand annealing (SSA) and extended synthesis-dependent strand annealing (ESDSA), to efficiently restore damaged genome. In this study, we investigate the role of the natural transformation-specific protein DprA in DNA repair pathways following acute gamma radiation exposure. Our findings demonstrate that the absence of DprA leads to rapid repair of gamma radiation-induced DNA double-strand

To advance the scientific understanding of bacteria-driven mercury (Hg) transformation processes in natural environments, thermodynamics and kinetics of divalent mercury Hg(II) chemical speciation need to be understood. Based on Hg LIII-edge extended X-ray absorption fine structure (EXAFS) spectroscopic information, combined with competitive ligand exchange (CLE) experiments, we determined Hg(II) structures and thermodynamic constants for Hg(II) complexes formed with thiol functional groups in bacterial cell membranes of two extensively studied Hg(II) methylating bacteria: Geobacter sulfurredu

In order to explore immobilization characteristic and its possible mechanism of various feedstocks-based biochars, deriving from chicken manure (CMB), rice straw (RSB) and sewage sludge (SSB), the effect of three biochars on speciation transformation of Cd in soil was compared, and soil properties, microbial community diversity were also examined. The treatment of CMB and RSB reduced significantly the content of acid-extractable Cd by 53.62% and 41.18%, respectively, but this available fraction was increased by 23.44% after SSB treatment. Simultaneously, CMB and RSB treatment significantly inc

Recent advances in microscopy techniques has uncovered unique aspects of flagella-driven motility in bacteria. A remarkable example is the discovery of flagellar wrapping, a phenomenon whereby a bacterium wraps its flagellum (or flagellar bundle) around its cell body and propels itself like a corkscrew, enabling locomotion in highly viscous or confined environments. For certain bacterial species, this flagellar-wrapping mode is crucial for establishing selective symbiotic relationships with their hosts. The transformation of a flagellum from an extended to a folded (wrapped) state is triggered

Chronic infection with Helicobacter pylori is the leading environmental cause of gastric carcinogenesis, yet the molecular pathways remain incompletely defined. This review links H. pylori-derived outer membrane vesicles (OMVs) and host epithelial exosomes through their shared cargo of heat shock protein 60 (GroEL/Hsp60). We proposed the concept of the "muco-microbiotic layer" as a fifth, functionally distinct layer of the gastric wall, where bacterial and host extracellular vesicles (EVs) interact within the mucus-microbiota interface. In this compartment, OMVs carrying bacterial GroEL and ex

Anaerobic digestion is a complex microbial process that mediates the transformation of organic waste into biogas. The performance and stability of anaerobic digesters relies on the structure and function of the microbial community. In this study, we asked whether the deterministic effect of wastewater composition outweighs the effect of reactor configuration on the structure and dynamics of anaerobic digester archaeal and bacterial communities. Biotic and abiotic factors acting on microbial community assembly in two parallel anaerobic digestion systems, an upflow anaerobic sludge blanket diges

Glaesserella parasuis is the pathogen that causes Glässer's disease in pigs, which is characterized by fibrinous polyserositis, arthritis and meningitis. Research on ribosomal protein L32 in microorganisms has mainly focused on regulating gene transcription and translation, but its effect on bacterial virulence is unclear. The role of L32 gene in G. parasuis is not clear, and in order to study the function of L32 gene, a suicide plasmid-mediated natural transformation method was used to construct a L32 gene deletion mutant. We found that although L32 was shown to be non-essential for cell prol
