The first outbreak of equine coronavirus infection in 13 years among draft horses at Obihiro Racecourse in Japan in 2025.

Equine coronavirus (ECoV) causes fever, anorexia and sometimes enteric signs (e.g. diarrhea) in adult horses. Three ECoV outbreaks among draft horses at the same Japanese racecourse have been reported previously, in 2004, 2009 and 2012. We describe the epidemiological, clinical and genetic features of the fourth ECoV outbreak at the same racecourse in 2025. In January 2025, a sudden increase in the number of draft horses showing various combinations of fever, anorexia and enteric signs was observed at Obihiro Racecourse, in Hokkaido, Japan. Of about 800 horses, 176 showed clinical signs. Speci

Development and application of a dual RT-qRPA assay for rapid differentiating of goose astrovirus genotypes 1 and 2.

Goose astrovirus (GAstV) is an important pathogen causing joint and visceral gout in goslings. Its high incidence and mortality had caused enormous economic losses to the goose farming industry. To achieve the simultaneous and rapid identification and detection of Goose astrovirus genotype 1 (GAstV-1) and genotype 2 (GAstV-2), the present study aimed to design and synthesize specific recombinase polymerase amplification (RPA) primers and exo probes targeting the ORF1b gene of GAstV. Utilizing a portable fluorescence detector (Genie III), the RPA primers and exo probes were optimized for concen

Rickettsia detection in fleas associated with cricetid rodents in a periurban area of Argentina.

This study analysed the presence of Rickettsia (Rickettsiales: Rickettsiaceae) in fleas (Siphonaptera) associated with cricetid rodents in periurban localities of Gran La Plata, Argentina. Rodents were captured in three localities and fleas were collected directly from their fur. After DNA extraction, fleas were prepared for microscopic identification. PCR amplification of gltA and ompB genes was performed on each individual flea to detect and identify Rickettsia. The OmpB gene was sequenced and compared using nBLAST to initially identify its similarity with other sequences from GenBank. A phy

Broad geographical circulation of a novel vesiculovirus in bats in the Mediterranean region.

Bats are the natural reservoirs for a variety of emerging and re-emerging viruses. Among them, rabies virus (genus Lyssavirus, family Rhabdoviridae) is one of the first and most emblematic described in these animals. Since its first description, several new bat lyssaviruses have been regularly identified. In addition to lyssaviruses, other bat rhabdoviruses have also been discovered, including members of the genera Vesiculovirus, Ledantevirus and, more recently, Alphanemrhavirus and Tupavirus. However, the family Rhabdoviridae is one of the most abundant and diverse viral families, with 434 of

Clinical evaluation of a sensitive fluorescent PCR assay for trichomonas vaginalis.

Trichomoniasis, caused by Trichomonas vaginalis (T. vaginalis), is a common sexually transmitted disease (STD) that primarily affects the genital and urinary tracts. Trichomoniasis has a worldwide distribution and is also widely endemic in China. In recent years, due to the continuous increase in the proportion of recurrent or persistent vaginal trichomoniasis, the related clinical diagnosis and treatments are facing new challenges. Currently, clinical diagnosis relies on clinical features and laboratory tests. Microscopic examination of vaginal secretions is a commonly used diagnostic method

Diagnostic Accuracy of Multiplex NAAT/PCR and Culture Against Salmonella spp.: A Comparison of Meta-Analytical Methods.

BACKGROUND: Non-typhoidal (NT) Salmonella spp. constitutes a major cause of foodborne illness. Culture is the gold standard, but it is time consuming, whereas multiplex nucleic acid amplification tests (NAATs)/Polymerase Chain Reaction (PCR) offer faster detection with variable reported performance. OBJECTIVES: To compare the diagnostic accuracy of multiplex NAAT/PCR and culture for Salmonella spp. using various statistical models with or without a gold standard assumption. METHODS: A systematic search (PubMed, Web of Science, Scopus; up to April 2024) identified 44 studies (55 comparisons).

