Evolution of neoplastic cell lineages in Barrett oesophagus.

It has been hypothesized that neoplastic progression develops as a consequence of an acquired genetic instability and the subsequent evolution of clonal populations with accumulated genetic errors. Accordingly, human cancers and some premalignant lesions contain multiple genetic abnormalities not present in the normal tissues from which the neoplasms arose. Barrett oesophagus (BE) is a premalignant condition which predisposes to oesophageal adenocarcinoma (EA) that can be biopsied prospectively over time because endoscopic surveillance is recommended for early detection of cancer. In addition,

Cryoglobulinemic glomerulonephritis: a membranoproliferative glomerulonephritis induced by hepatitis C virus.

Mixed cryoglobulins (MCs) are proteins that precipitate from cooled serum, and are composed of a polyclonal immunoglobulin G (IgG) bound to another immunoglobulin that acts as an anti-IgG rheumatoid factor (RF). In type II mixed cryoglobulinemia, the antiglobulin component, usually of the IgM class, is monoclonal; it is polyclonal in type III mixed cryoglobulinemia. The majority of MCs are found in patients with connective tissue diseases, infectious or lymphoproliferative disorders, hepatobiliary diseases, or immunologically mediated glomerular diseases (secondary MCs). The etiology is not cl

Magnetic resonance imaging or metaiodobenzylguanidine scintigraphy for the demonstration of paragangliomas? Correlations and disparities.

Paragangliomas are tumours arising from paraganglionic tissue dispersed from the base of the skull to the pelvic diaphragm. These tumours produce symptoms by secreting catecholamines (functioning tumours) or by local tumour expansion. They can be part of several hereditary disorders. The introduction of magnetic resonance (MR) imaging and metaiodobenzylguanidine (MIBG) scintigraphy has provided new insights into paragangliomas and has tremendously changed the topographic diagnosis of paragangliomas. Both techniques have proven to be adequate in localising paragangliomas. In this report, the pe

Gram-negative sepsis and the adult respiratory distress syndrome.

Gram-negative sepsis has dramatically increased in frequency throughout the twentieth century in the United States. Currently, approximately 200,000 patients develop gram-negative sepsis each year in this country. Of these, about one-quarter develop the adult respiratory distress syndrome (ARDS). Among these critically ill patients, mortality is estimated at 60%-90%. In the complex series of events leading to acute lung injury in gram-negative sepsis, endotoxin is the proximal mediator. Although endotoxin may be capable of causing direct injury to the pulmonary endothelium, its primary role is

Myeloma phenotype: clues to disease origin and manifestation.

Phenotypic analysis of myeloma cells has had a major impact on our understanding of the development of the disease. Heterogeneity in the expression of lineage- and differentiation-associated antigens has helped delineate a circulating clonal premyeloma cell compartment coexpressing CD19 and CD11b. These cells can be stimulated in vitro to proliferate and differentiate into the mature myeloma cells. Other studies have demonstrated the involvement of very early bone marrow B lymphocytes, which could be differentiated into myeloma cells through a CD10-positive intermediate stage. These data sugge

Congenital goiter sustaining normal level of serum triiodothyronine.

We attempted to elucidate the deficient site of thyroid hormone biosynthesis in the thyroid gland and the mechanism of sustaining normal T3 level in sera of a patient with congenital goiter. TY, a 8-yr-old boy, first noted the onset of a diffuse goiter at the age of 2. There was no clinical evidence of hypothyroidism except for the slight impairment of intellectual development and the awkward physical activity. BMR, T3-RSU and T4 showed low values (-13%, 20.8% and 2.2 micrograms/dl), but serum T3 was normal (180 ng/dl). Serum TSH was 18 microU/ml. The intrathyroidal T3 and T4 were slightly low

Effects of albumin-bound paclitaxel combined with Sophora subprostrate polysaccharide on inflammatory factors and immune function in breast cancer rats.

