Pre-B-cell leukemia homeobox 3 (PBX3) is a homeobox transcription factor that belongs to the pre-B-cell leukemia homeobox family. Studies have revealed that PBX3 plays crucial roles in tumor progression and metastasis. PBX3 upregulation has been linked to various tumors, where it contributes to cell growth and development. One of its primary functions in tumor biology is promoting cell proliferation by modulating the cell cycle. Moreover, PBX3 inhibits apoptotic pathways, allowing tumor cells to survive and resist treatment. In addition to its role in survival, PBX3 is involved in epithelial-m

M2 macrophages play a critical role in the tumor microenvironment of invasive solid tumors. They are closely associated with perineural invasion (PNI) and are often linked to poor prognosis. In this context, tumor-derived exosomes serve as important mediators of intercellular communication. However, the relationship between tumor cell-induced M2 macrophages and PNI in cholangiocarcinoma remains unexplored. In this study, we utilized multiplex immunofluorescence and transcriptomic sequencing to demonstrate the upregulation of LINC01812 in cholangiocarcinoma tissues and its positive correlation

OBJECTIVES: Human RECQL4 and BLM helicases participate in all DNA dependent processes, including replication stress, DNA damage repair. Both helicases are overexpressed in glioblastoma (GBM), a lethal primary brain tumour, characterised by resistance to radio- and chemotherapy. BLM-depleted glioma cells exhibit senescence-associated or polypoid phenotype when exposed to temozolomide (TMZ) and olaparib (OLA), a PARP inhibitor. This study aims to investigate how RECQL4 depletion influences the response of malignant gliomas to chemotherapeutics.
METHODS: We investigated the effect of RECQL4 depl

Solute carrier family 16 member 3 (SLC16A3), a key protein mediating lactic acid efflux, promotes immune evasion and angiogenesis in tumour, correlating with poor prognosis and potentially impairing anti-angiogenic therapies like sorafenib. To elucidate SLC16A3's role and mechanism in hepatocellular carcinoma (HCC), particularly concerning sorafenib, we analysed public datasets and employed molecular biology techniques, animal experiments, and transcriptome sequencing. Our findings revealed that elevated SLC16A3 protein expression correlates with poor prognosis, reduced sorafenib response, and

SLC5A8 is a protein coded by the SLC5A8 gene, and has been proposed as a tumor suppressor and iodide transporter. Its expression is reduced in papillary thyroid carcinoma (PTC), yet the mechanisms underlying this phenomenon are largely unknown. We hypothesized that SLC5A8 expression in PTC is reduced by microRNAs and can be modulated by their inhibition. We used real-time PCR to analyze the expression of SLC5A8 and the microRNAs of interest in a set of 49 PTC/normal tissue pairs. We used an in silico approach to identify microRNAs upregulated in PTC and putatively binding to the SLC5A8 transcr

To explore potential biomarkers in the progression of intrahepatic cholangiocarcinoma (ICC), the gene expression profiles in ICC and paired normal tissues were analyzed using mRNA sequencing. E3 ubiquitin ligase tripartite motif containing 47 (TRIM47) was found highly expressed in ICC tissues (log2 fold change = 2.473, p < 0.001), which was also observed in seven ICC samples compared with paired normal tissues. Patients with higher TRIM47 expression exhibited shorter disease-free survival period. Functional studies indicated that TRIM47 knockdown in ICC cells suppressed cell growth in vitro an

Multiple myeloma (MM) is a complex hematological malignancy characterized by genomic changes and transcriptomic dysregulation. Initial exome sequencing approaches have failed to identify any single, frequent (>25%) mutation in the coding genome. However, using whole-genome sequencing, we found that one of the genomic regions most frequently mutated (62% of the patients with MM) was the 5' untranslated region and/or intron 1 of the BCL7A gene. RNA-sequencing data from a large cohort suggest a loss of BCL7A expression in a large majority of patients with MM as compared with normal plasma cells.

Comprehensive epigenomic studies in multiple myeloma (MM) that unravel the connections between major epigenetic regulators, their intertwined collaboration and the potential of combinatorial targeting remain limited. Utilizing ChIP-seq, ATAC-seq, RNA-seq, and DNA methylation (DNAme) data, we generated whole-genome chromatin annotations from normal plasma cells and MM patients, revealing epigenomic re-configuration affecting downstream genes involved in tumour growth and survival. Primary MM samples showed global DNA hypomethylation but site-specific hypermethylation was observed at transcripti

BACKGROUND: Anaplastic thyroid cancer (ATC) is a rare lethal human malignancy with a poor prognosis. Multimodality treatment, including radiotherapy, is recommended to improve local control and survival. Valproic acid (VA) is a clinically available anticonvulsant and histone deacetylase inhibitor with a well-documented side effect profile. Furthermore, VA radiosensitizes various cancer cells, including thyroid cancer. MicroRNAs (miRs) are deregulated in several cancers and may modulate radiation response. Therefore, the aim of this study was to analyze the effect of VA combined with gamma radi