Release of Angiostrongylus cantonensis larvae from live intermediate hosts under stress.

The metastrongyloid nematode Angiostrongylus cantonensis causes eosinophilic meningitis in a variety of homeothermic hosts including humans. Third-stage infectious larvae develop in gastropods as intermediate hosts. Humans are usually infected by intentional or incidental ingestion of an infected mollusk or paratenic host (poikilothermic vertebrates and invertebrates). The infection may also hypothetically occur through ingestion of food or water contaminated by third-stage larvae spontaneously released from gastropods. Larvae are thought to be released in greater numbers from the intermediate

Implementation of pooled saliva tests for universal screening of cCMV infection.

Congenital cytomegalovirus (cCMV) is the most common intrauterine infection, leading to neurodevelopmental disabilities. Universal newborn infant screening of cCMV has been increasingly advocated. In the absence of a high-throughput screening test, which can identify all infected newborn infants, the development of an accurate and efficient testing strategy has remained an ongoing challenge. Here we assessed the implementation of pooled saliva polymerase chain reaction (PCR) tests for universal screening of cCMV, in two hospitals of Jerusalem from April 2022 through April 2023. During the 13-m

Rapid detection of pigeon adenovirus 2 using a TaqMan real-time PCR assay.

Pigeons infected with aviadenoviruses have been found worldwide. Recently, pigeon adenovirus 2 (PiAdV-2) has been widely distributed in racing pigeons in Germany. However, the epidemiology of this virus remains unclear due to the lack of a specific detection platform for PiAdV-2. In this study, we first detected PiAdV-2 positivity in racing pigeons (designated FJ21125 and FJ21128, which share 100% nucleotide identity with each other based on the fiber 2 gene) in Fujian, Southeast China. These genes shared 99.8% nucleotide identity with PiAdV-2 (GenBank No. NC_031501) but only 54.1% nucleotide

Development and validation of a quantitative PCR assay for detection of Sulawesi tortoise adenovirus.

In 2007, a mortality event involving over 100 Sulawesi tortoises (Indotestudo forsteni), two Impressed tortoises (Manouria impress) and a critically endangered Burmese star tortoise (Geochelone platynota) was attributed to Sulawesi tortoise adenovirus (STADV; genus Siadenovirus). We developed a TaqMan quantitative PCR assay targeting the DNA polymerase gene of STADV for use in clinical diagnosis and epidemiologic surveillance. This assay failed to amplify five closely-related chelonian adenoviruses, indicating high analytical specificity. The assay performed with high efficiency (slope = -3.33

Detection and differentiation of fowl adenovirus serotype 4 and duck adenovirus 3 using high resolution melting curve assay.

Fowl adenovirus type 4 (FAdV-4) and duck adenovirus type 3 (DAdV-3) are the causative agents of clinical diseases in poultry and have caused considerable economic losses to the waterfowl industry in China. Both FAdV-4 and DAdV-3 are classified into the genus Aviadenovirus under the family Adenoviridae. The high-resolution melting (HRM) assay has become a useful method for virus genotyping, which offers the possibility of rapidly developing a differentiation technique in which the melting profile depends on the GC content of the product in the qPCR platform. The aim of this study was to develop

Molecular detection of Usutu virus in pools of Culex pipiens mosquitoes in Greece.

Usutu virus (USUV) is a mosquito-borne flavivirus originating from Africa, that belongs to the Japanese encephalitis virus (JEV) complex. In nature, USUV involves Culex spp. mosquitoes acting as vectors and birds as amplifying hosts. The virus has recently spread in Europe and is considered an emerging human pathogen. This is the first research study performed in Greece revealing the presence and circulation of USUV in Culex spp. mosquito populations. Out of the 1,500 mosquito pools tested with real-time RT-PCR, four (Roesch et al., 2019) were positive for USUV. All four pools were collected f
