Copper peroxide/manganese dioxide nanoparticles crosslinked hyaluronan hydrogel for enhanced chemodynamic/photodynamic/photothermal therapy in breast cancer.

Therapeutic hydrogels are becoming a promising option of cancer treatment. However, the simple construction of multifunctional hydrogels under physiological conditions is still a huge challenge. Herein, a chlorin e6 (Ce6)-embedded hyaluronan/CuO2/MnO2 nanocomposite hydrogel (Ce6@HHCM) was developed at physiological pH through an innovative mineralization-induced metal-hydrazide coordinative crosslinking technique. The hydrogel exhibited adjustable degradation rate, outstanding mechanical performance, excellent biosuitability, stimulus-responsive behavior, injectability, and self-healing functi

Biotinylated Self-Assembled Cytarabine Nano-Prodrug with Enhanced Targetability and Anticancer Efficacy: An In Vitro Study.

In this study, we developed a tumor-target biotinylated cytarabine (Ara-C) prodrug linked through a disulfide bond (Bio-SS-Ara). To enhance its delivery, Bio-SS-Ara was found to be able to self-assemble into uniform nanoparticles (BSSA NPs) with an average particle size of 230 nm, a PDI of 0.265, a critical micelle concentration (CMC) of 5.89 μg/mL, and a zeta potential of -15 mV. BSSA NPs showed good storage stability over 6 weeks. In a glutathione-rich environment, the drug release of BSSA NPs was more rapid than that of BCCA NPs. Concentration-dependent size analysis suggested that BSSA NPs

Elucidating the activation mechanism of botulinum neurotoxin a: role of α-clostripain and NTNH.

Botulinum neurotoxin A (BoNT/A) is one of the most toxic substances known. The bacterium Clostridium botulinum produces it as a 150-kDa polypeptide with relatively low potency. Surprisingly, the endogenous protease that transforms this polypeptide into an activated, full-potency toxin, consisting of a 100-kDa heavy chain connected to a 50-kDa light chain (LC) by a disulfide bond, is still unknown. This study aimed to identify the BoNT/A-activating protease. We screened cation-exchange chromatography fractions of C. botulinum A culture supernatant for activity using a toxin-simulating substrate

Insights into platelet factor 4-derived peptide macrocycles; the mechanistic basis of their rapid and selective antiplasmodial actions.

The malarial parasite Plasmodium can acquire resistance to most mainstay antimalarial drugs, necessitating the development of new antiplasmodial agents with different modes of action. The innate defense protein, human platelet factor 4 (PF4), has a unique antiplasmodial action that involves selective entry into Plasmodium-infected red blood cells (RBC) and subsequent destruction of the parasite's digestive vacuole (DV). This activity is recapitulated in PF4-derived internalization peptides (PDIPs). Here, we characterized the actions of PDIP analogs and PF4 in live P. falciparum-infected human

Human serum albumin-adduct biomarkers to prove human poisoning with methanethiol.

Exposure to methanethiol (MT) presents a significant challenge in public healthcare and could be a concern in the context of terrorist attacks. Therefore, reliable verification procedures for MT intoxication are essential for forensic, toxicological, and clinical purposes. We developed and validated a bioanalytical method for the simultaneous detection and identification of biomarkers indicative of MT exposure. Neat human serum albumin (HSA) and human plasma were incubated with MT to form adducts, which served as references. Subsequently, HSA and human plasma were subjected to proteolysis usin

ADME analysis, metabolic prediction, and molecular docking of lipoic acid with SARS-CoV-2 Omicron spike protein.

Lipoic acid (ALA), also known as 1,2-dithiolane-3-pentanoic acid, is a natural antioxidant and a critical component of mitochondrial function, where it participates in several key enzymatic processes. This organosulfur compound is derived from both plant and animal sources and was initially studied as a potential substitute for acetate. This study aims to examine the parameters related to ADME, metabolic, and structural binding analyses. Furthermore, a molecular docking analysis was performed to explore how ALA interacts with the Omicron variant of SARS-CoV-2. The study revealed that ALA exhib

Secreted protein acidic and rich in cysteine-guided biomimetic delivery of nano-antioxidants reverses muscle atrophy in a mouse model of sarcopenia.

Sarcopenia is currently classified as an unmet medical need with an increasing incidence because of population aging. Excessive reactive oxygen species (ROS) production plays a critical role in the pathogenesis of muscle atrophy, a key feature of sarcopenia, whereas edaravone has been identified as a potent ROS scavenger in screening assays using C2C12 myocytes. In this study, we established an injured muscle-targeted drug delivery system (imDDS), which mimics the secreted protein acidic and rich in cysteine (SPARC)-mediated albumin uptake pathway, to deliver edaravone to damaged muscle tissue

A smart ultra-small near infrared persistent luminescence nanoprobe for non-invasive detection of early rheumatoid arthritis.

