Tendon and ligament regeneration and repair: clinical relevance and developmental paradigm.

As dense connective tissues connecting bone to muscle and bone to bone, respectively, tendon and ligament (T/L) arise from the somitic mesoderm, originating in a recently discovered somitic compartment, the syndetome. Inductive signals from the adjacent sclerotome and myotome upregulate expression of Scleraxis, a key transcription factor for tenogenic and ligamentogenic differentiation. Understanding T/L development is critical to establishing a knowledge base for improving the healing and repair of T/L injuries, a high-burden disease due to the intrinsically poor natural healing response. Design for a lithographically patterned bioartificial endocrine pancreas.

Cell encapsulation provides a means to transplant therapeutic cells for a variety of diseases including diabetes. However, due to the large numbers of cells, approximately on the order of a billion, that need to be transplanted for human diabetes therapy, adequate mass transport of nutrients such as oxygen presents a major challenge. Proof-of-concept for the design of a bioartificial endocrine pancreas (BAEP) that is optimized to minimize hypoxia in a scalable and precise architecture is demonstrated using a combination of simulations and experiments. The BAEP is composed of an array of porous

Human elastic cartilage engineering from cartilage progenitor cells using rotating wall vessel bioreactor.

Transplantation of bioengineered elastic cartilage is considered to be a promising approach for patients with craniofacial defects. We have previously shown that human ear perichondrium harbors a population of cartilage progenitor cells (CPCs). The aim of this study was to examine the use of a rotating wall vessel (RWV) bioreactor for CPCs to engineer 3-D elastic cartilage in vitro. Human CPCs isolated from ear perichondrium were expanded and differentiated into chondrocytes under 2-D culture conditions. Fully differentiated CPCs were seeded into recently developed pC-HAp/ChS (porous material

Enhanced bone formation in large segmental radial defects by combining adipose-derived stem cells expressing bone morphogenetic protein 2 with nHA/RHLC/PLA scaffold.

In this study, rabbit adipose-derived stem cells (rASCs) were isolated, cultured in vitro, and transfected with recombinant adenovirus vector containing human bone morphogenetic protein 2 (Ad-hBMP2). These cells were combined with a nano-hydroxyapatite/recombinant human-like collagen/poly(lactic acid) scaffold (nHA/RHLC/PLA) to fabricate a new biocomposite (hBMP2/rASCs-nHA/RHLC/PLA, group 1) and cultured in osteogenic medium. Non-transfected rASCs mixed with nHA/RHLC/PLA (rASCs-nHA/RHLC/PLA, group 2) and nHA/RHLC/PLA scaffold alone (group 3) served as controls. Scanning electron microscope (SE

Effect of self-assembling peptide, chondrogenic factors, and bone marrow-derived stromal cells on osteochondral repair.

OBJECTIVE: The goal of this study was to test the ability of an injectable self-assembling peptide (KLD) hydrogel with or without chondrogenic factors (CF) and allogeneic bone marrow stromal cells (BMSCs) to stimulate cartilage regeneration in a full-thickness, critically-sized, rabbit cartilage defect model in vivo. We used CF treatments to test the hypotheses that CF would stimulate chondrogenesis and matrix production by cells migrating into acellular KLD (KLD+CF) or by BMSCs delivered in KLD (KLD+CF+BMSCs). DESIGN: Three groups were tested against contralateral untreated controls: KLD, KL

Chest wall injuries and the seat belt syndrome.

A 3-year prospective study of road traffic accidents in which injuries of the chest wall were accurately recorded and related to the use of seat belts was performed in Oxfordshire by a combined team of surgeons and engineers. An increased incidence of soft tissue injuries was noted in restrained front seat occupants though this was not the case for fractures of the clavicle or sternum. The overall incidence of broken ribs in front seat passengers was not affected by the wearing of belts but the incidence of those with four or more broken ribs was considerably reduced by wearing belts.

Genotoxic and Physiological Responses to Repeated Exposure to Low-Concentration Silver Nanoparticles in Vicia faba Through Root and Foliar Uptake Pathways.

Silver nanoparticles (AgNPs) are among the most widely used engineered nanomaterials, increasing the likelihood of repeated plant exposure through soil, water, and atmospheric deposition. However, the biological consequences of repeated exposure through different uptake pathways remain insufficiently understood. This study characterized citrate-reduced AgNPs and evaluated their genotoxic and physiological effects in Vicia faba following repeated root and foliar exposure. Physicochemical characterization included SEM, energy-dispersive X-Ray spectroscopy, dynamic light scattering, zeta potentia

High-efficiency, transgene-free plant genome editing by viral delivery of an engineered TnpB.

Genome editing has revolutionized plant biology research1, yet the efficient delivery of editing reagents remains a challenge. Current methods are labour intensive, involving lengthy tissue culture and complex transformation and regeneration steps. Viral delivery can mitigate these issues2 but CRISPR-Cas nucleases exceed viral cargo limits, restricting guide RNA (gRNA) delivery into Cas9-expressing transgenic plants2-11. This requires generating an initial Cas9 transgenic line. Furthermore, gRNAs delivered by plant viral vectors can induce somatic edits, although only a few produce heritable e

Adeno-Associated virus-based approaches for mitochondrial diseases: advances and challenges.

Mitochondrial diseases, caused by mutations in either mitochondrial or nuclear DNA, are highly complex genetic disorders characterized by faulty oxidative phosphorylation. Adeno-associated virus (AAV)-based gene therapy with its broad and customizable tissue tropism achieved through natural and engineered serotypes offers a highly effective platform for delivering therapeutic genes to affected tissues. However, the intricate genetics and biology of mitochondria present unique challenges for the development of AAV-based therapies. While gene replacement therapy remains a viable strategy for cor

Gene-edited live-attenuated vaccines against Toxoplasma gondii: recent advances and future frontiers.

Toxoplasma gondii is an infectious disease that infects nearly one third of the world's population and endangers the health of immunocompromised people, pregnant women, and livestock. There are no existing drugs able to treat the infection and prevent tissue cysts from developing. Therefore, the creation of a safe vaccine should be considered as a matter of great importance. While many of the vaccines have not proven successful in the past, recently created live-attenuated vaccines (LAVs) using CRISPR-Cas9 gene editing technology were able to outperform the previous ones. The current review ai

Covalent Organic Framework-Based Artificial Antioxidases with π-Electron Delocalization and Asymmetric Coordination Sites for Superior Inflammation Inhibition and Oral Bone Modulation.

Periodontitis-induced alveolar bone loss represents a complex therapeutic challenge that demands simultaneous resolution of chronic inflammation and restoration of bone homeostasis. Drawing inspiration from natural antioxidases, the de novo design of an interlaminar ruthenium-coordinated covalent organic framework (COF) is reported that functions as an artificial antioxidase system. This fully condensed COF architecture (TTf-Ru) combines extended π-electron delocalization with precisely engineered asymmetric ruthenium coordination sites, thereby creating optimal electronic environments for sca

Urease-Powered Nanomotors for Enhanced Mucosal and Tumor Penetration To Augment Sonodynamic Therapy in Bladder Cancer.

Sonodynamic therapy (SDT) represents a promising minimally invasive approach for treating bladder cancer, however, its efficacy following intravesical instillation is substantially compromised by the glycosaminoglycan-rich mucosal barrier and the cyclic voiding process of the bladder. To address these limitations, nanomotors coloaded with enzyme and sonosensitizer were engineered to enhance self-propelled tissue penetration and prolong retention of SDT for bladder cancer. Herein, urine-derived exosomes (EXO) were isolated using density gradient ultracentrifugation and subsequently engineered i
