Similarities in the physical and chemical properties of the phosphoribosyltransferase family of enzymes suggest that they may share common structural features as observed in other functionally related proteins. The unusually high incidence of structural gene mutations of these enzymes in man are associated with several metabolic diseases of purine and pyrimidine metabolism. It is proposed that these disorders are the consequence of structural mutations to an architectural domain common to all of the phosphoribosyltransferases.

This paper describes the transition from separate study-specific databases to a unified SQL Server-based system with a modular architecture and a web interface developed in Blazor Server. The new design enhances data interoperability, supports future studies, and simplifies clinician interaction, paving the way for future alignment with the OMOP Common Data Model.

Circulating tumor cell (CTC) clusters are highly efficient metastatic seeds in various cancers. Yet, their genetic heterogeneity and clonal architecture is poorly characterized. Using whole-exome sequencing coupled with phylogenetic inference from CTC clusters of patients with breast and prostate cancer, as well as mouse cancer models alongside barcode-mediated clonal tracking in vivo, we demonstrate oligoclonal composition of individual CTC clusters. These results improve our understanding of metastasis-relevant clonal dynamics.

The glomerular endothelium produces the key complement regulator factor H (FH), but its role in the endothelial cells protection and functional integrity is unclear. In this edition of Kidney International, Mahajan et al. demonstrate that the endothelial-intrinsic FH is important for the cytoskeletal architecture, monolayer integrity, proliferation control, metabolism, and inflammatory signaling regulation. These findings place the endothelium-derived FH in the center of the pathological process of diseases, characterized with FH genetic abnormalities.

This research argues that the spaces of skateboarding are hybrid. This is evidenced by not only the construction of numerous skateparks throughout the world, and their incorporation of design elements from the city, but the exclusion of skateboarders from urban spaces through hostile architecture. More specficially, skateparks, which are unique among sport facilities, are shown to be evolving hybrid places in a continuing dialogue with the city under which they are geographically and politically contained.

We describe a rare case of extralobar pulmonary sequestration with hemorrhagic infarction in a 10-year-old boy who presented with acute abdominal pain and fever. In our case, internal branching linear architecture, lack of enhancement in the peripheral portion of the lesion with internal hemorrhage, and vascular pedicle were well visualized on preoperative magnetic resonance imaging that led to successful preoperative diagnosis of extralobar pulmonary sequestration with hemorrhagic infarction probably due to torsion.

We present the case of 80-year-old male with superficial papillary urothelial carcinoma of the urinary bladder with striking multicystic architecture with a combination of features of urothelial carcinoma with gland-like lumina, with signet-ring cell differentiation and microcystic pattern. However, the tumor shared the morphologic features of several variants of urothelial carcinoma, the most important differential diagnosis covered so-called florid Brunneriosis, cystitis cystica, and primary adenocarcinomas of the urinary bladder.

A woman whose condition was diagnosed as agoraphobia with panic attacks participated in an all-night polysomnographic study. During the night she experienced a panic attack that was phenomenologically similar to her daytime attacks. The attack occurred suddenly, arising from stage 3-4 (delta) sleep. There was no disruption of sleep architecture immediately before awakening. The occurrence of this panic attack during sleep is discussed in the context of other nighttime sleep disorders.
