BACKGROUND: Periostin is an extracellular matrix protein that participates in tissue remodeling and inflammation and may be related to cardiometabolic abnormalities. However, its association with metabolic and lipid disorders in newly diagnosed type 2 diabetes mellitus (T2DM) patients with dyslipidemia is poorly defined. This study evaluated serum periostin and its associations with glycemic, lipid, and atherogenic measures. We also performed exploratory molecular docking to investigate its predicted interactions with metformin and atorvastatin.
METHODS: This was a case-control study with 45

In vivo molecular imaging tools hold immense potential to drive transformative breakthroughs by enabling researchers to visualize cellular and molecular interactions in real-time and/or at high resolution. These advancements will facilitate a deeper understanding of fundamental biological processes and their dysregulation in disease states. Here, we develop and characterize a self-assembling protein nanomicelle called collagen type I binding - thermoresponsive assembled protein (Col1-TRAP) that binds tightly to type I collagen in vitro with nanomolar affinity. For ex vivo visualization, Col1-T

An in-depth literature review of up to 2023 reveals 330 risk loci found by genetic association at p ≤ 5 × 10-8, with systemic lupus erythematosus (SLE) in at least one study of 160 pertinent publications. There are 225 loci found in East Asian (EAS), 106 in European (EU), 11 in African-American (AA), 18 Mixed American (MA), and 1 in Egyptian ancestries. Unexpectedly, most of these associations are found to date at p ≤ 5 × 10-8 in a single ancestry. However, the EAS and EU share 40 risk loci that are independently established. The great majority of the identified loci [250 (75.8%) of 330] do no

The presence of HLA-DRB1 alleles that encode critical points associated with environmental interactions is associated with increased risk of rheumatoid arthritis caused by anti-citrullinated protein antibodies. Therefore, interleukin-6 (IL-6), a multifunctional cytokine that controls both local and systemic acute inflammatory responses through its ability to induce a phase response, plays a serious role. Its overexpression leads to pathological challenges such as rheumatoid arthritis and menopausal osteoporosis. However, targeting the IL-6 receptor and its region could be the major step in con

Fetal exposure to bisphenols and phthalates may influence development of the reproductive system. In a population-based, prospective cohort study of 1059 mother-child pairs, we examined the associations of maternal gestational urinary bisphenols and phthalates concentrations with offspring reproductive development from infancy until 13 years. We measured urinary bisphenol and phthalate concentrations in each trimester. We obtained information on cryptorchidism or hypospadias after birth from medical records. At 9.7 years, we measured testicular and ovarian volume by MRI. At 13.5 years, we meas

Non-Structural Protein 6 (NSP6) is a crucial protein for SARS-CoV-2 as it performs a vital role in the replication and transcription of the virus. NSP6 plays a role in the stress response of the endoplasmic reticulum through binding to the sigma receptor 1 (SR1). Therefore, NSP6 is an interesting target for fighting SARS-CoV-2. The best model of the tertiary structure of NSP6 was predicted using the AlphaFold server, then this model was refined using the DeepRefiner server to construct a good-quality model. The current study utilized the virtual screening of the ZINC20 database for the identif

In the pursuit of novel antioxidant and anti-inflammatory agents, we investigated 5-hydroxyferulic acid (5-OHFA), a hydroxylated derivative of the well-known phenolic compound ferulic acid (FA). This study aimed to determine whether structural modification enhances the biological activity of FA. To this end, both compounds were subjected to a series of in vitro antioxidant assays (DPPH, ABTS, FRAP, and Fe(II)-chelating) and anti-inflammatory evaluations, complemented by in silico predictions. 5-OHFA consistently exhibited superior antioxidant capacity, with significantly lower IC50 values than

CGA-N9 is an antifungal peptide that primarily targets Candida spp. with a mild activity. Our preceding research confirmed that the CGA-N9 crosses cell membrane with the assistance of C. tropicalis oligopeptide transporter (CtOPT) -1 and - 9. In this study, CGA-N9-derived peptides were designed following the molecular docking results with CtOPT-1 and -9. Compared with CGAN9, they exhibit higher transmembrane efficiency with the assistance of CtOPT-1 during the early phase of transmembrane processes and CtOPT-9 in the late phase. And they displayed significantly enhanced antifungal activity, wi

Neospora caninum (NC) is a protozoan infection causing neosporosis, a disease that leads to substantial economic loss in livestock, especially in cattle, sheep, and goats. Unfortunately, there is presently no viable vaccination or authorized therapy available. Our research applied a bioinformatics approach to design a multi-epitope peptide (MEP) vaccine aimed at the pathogenic proteins GRA2 and Nc-p43 of NC, which are critical in parasite-mediated antigenicity and host interactions. Consequently, our research employs an in-silico methodology, including protein sequence retrieval, epitope predi

Antibiotic resistance poses a global crisis fuelled by widespread antibiotic use, particularly against Gram-negative bacteria like Klebsiella pneumoniae, a leading cause of hospital-acquired infections with high mortality rates. Urgent identification of effective drug targets is imperative, with a focus on metabolic pathways to inhibit bacterial growth. Targeting the crucial metabolic pathways of K. pneumoniae would be a more efficient way to prevent its growth and the diseases that it causes. The present study focused on inhibiting the UDP-N-acetylglucosamine--N-acetylmuramyl-(pentapeptide)py

Japanese encephalitis (JE), a neurological infection of severe nature, is caused by the Japanese encephalitis virus (JEV) and is transmitted by the mosquito vector. The polymerase domain of Non-structural 5 (NS5), which is also referred to as RdRp (RNA-dependent RNA polymerase), is considered a potential therapeutic target for JEV. The present study employed molecular dynamics modelling and high-throughput virtual screening to evaluate the possible antiviral activity of anti-dengue drugs against JEV RdRp. Furthermore, a ranking was performed utilising the MM/GBSA analysis to identify the three
