Prazosin for vasodilator treatment of acute pulmonary oedema due to scorpion sting.

Prazosin hydrochloride, a post-synaptic alpha receptor blocking agent, was administered to twelve patients with acute pulmonary oedema due to scorpion sting in order to lower their mean blood pressure (diastolic blood pressure plus one-third of pulse pressure) 10 to 20 mmHg. Following an oral dose (0.125 mg in children and 0.5 mg in adults), clinical improvement was seen in the form of disappearance of apical systolic murmur and pulmonary oedema, a reduction in heart rate observed at the end of the first hour, and maximal improvement seen at the end of the third hour. Chest radiographs showed

Hemodynamic properties of bucindolol, a beta-adrenoreceptor blocking drug with vasodilator activity.

Bucindolol is a new nonselective beta-adrenergic blocking agent with direct vasodilator, partial agonist and alpha-adrenergic blocking properties. Its hemodynamic effects were evaluated in 13 healthy young men. In an open study, the effects of increasing single oral doses (50 to 200 mg) were examined. Two hours after each dose there was little effect at rest, but the responses of heart rate and systolic blood pressure to cycle exercise (50 to 150 W) were significantly reduced. There was no greater effect with larger doses. The second phase was a 3-way double-blind study comparing the effects o

Regional vasodilating and cardiac effects of nicardipine in anaesthetized open chest dog.

In this study the effects of intravenous nicardipine on the cardiovascular system in anaesthetized open chest dogs were assessed. In the dose range of 3-30 nmol/kg it induced a dose-dependent and long lasting reduction of the vertebral, coronary and femoral vascular resistance. Nicardipine proved to be equipotent, but longer lasting than nifedipine, and about 10 and 100 times more potent than verapamil and papaverine respectively. Unlike papaverine, calcium entry blockers caused a preferential vasodilation of the vertebral and coronary arteries. Hypotension and a reduction of LVSP and dP/dt ap…

[Intravenous nitroglycerin as vasodilator therapy in acute myocardial infarction (author's transl)].

Thirteen patients with left ventricular failure complicating acute myocardial infarction were treated with intravenous nitroglycerin. The hemodynamic criterium for admission to this study was a pulmonary capillary wedge pressure greater than 15 mm Hg. All patients received intravenous nitroglycerin at infusion rates sufficient to lower pulmonary capillary wedge pressure by an average of 15 mm Hg, corresponding to the optimal left ventricular filling pressure in acute myocardial infarction. During this study, heart rate did not vary significantly; pulmonary capillary wedge pressure decreased fr

Chemical control of cerebral circulation. Modification by a new vasodilator (YC-93).

This study was undertaken to investigate influences of a new vasodilator, YC-93, a derivative of 1,4-dihydropyridine, on cerebral circulation and cerebrovascular CO2 reactivity. Cerebrocortical PO2, cerebrocortical PCO2, cerebrocortical blood flow and arterial blood pressure were continuously recorded by means of a PO2 electrode, a PCO2 electrode and a plate-type crossed thermocouple flowmeter placed on an exposed pial surface of the cat brain. The changes in each parameter induced by YC-93 were compared with those induced by papaverine hydrochloride. The effects of 5% CO2 inhalation were comp

Sympathetic and parasympathetic components of reflex cardiostimulation during vasodilator treatment of hypertension.

1. Haemodynamic responses to diazoxide (300 mg intravenously) were studied in 15 hypertensive patients before and after chronic beta-adrenoreceptor blockade by 320 mg of propranolol daily. After diazoxide alone, mean arterial pressure and total peripheral resistance were lowered by 24 +/- 3 and 35 +/- 5% (mean +/- SEM) respectively. Cardiac output and heart rate rose by 25 +/- 9 and 21 +/- 3%. During beta-adrenoreceptor blockade, the percentage changes of mean arterial pressure, heart rate, cardiac output and total peripheral resistance after vasodilatation were not significantly different fro

Hemodynamic effects of vasodilator agents in dogs with experimental ventricular septal defects.

