Neuroligin-2 is ubiquitinated by Nedd4l to control developmental astrocyte morphogenesis.

Central nervous system astrocytes have an intricate, highly branched morphology. Proper development of perisynaptic astrocyte processes is n … rocyte morphogenesis. Biochemical assays revealed Nedd4l ubiquitinates and stabilizes NL2, and this ubiquitination is required for astrocyte morphogenesis. This study shows that NLs have nonoverlapping roles in controlling astrocyte growth and uncovers a molecular mechanism of how NL2 mediates astrocyte morphogenesis.

The ubiquitin system in normal and infected germinal center B cells.

The ubiquitin system is a highly regulated cellular mechanism that plays a central role in germinal center (GC) B cells, essential for protective humoral immunity and the site of gammaherpesvirus latency establishment. GC B cells undergo extraordinary levels of proliferation, DNA damage, and apoptosis prior to differentiating to long-lived memory or plasma cells, and ubiquitin pathways underpin these activities. Oncogenic gammaherpesviruses infect B cells and drive them through a GC activation program. Viral programs exploit ubiquitin biology to establish latency in GC B cells and persist for

Photoreactive crosslinkers capture the ubiquitin-E2-E3 ligases intermediates.

E3 ligases catalyze the final step in transferring ubiquitin to protein substrates, but resulting intermediates are characterized by conformational flexibility. Recent work by Chandler and colleagues ( Biochem J. (2026) 483, 1115-1130) demonstrates that photocross-linking activity-based probes can trap transient conformations of the intermediates, providing a valuable tool to map interfaces during the ubiquitin transfer process.

Viral hijacking of the host ubiquitin-proteasome system for immune escape.

E3 ubiquitin ligases regulate protein turnover and signaling by mediating degradative or nondegradative types of ubiquitylation. In the 20 August 2026 issue of Science, Glassman et al. identified mechanisms by which viral proteins commandeer the ubiquitin-proteasome system to dysregulate innate immunity, unveiling potential avenues for designing antiviral interventions.

Quinazolinone and Phthalazinone Inhibitors of the HDAC6/Ubiquitin Protein-Protein Interaction.

Histone deacetylase 6 (HDAC6) is a class IIb histone deacetylase that regulates diverse cytosolic acetylation through its two catalytic deacetylase domains and a C-terminal zinc finger ubiquitin-binding domain (ZnF-UBD). This ZnF-UBD mediates key protein-protein interactions (PPIs) that couple deacetylation and ubiquitin-dependent degradation. While most HDAC6 inhibitors target the catalytic domains, the ZnF-UBD represents an underexplored target. Here, we validate previously reported small-molecule inhibitors of the HDAC6 ZnF-UBD/ubiquitin interaction and describe novel N-alkyl moieties based

Ubiquitin Ligases in pro-atrophic and antiatrophic signaling cascades in muscles.

Skeletal muscle (SkM) atrophy is an associated disorder of cachexia, sarcopenia, immobilization, and denervation and is responsible for increased mortality and morbidity. SkM atrophy is often characterized by increased protein degradation and decreased protein synthesis in skeletal muscle. Increased protein catabolism is firmly associated with protein ubiquitination, an associated post-transcriptional modification of proteins that mediate diverse cellular functions like cell growth, cell death, DNA damage repair, and protein degradation. During the SkM atrophy, the extents of ubiquitination de…

Ubiquitin-proteasome system regulation of bone remodeling in postmenopausal osteoporosis.

Postmenopausal osteoporosis (PMOP) is associated with declining estrogen levels, and this hormonal deficiency alters the physiological balance between bone formation and resorption. Substantial evidence indicates that protein homeostasis disorders play a significant role in the pathological process of PMOP. As the core system regulating protein homeostasis, the ubiquitin-proteasome system exerts crucial regulatory effects on bone metabolism under estrogen-deficient conditions. Abnormal activity of these ubiquitin-associated enzymes often leads to excessive degradation or abnormal accumulation

Ubiquitin ligase Nedd4 regulates the abundance and toxicity of mutant huntingtin.

Huntington's disease (HD) is a neurodegenerative disorder caused by the expansion of CAG repeats in the gene encoding huntingtin. Since accumulation of mutant huntingtin (mHtt) leads to dysfunction of numerous cellular pathways and toxicity, reducing levels of the mutant protein represents a key therapeutic objective in HD. We found that ubiquitination of mHtt by E3 ubiquitin ligase Nedd4 promotes clearance of the mutant protein. Knockdown of Nedd4 increased toxicity of mHtt in mouse primary neurons and in a fly model of HD, suggesting the protective role of Nedd4. Importantly, levels of Nedd4…

Breaking the proteasome balance: α-synuclein and the ubiquitin-proteasome system.

