Calcium gradient in plant cells with polarized growth in simulated microgravity.

Plant cells characterized by apical growth, for example, root hairs and apical cells of moss protonema, are a convenient model to address the problem of gravity response mechanisms including initiation of cell polarity. The fluorescent calcium probe, chlorotetracycline, allowed us to display the calcium distribution gradient in these cells. Irradiation by red light led to a sharp decrease in the Ca2+ ion activity in cells. During clinostatting in darkness the pattern of calcium influx and distribution changes inconsiderably as compared with control; in root hairs calcium is detected mainly in

Execution of "ARC" experiment on space shuttle "Discovery" STS 51-C: some results on aggregation of red blood cells under zero gravity.

A project on "Aggregation of Red Cells" has been accepted by NASA in 1977. An automated slit-capillary photo-viscometer has been designed during 1979-1984, and its last version met NASA's space hazards requirements. The 'heart' of instrument is a set of two highly polished glass plates, spaced by a gap of 12.5 micrometers. An original drum-like infusion pump allows utilization of up to eight blood samples. During a sequential process, blood flows through the slit, and then stops to allow formation of aggregates. Micro- and macro-photography is carried out, and 500 photographs are obtained. Blo

Etiological factors in space motion sickness.

We compared susceptibility to motion sickness during exposure to sudden-stop stimulation as a function of gravitoinertial force level. Our findings show that susceptibility is greatly enhanced, both with eyes-closed and eyes-open, for zero-g and 2-g conditions in parabolic flight compared with 1-g test conditions. The change in susceptibility is likely related to three factors: alterations in vestibulo-ocular function which result from variations in gravitoinertial force level (28,29); the altered pattern of otolithic activity resulting during variations in gravitoinertial force level; and the

[Bioelectric activity of the human brain during 180-day antiorthostatic hypokinesia and during the rehabilitation period].

Cerebral bioelectric activity of 18 healthy test subjects, aged 31-40, was examined during and after their 182-day head-down tilting. The test subjects were divided into a group that performed countermeasures (exercise, muscle stimulation) against hypokinesia-induced disorders and a control group. During the study EEG changes were most pronounced in the controls: alpha-rhythm amplitude, frequency and index decreased, slow wave frequency reduced, whereas index of zonal differences and amplitude of beta- and slow waves increased. Phasic changes in the central nervous system excitability in respo

Effect of lower body negative pressure against orthostatic intolerance induced by 21 days head-down tilt bed rest.

BACKGROUND: Exposure to actual or simulated weightlessness is known to induce orthostatic intolerance in humans. Many different methods have been suggested to counteract orthostatic hypotension. The repetitive or prolonged application of lower body negative pressure (LBNP) has shown beneficial effects to counter orthostatic intolerance, but devoting so much time to countermeasures is not compatible with space mission objectives or costs. The purpose of the present study was to assess the effects of brief LBNP sessions against orthostatic intolerance during a 21-d head-down tilt (HDT) bed rest.…

Space motion sickness medications: interference with biomedical parameters.

The possibility that drugs administered to Skylab 3 (SL-3) and 4 (SL-4) crewmen for space motion sickness may have interfered with their bio … sterone excretion during the first three days of spaceflight probably can be directly attributed to weightlessness, whereas the antimotion sickness medication could have substantially contributed to the early increased excretion of epinephrine and cortisol during these missions.

Daily use of countermeasures to prevent headward fluid shifts throughout 30 days of strict head-down tilt bed rest.

Strict 6° head-down tilt bed rest (HDTBR) induces a chronic headward fluid shift similar to weightlessness that may underlie the development of spaceflight-associated neuro-ocular syndrome. We tested the efficacy of two headward fluid shift countermeasures, using either 25 mmHg lower body negative pressure (LBNP, 2 × 3 h daily, n = 12) or use of venoconstrictive thigh cuffs (VTC) following 1 h of moderate aerobic cycle exercise (Ex + CUFF, 6 h daily for 6 of 7 days, n = 12) across 30 days of HDTBR. The study also included a seated posture group (Upright, 2 × 3 h daily, n = 11), which was condu…

Effects of Spaceflight and Simulated Microgravity on YAP1 Expression in Cardiovascular Progenitors: Implications for Cell-Based Repair.

Spaceflight alters many processes of the human body including cardiac function and cardiac progenitor cell behavior. The mechanism behind these changes remains largely unknown; however, simulated microgravity devices are making it easier for researchers to study the effects of microgravity. To study the changes that take place in cardiac progenitor cells in microgravity environments, adult cardiac progenitor cells were cultured aboard the International Space Station (ISS) as well as on a clinostat and examined for changes in Hippo signaling, a pathway known to regulate cardiac development. Sex-Based Differences in Intraocular Pressure During Head-Down Tilt in a Spaceflight Analog Model.

INTRODUCTION: Spaceflight-associated neuro-ocular syndrome is a high-priority research gap, especially regarding its mechanisms and individual susceptibility, including sex-based differences. This study investigates sex-specific intraocular pressure (IOP) responses during head-down tilt (HDT) to improve understanding of differential risk profiles and inform the development of targeted countermeasures. METHODS: A total of 15 healthy subjects (8 males, 7 females) completed a 60-min -12° HDT protocol. IOP was measured in a seated baseline condition and at 0, 30, and 60 min during HDT using a reb

Bone mineral loss damages renal tubules in mice.