The ratio of pulmonary to systemic vascular resistance (Rp/Rs) largely determines the amount of left-to-right shunting and pulmonary to systemic flow rat (Qp/Qs) in the presence of a large isolated ventricular septal defect. The possibility that pharmacologic reduction of systemic vascular resistance with alpha-adrenergic receptor blockade or beta-adrenergic receptor stimulation would increase the ratio Rp/Rs, and therefore reduce the ratio Qp/Qs, was studied in dogs in which ventricular septal defects had been surgically created. Administration of phentolamine and phenoxybenzamine caused a 42

Insulin augments vasodilatory response elicited by amlodipine via nitric oxide-dependent vasodilation in isolated rat aortas.

BACKGROUND: High-dose insulin and euglycemic therapy are widely used to treat calcium channel blocker toxicity. However, the effect of insulin on vasodilation evoked by the dihydropyridine calcium channel blocker amlodipine remains unknown. This study examined the effect of insulin on amlodipine-induced vasodilation in isolated rat aortas with specific emphasis on mechanisms associated with nitric oxide (NO). METHODS: The study assessed the roles of NW-nitro-L-arginine methyl ester (L-NAME), a nitric oxide synthase inhibitor; methylene blue, a general guanylate cyclase suppressor; 1H-[1,2,4]o…

Cholinergic nerve contribution to cutaneous active vasodilation during exercise is similar to whole body passive heating.

Sympathetic cholinergic nerve cotransmission is widely accepted as the mechanism of cutaneous active vasodilation (CAVD) during whole body passive heating (passive heating). However, recent research suggests that there may be mechanistic differences in CAVD to heating, depending on the modality of thermal loading. It is unknown whether sympathetic cholinergic cotransmission explains CAVD during exercise. This study sought to confirm the role of cholinergic nerves in CAVD during passive heating and expand these findings to exercise. It was hypothesized that CAVD during both exercise and passive…

The effect of temperature on vascular smooth muscle: cooling-induced vasodilation in deep arteries and veins.

Blood vessels in the cardiovascular system include arteries and veins, which are responsible for moving blood to and from tissues around the body. Our previous studies showed that cooling induces relaxation of arteries. The aim of this study is to investigate the effect of cooling on both arteries and veins pairs. Isometric tension was recorded in rat artery ring preparations (aorta, carotid, pulmonary arteries) and their vein pairs (vena cava, jugular, pulmonary veins) in organ baths during stepwise cooling from 37 to 4 °C. Cooling responses were tested before and after the addition of variou

Mitochondrial DNA integrity and function are critical for endothelium-dependent vasodilation in rats with metabolic syndrome.

Endothelial dysfunction in diabetes is generally attributed to oxidative stress, but this view is challenged by observations showing antioxidants do not eliminate diabetic vasculopathy. As an alternative to oxidative stress-induced dysfunction, we interrogated if impaired mitochondrial function in endothelial cells is central to endothelial dysfunction in the metabolic syndrome. We observed reduced coronary arteriolar vasodilation to the endothelium-dependent dilator, acetylcholine (Ach), in Zucker Obese Fatty rats (ZOF, 34 ± 15% [mean ± standard deviation] 10-3 M) compared to Zucker Lean rats…

Characterisation of P2Y receptor subtypes mediating vasodilation and vasoconstriction of rat pulmonary artery using selective antagonists.

Pulmonary vascular tone is modulated by nucleotides, but which P2 receptors mediate these actions is largely unclear. The aim of this study, therefore, was to use subtype-selective antagonists to determine the roles of individual P2Y receptor subtypes in nucleotide-evoked pulmonary vasodilation and vasoconstriction. Isometric tension was recorded from rat intrapulmonary artery rings (i.d. 200-500 µm) mounted on a wire myograph. Nucleotides evoked concentration- and endothelium-dependent vasodilation of precontracted tissues, but the concentration-response curves were shallow and did not reach …
