Prevalence, transmission, and molecular epidemiology of tet(X)-positive bacteria among humans, animals, and environmental niches in China: An epidemiological, and genomic-based study.

Plasmid-mediated, transmissible, tigecycline-inactivating enzyme Tet(X) has attracted considerable public attention. However, so far studies have not addressed its impact on public health and the ecosystem. Herein, we report the prevalence and molecular epidemiology of tet(X)-positive bacteria (TPB) from diverse sources, investigate the host-specificity of TPB and the transferability of tet(X). Sample collection was conducted between 2018 and 2020 in 30 provinces in China. PCR screening suggested tet(X) was prevalent among freshwater fishes (24.7%, 95% CI 19.4-30.7%), followed by chickens (23.

Host blood transcriptomic biomarkers of tuberculosis disease in people living with HIV: a systematic review protocol.

INTRODUCTION: Current tuberculosis triage and predictive tools offer poor accuracy and are ineffective for detecting asymptomatic disease in people living with HIV (PLHIV). Host tuberculosis transcriptomic biomarkers hold promise for diagnosing prevalent and predicting progression to incident tuberculosis and guiding further investigation, preventive therapy and follow-up. We aim to conduct a systematic review of performance of transcriptomic signatures of tuberculosis in PLHIV. METHODS AND ANALYSIS: We will search MEDLINE (PubMed), WOS Core Collection, Biological Abstracts, and SciELO Citati

Antibody-Dependent Enhancement: A Challenge for Developing a Safe Dengue Vaccine.

In 2019, the United States Food and Drug Administration accorded restricted approval to Sanofi Pasteur's Dengvaxia, a live attenuated vaccine (LAV) for dengue fever, a mosquito-borne viral disease, caused by four antigenically distinct dengue virus serotypes (DENV 1-4). The reason for this limited approval is the concern that this vaccine sensitized some of the dengue-naïve recipients to severe dengue fever. Recent knowledge about the nature of the immune response elicited by DENV viruses suggests that all LAVs have inherent capacity to predominantly elicit antibodies (Abs) against the pre-mem

Baicalein inhibits growth of Epstein-Barr virus-positive nasopharyngeal carcinoma by repressing the activity of EBNA1 Q-promoter.

Epstein-Barr virus (EBV) can establish a life-long latent infection in the host and is associated with various human malignancies, including nasopharyngeal carcinoma (NPC), the most common cancer originated from nasopharynx. EBV nuclear antigen 1 (EBNA1) is the only viral protein absolutely demanded for segregation, replication, transcription and maintenance of EBV viral genome in host cells. Baicalein, a bioactive flavonoid compound purified from the root of Scutellariae baicaleinsis, displays anti-inflammatory, immunosuppressive, and anti-tumor properties. In this study, the therapeutic effe

Structure of the Neisseria Adhesin Complex Protein (ACP) and its role as a novel lysozyme inhibitor.

Pathogenic and commensal Neisseria species produce an Adhesin Complex Protein, which was first characterised in Neisseria meningitidis (Nm) as a novel surface-exposed adhesin with vaccine potential. In the current study, the crystal structure of a recombinant (r)Nm-ACP Type I protein was determined to 1.4 Å resolution: the fold resembles an eight-stranded β-barrel, stabilized by a disulphide bond between the first (Cys38) and last (Cys121) β-strands. There are few main-chain hydrogen bonds linking β4-β5 and β8-β1, so the structure divides into two four-stranded anti-parallel β-sheets (β1-β4 an

Radiolabeled antibodies in prostate cancer: a case study showing the effect of host immunity on antibody bio-distribution.

OBJECTIVES: Human tumors xenografted in immunodeficient mice are crucial models in nuclear medicine to evaluate the effectiveness of candidate diagnostic and therapeutic compounds. However, little attention has been focused on the biological profile of the host model and its potential effects on the bio-distribution and tumor targeting of the tracer compound under study. We specifically investigated the dissimilarity in bio-distribution of (111)In-DTPA-5A10, which targets free prostate specific antigen (fPSA), in two animal models. METHODS: In vivo bio-distribution studies of (111)In-DTPA-5A1

Molecular characterization of a H5N1 highly pathogenic avian influenza virus clade 2.3.2.1b circulating in Vietnam in 2011.

The emergence of highly pathogenic avian influenza (HPAI) H5N1 viruses and the risk of a human pandemic have highlighted the need for advance stockpiling of vaccine. Current vaccines may be sub-optimally matched to the actual pandemic virus due to the rapid dissemination and ongoing evolution of avian H5N1 viruses. We report here the evaluation of efficacy of NIBRG-14 vaccine (clade 1 A/Vietnam/1194/2004) against the H5N1 HPAI virus strains circulating in Vietnam. The birds were either vaccinated with a single or booster dose of vaccine by subcutaneous injection; then challenged with three H5N

Phase variation and microevolution at homopolymeric tracts in Bordetella pertussis.