Fibroblast growth factor-23 (FGF23) is a bone-derived hormone that promotes urinary phosphate excretion in response to phosphate loading. While essential for phosphate homeostasis, elevated FGF23 increases phosphate concentration in the renal tubular fluid, promoting calcium-phosphate crystal formation and tubular injury. Here we show that bone resorption mobilizes phosphate into the circulation and mimics the pathophysiology of dietary phosphate loading. Enhanced bone resorption, induced by soluble receptor activator of NF-κB ligand (sRANKL) administration or microgravity exposure on the Inte

Biological sex as a variable in immunity does not affect parabolic flight-induced alterations in immune responses.

The present era of spaceflight is accompanied by two meaningful breakthroughs. The access to in-orbit missions for civilians and the increasing enrolment of female astronauts require detailed investigations on the impact of gravitational stress on human physiology with focus on sex-specific differences. To assess the responsive capacities of innate and adaptive immunity in this context, functional characterizations were performed in women and men in a parabolic flight study. Blood and saliva were collected 1 month prior and on the day before the flight, as well as directly after flight and on

Aerobic Exercise and Subsequent Venoconstrictive Thigh Cuffs Preserve Cardiorespiratory Fitness but Not Muscle Function after 30 Days of Bed Rest.

PURPOSE: During spaceflight, countermeasures are crucial to counteract cardiorespiratory and musculoskeletal deconditioning, similar to effects seen after prolonged immobilization. Within a bed rest study simulating the effects of microgravity, we investigated the effectiveness of submaximal aerobic exercise and postexercise venoconstrictive thigh cuffs (VTC) for maintaining cardiorespiratory fitness and muscle function. METHODS: Twenty-four males and females were randomly allocated to a countermeasure ( n = 12, age = 34 ± 9 yr, body mass index = 22.9 ± 2.9 kg·m -2 ) or control group ( n = 12

Celastrol and Cblin peptide activation of IGF-1 signaling prevents microgravity-induced atrophy in rat L6 myotubes.

This study investigated the efficacy of two natural compounds-celastrol, a heat shock protein (HSP) inducer, and Cblin peptide, a ubiquitination inhibitor-in counteracting muscle atrophy under real microgravity conditions. Both agents independently attenuated microgravity-induced reductions in myotube thickness, myosin heavy chain protein levels, and atrogene expression. Celastrol primarily enhanced HSP expression, whereas Cblin peptide inhibited insulin receptor substrate-1 degradation, thereby promoting insulin-like growth factor-1 signaling. Despite their distinct molecular actions, no syne

Human cranial bone-derived mesenchymal stem cells cultured under simulated microgravity can improve cerebral infarction in rats.

The efficacy of transplanting human cranial bone-derived mesenchymal stem cells (hcMSCs) cultured under simulated microgravity (sMG) conditions has been previously reported; however, their effect on cerebral infarction remains unknown. Here, we examined the efficacy of transplanting hcMSCs cultured in an sMG environment into rat models of cerebral infarction. For evaluating neurological function, hcMSCs cultured in either a normal gravity (1G) or an sMG environment were transplanted in rats 1 day after inducing cerebral infarction. The expression of endogenous neurotrophic, axonal, neuronal, s

Simulated microgravity-induced oxidative stress and loss of osteogenic potential of osteoblasts can be prevented by protection of primary cilia.

Oxidative stress has been considered to be closely related to spaceflight-induced bone loss; however, mechanism is elusive and there are no effective countermeasures. Using cultured rat calvarial osteoblasts exposed to microgravity simulated by a random positioning machine, this study addressed the hypotheses that microgravity-induced shortening of primary cilia leads to oxidative stress and that primary cilium protection prevents oxidative stress and osteogenesis loss. Microgravity was found to induce oxidative stress (as represented by increased levels of reactive oxygen species (ROS) and ma

The effects of a spaceflight analog with elevated CO2 on sensorimotor adaptation.

Aboard the International Space Station (ISS), astronauts must adapt to altered vestibular and somatosensory inputs due to microgravity. Sensorimotor adaptation on Earth is often studied with a task that introduces visuomotor conflict. Retention of the adaptation process, known as savings, can be measured when subjects are exposed to the same adaptive task multiple times. It is unclear how adaptation demands found on the ISS might interfere with the ability to adapt to other sensory conflict at the same time. In the present study, we investigated the impact of 30 days' head-down tilt bed rest c

[Effect of vibrostimulation on support zones of rat's feet, and support loading on titin N2A-isoform and T2-fragment in m. soleus under the conditions of simulated microgravity].

Effects of daily 3-hr vibrostimulation of foot support zone and 3 hrs. of usual locomotion on titin N2A-isoform and proteolytic T2-fragment were studied in m. soleus of rats during 7-d suspension. Besides, effects of single one-hour support loading following 7-d weight-deprivation on titin N2A- and T2-forms were also evaluated in the muscle. Relative N2A content in m. soleus of the suspended group was noted to reduce approximately 25% as compared with the group of control. In the meantime, relative T2 content grew 2-3 times. Along with other changes in the muscle apparatus, N2A reduction contr