Aging is the major risk factor for synucleinopathies, including Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. However, the molecular mechanisms linking aging to. α-Synuclein cytotoxicity remain incompletely understood. The progressive decline of proteostasis is central among these mechanisms, as the ubiquitin-proteasome system (UPS) and the autophagy-lysosome pathway fail to maintain the turnover of aggregation-prone proteins. In this review, we focus on the bidirectional crosstalk between α-Synuclein and the proteasome as a key driver of proteostasis collapse in

Midnolin: A ubiquitin-independent proteasome adapter in development, neurodegeneration, and cancer.

A seminal 2023 study uncovered a groundbreaking function of midnolin (MIDN): it acts as a novel proteasome adapter that directly targets nuclear transcription factors via its unique "capture-ubiquitin-α-helix-C-terminal (CUHC)" domain, thereby defining a novel midnolin-proteasome pathway. Before this pivotal discovery, midnolin was only known for its cellular localization and associations with nervous system, cancer, and cell differentiation, remaining a largely functionally uncharacterized protein in mammals. Importantly, this newly identified pathway plays indispensable roles in neurodevelop

Phosphorylated Ubiquitin as a Clinical Biomarker for Mitochondrial Damage in Neurodegenerative Diseases.

Phosphorylated ubiquitin (pS65-Ub) is generated by the kinase-ligase pair PINK1-Parkin to selectively label damaged mitochondria for degradation via the autophagy-lysosome system (mitophagy). Consistent with increasing mitochondrial and lysosomal dysfunctions, pS65-Ub accumulates with aging in human autopsy brain and in mice. pS65-Ub levels are strongly and independently elevated in brains from subjects with Alzheimer's or Parkinson's disease compared to age-matched, neurologically normal controls. Furthermore, pS65-Ub levels have been used to identify disease risk and potential resilience fac

Ubiquitin-proteasome pathway activation in the diaphragm of humans with reflux esophagitis.

Some forms of gastroesophageal reflux disease (GERD) are associated with crural diaphragm (CD) dysfunction, suggesting that GERD may be influenced by skeletal muscle deficiencies. Skeletal muscle atrophy has been strongly linked to alterations in the ubiquitin-proteasome system, the primary pathway for protein degradation. This study aimed to assess the expression of muscle atrophy-related proteins in the CD of patients with reflux esophagitis compared with those without esophagitis. In addition, we examined the correlation between these proteins, esophagitis severity, and esophageal acid expo

Ubiquitin E3 ligases in the plant Arg/N-degron pathway.

Regulation of protein longevity via the ubiquitin (Ub) - proteasome pathway is fundamental to eukaryotic biology. Ubiquitin E3 ligases (E3s) interact with substrate proteins and provide specificity to the pathway. A small subset of E3s bind to specific exposed N-termini (N-degrons) and promote the ubiquitination of the bound protein. Collectively these E3s, and other N-degron binding proteins, are known as N-recognins. There is considerable functional divergence between fungi, animal, and plant N-recognins. In plants, at least three proteins (PRT1, PRT6, and BIG) participate in the Arg/N-degro…

Ubiquitin-Mediated Effects on Oncogenesis during EBV and KSHV Infection.

The Herpesviridae include the Epstein-Barr Virus (EBV) and the Kaposi Sarcoma-associated Herpesvirus (KSHV), both of which are oncogenic gamma-herpesviruses. These viruses manipulate host cellular mechanisms, including through ubiquitin-mediated pathways, to promote viral replication and oncogenesis. Ubiquitin, a regulatory protein which tags substrates for degradation or alters their function, is manipulated by both EBV and KSHV to facilitate viral persistence and cancer development. EBV infects approximately 90% of the global population and is implicated in malignancies including Burkitt lym

Cross-regulation of Listeria monocytogenes and the host ubiquitin system in listeriosis.

The facultative intracellular bacterium Listeria (L.) monocytogenes may cause severe diseases in humans and animals. The control of listeriosis/L. monocytogenes requires the concerted action of cells of the innate and adaptive immune systems. In this regard, cell-intrinsic immunity of infected cells, activated by the immune responses, is crucial for the control and elimination intracellular L. monocytogenes. Both the immune response against L. monocytogenes and cell intrinsic pathogen control are critically regulated by post-translational modifications exerted by the host ubiquitin system and