BACKGROUND: Bordetella pertussis, the causative agent of whooping cough, is a highly clonal pathogen of the respiratory tract. Its lack of genetic diversity, relative to many bacterial pathogens, could limit its ability to adapt to a hostile and changing host environment. This limitation might be overcome by phase variation, as observed for other mucosal pathogens. One of the most common mechanisms of phase variation is reversible expansion or contraction of homopolymeric tracts (HPTs). RESULTS: The genomes of B. pertussis and the two closely related species, B. bronchiseptica and B. parapert

Feeder- and Drinker-Associated Microbiota Differ from Host-Associated Fecal Microbiota During a Commercial Broiler Production Cycle with Age-Associated Changes in Taxonomic Composition.

Broiler chickens are continuously exposed to microorganisms in the production environment, but the relationship between microorganisms on bird-contact surfaces and fecal microbiota remains unclear. In this exploratory observational study, shotgun metagenomic sequencing was used to compare microbial communities in feeder- and drinker-associated surface samples and individual fecal samples during one production cycle in a single commercial system housing Ross 308 broilers. The dataset comprised 22 individual fecal samples collected on days 1, 7, 19, 27, and 39 and four composite feeder- or drink

Fasciolagigantica Recombinant Bone Morphogenetic Protein (FgBMP) Modulates Various Functions of Buffalo Peripheral Blood Mononuclear Cells.

Fasciolosis, a disease induced by liver fluke infection, poses a major threat to the livestock industry. The emergence of drug resistance and the lack of commercial vaccines necessitate the development of novel control strategies. Studies on helminth immunoregulatory molecules have become a key research focus. Bone morphogenetic proteins (BMPs), ligand members of the TGF-β superfamily, have been identified in Fasciola gigantica and designated FgBMPs, and are hypothesized to regulate host immune responses. To explore the effects of FgBMP on hosts, the mechanistic actions of FgBMP-1 on buffalo p

Decoding the interconnected splicing patterns of hepatitis B virus and host using large language and deep learning models.

Hepatitis B virus (HBV) infection causes one million deaths annually and remains a major driver of hepatocellular carcinoma. Despite its compact 3.2 kb genome, HBV exhibits extensive alternative splicing. HBV splice variants contribute to immune evasion and reduce the likelihood of achieving a functional cure. Here, we show that HBV splicing efficiency - quantified from 279 RNA-sequencing libraries of HBV-associated liver biopsies and cultured cells - correlates more strongly with disease progression than the overall proportion of spliced HBV RNA, the latter of which has been proposed as an em

Integrative hologenomic analysis reveals apolipoprotein D-associated host-microbe metabolic crosstalk linked to fat deposition in Jinhua pigs.

BACKGROUND: Adipose deposition is a complex trait shaped by both host genetics and environmental factors, including the gut microbiota. While quantitative trait loci (QTLs) and candidate genes underlying fat accumulation have been extensively identified in diverse pig breeds, the hologenomic regulatory networks integrating host genetic variation and microbial signals remain to be fully deciphered. METHODS: In this study, we utilized a strictly controlled dietary cross-fostering model involving Jinhua and Landrace×Yorkshire pigs across developmental stages (day60, day90 and day180). We integra

Exploiting metabolic shifts: gut colonization strategies of the microaerophilic pathogen Campylobacter jejuni.

Campylobacter jejuni is a microaerophilic, gram-negative bacterium and one of the leading causes of bacterial gastroenteritis worldwide, most commonly transmitted through contaminated poultry products. The organism exhibits striking host-specific behavior, persisting as a harmless commensal in the chicken intestine while causing inflammatory disease and rapid proliferation in the human gut. Despite its clinical importance, the mechanisms underlying nutrient acquisition, metabolic adaptation, and fitness during infection remain incompletely understood. The ferret model of campylobacteriosis clo

Lobosorchis labri n. sp. (Trematoda: Cryptogonimidae): a host switch from snappers (Lutjanidae) to wrasses (Labridae).

The first cryptogonimid trematode with adults parasitic in wrasses (Labriformes: Labridae) is proposed. Lobosorchis labri n. sp. was recovered at a substantial combined prevalence (63%, 15 of 24) from the thicklipped wrasses Hemigymnus melapterus (Bloch) and H. fasciatus (Bloch) at Ningaloo Reef, Western Australia. Definitive hosts for all other Lobosorchis spp. are exclusively tropical snappers, Lutjanus spp. (Lutjaniformes: Lutjanidae). Novel material for Lobosorchis spp. collected from snappers is also reported, including for L. tibaldiae Miller & Cribb, 2005 and L. polygongylus Miller et a

Genetic diversity and host-symbiont cophylogenetic analysis of Neorickettsia endobacteria of Fasciola hepatica.

Neorickettsia are obligate intracellular bacteria that persist as endosymbionts of digenean trematodes and include several vertebrate pathogens. However, many digenean-associated lineages remain poorly characterized, including Neorickettsia associated with the liver fluke Fasciola hepatica (nFh). Here, we present a large-scale population genomic analysis of nFh to assess its prevalence, genetic diversity, evolutionary history, and transmission dynamics. We screened whole-genome sequencing data from 502 F. hepatica samples collected across Peru, the UK, Uruguay, and the USA, detecting Neoricket