Early detection of rheumatoid arthritis (E-RA) is crucial for long-term patient health, but current methods lack precision. Overproduced reactive oxygen species (ROS) in hypoxic and inflammatory synovium are early markers of RA, appearing years before significant radiological findings, making them attractive biomarkers for early detection. Here, an ultrasensitive ROS-responsive nanoprobe are developed based on glutathione (GSH) modified Cr3 + doped ZnGa2O4 ultra-small near-infrared (NIR) persistent luminescence nanoparticles (denoted as ZnGa2O4:Cr3+-GSH). Dual Targeting and Cascaded Aggregation of Luminogen for Washing-Free and Enhanced Fluorescence Imaging of Intracellular Bacteria.

The invasion and intracellular survival of Staphylococcus aureus (S. aureus) within phagocytic cells is a major cause of serious antibiotic resistance and recurrent infections in clinical settings, making the detection of intracellular S. aureus highly desirable prior to initiating effective treatment. In this study, we proposed a dual-targeting and cascaded aggregation-induced emission luminogen (AIEgen) probe for washing-free and enhanced fluorescence imaging of intracellular S. aureus. The AIEgen probes consist of an AIE molecule (TPE), cleavable peptide (YVADC(StBu)GKK), targeting peptide

Detection of reduced orexin-A/hypocretin-1 and its fragments by orexin-A "gold-standard" radioimmunoassay.

BACKGROUND: Orexin-A/hypocretin-1 level is determined in the cerebrospinal fluid samples as a part of clinical narcolepsy diagnostics utilizing a specific commercial radioimmunoassay (RIA); this assay is also widely used in research of many other conditions. The specificity of RIAs is in general variable, and little has been firmly disclosed about the specificity of this RIA assay. Thus, the validity of many research results obtained using the kit is unclear. At least metabolites of orexin-A have been proposed as potential interfering substances. METHODS: Since this issue has not been systema

Cysteine redoxome landscape in the liver of male mice fed a high-fat high-sucrose diet.

Reversible cysteine posttranslational modifications serve as a "switch" for protein structure-function dynamics. Herein, we applied a comprehensive strategy to map the cysteine redoxome by pinpointing over 5000 oxidized and reduced cysteine residues in the liver of male mice fed either a normal chow diet or a high-fat/high-sucrose diet (HFHSD). The global and subcellular distribution of oxidized and reduced cysteine residues remained stable across both diet groups, indicating that HFHSD does not induce widespread shifts in cysteine redox equilibrium. Proteomic analyses revealed that HFHSD upre

Alternative Splicing of the NMDA Receptor Subunit GluN1 Mediated by Polypyrimidine Tract-Binding Protein Dimerization in the Trigeminal Ganglion Contributes to Orofacial Allodynia.

Orofacial neuropathic pain, a debilitating condition associated with trigeminal nerve injury, is often characterized by allodynia. N-methyl-d-aspartate receptors (NMDARs), particularly the GluN1 subunit, play a central role in mediating this pain. The GluN1 subunit undergoes alternative splicing at exon 5, generating isoforms GluN1a (lacking the exon 5-encoded N1 cassette) and GluN1b (retaining the N1 cassette), which have distinct functional roles. However, the contribution of GluN1 exon 5 splicing and the mechanisms governing this process in orofacial neuropathic pain are not fully understoo

Combination treatment to improve mucociliary transport of Pseudomonas aeruginosa biofilms.

People with muco-obstructive pulmonary diseases such as cystic fibrosis (CF) and chronic obstructive pulmonary disease (COPD) often have acute or chronic respiratory infections that are difficult to treat due in part to the accumulation of hyperconcentrated mucus within the airway. Mucus accumulation and obstruction promote chronic inflammation and infection and reduce therapeutic efficacy. Bacterial aggregates in the form of biofilms exhibit increased resistance to mechanical stressors from the immune response (e.g., phagocytosis) and chemical treatments including antibiotics. Herein, combina

Structural and mechanistic analysis of Ca2+-dependent regulation of transglutaminase 2 activity using a Ca2+-bound intermediate state.

Mammalian transglutaminases, a family of Ca2+-dependent proteins, are implicated in a variety of diseases. For example, celiac disease (CeD) is an autoimmune disorder whose pathogenesis requires transglutaminase 2 (TG2) to deamidate select glutamine residues in diet-derived gluten peptides. Deamidation involves the formation of transient γ-glutamyl thioester intermediates. Recent studies have revealed that in addition to the deamidated gluten peptides themselves, their corresponding thioester intermediates are also pathogenically relevant. A mechanistic understanding of this relevance is hinde