BACKGROUND: Tumor occurrence and growth are highly correlated with the degree of inflammation and immunological activity. Reducing the level of inflammation in tumor-bearing body to relieve immune suppression and enhance anti-tumor immune function has become an important strategy for tumor treatment. OBJECTIVE: To investigate the effect of albumin-bound paclitaxel combined with Sophora subprostrate polysaccharide (SSP) on inhibiting inflammation, reducing immunosuppression, enhancing anti-tumor immune function and slowing the progression of tumor in tumor-bearing rats, and to provide certain

Safety and Biodistribution of NK Cell-Based Immunotherapy in Healthy and Cancer-Bearing Humanized-BLT Mice.

Natural killer (NK) cells are long known for their anti-cancer function; however, their use in clinical trials has not been well-positioned yet due to the lack of reliable methods to augment their expansion and anti-tumor potential. In our previous publications, we have demonstrated the methodology to enhance the number and function of NK cells by co-culturing peripheral blood-derived NK cells with osteoclasts (OCs) and sonicated probiotic bacteria (sAJ2). Due to significantly increased potency, these NK cells were named supercharged NK (sNK) cells. sNK cells have significantly high anti-tumor

Association of environmental xylene exposure with diabetic kidney disease: a cross-sectional analysis and multi-omics evaluation of immune microenvironment mechanisms.

Diabetic kidney disease (DKD) progression is driven by residual inflammation, potentially exacerbated by volatile organic compounds (VOCs). However, the impact of xylene exposure on the DKD immune microenvironment remains unclear. We evaluated the association between urinary VOC metabolites and DKD prevalence using NHANES data. Network toxicology and bulk transcriptomics (GSE142025) were integrated to identify core targets and profile immune infiltration. Single-cell RNA sequencing (GSE209781) validated macrophage heterogeneity and hub gene dynamics, and molecular docking assessed xylene-recep

Single-cell RNA sequencing reveals the role of neutrophils in intestinal ischemia-reperfusion injury in mice.

OBJECTIVE: Intestinal ischemia-reperfusion injury (IRI) severely compromises the mucosal barrier. While neutrophils are key responders, their specific roles and mechanisms remain unclear. This study aims to delineate the functional heterogeneity of neutrophils in intestinal IRI and identify the underlying molecular pathways. METHODS: A murine model of intestinal IRI was established by subjecting C57BL/6 mice to 45 min of ischemia and 4 h of reperfusion. Single-cell RNA sequencing (scRNA-seq) was performed on intestinal tissues. Neutrophil depletion, co-culture assays, and pharmacological modu

Two Decades of Nanotoxicology: Evolution, Impact, and Future Challenges.

The rapid expansion of nanotechnology, with over 11,100 products and 100,000 patents, has raised safety concerns for organisms and ecosystems. Nanotoxicology emerged from ultrafine particle studies in the 1990s and was formally established in 2004. This review synthesizes approximately 160 publications and regulatory documents, selected from 220 compiled, to analyze the field's evolution from 2004 to 2025. Early studies focused on nanostructures in medicine and their effects on organs and tissues, driving research aimed at mitigating risks. Current studies increasingly address the molecular me

Genome-wide identification of CHY zinc finger and RING finger (CHYR) genes in pepper and functional characterization of CaCHYR5 in response to Phytophthora capsici infection.

CHY zinc finger and RING finger (CHYR) proteins play crucial roles in the growth and development, as well as stress response. To date, no systematic or comprehensive analysis of the CHYR gene family has been performed in pepper (Capsicum annuum L.). In this study, we identified 8 CaCHYR genes (CaCHYR1-CaCHYR8), which were classified into 3 groups based on phylogenetic relationships. CaCHYR members within the same group exhibited similar distributions of conserved motifs and exon-intron structures. Chromosomal localization analysis showed that 8 CaCHYR genes were unevenly distributed on 6 chrom

Genome-Wide Characterization of the ZIP Transporter Family in Sea Island Cotton (Gossypium barbadense L.) and Expression Profiling Under Heavy Metal and Pathogen Stresses.

G. barbadense represents an indispensable germplasm resource for high-quality textile fiber and disease resistance; nevertheless, systematic information regarding its ZRT/IRT-like protein (ZIP) gene family remains limited. Here, a total of 46 GbZIP genes were identified across the G. barbadense genome. Comprehensive bioinformatic investigations revealed uneven chromosomal distribution and confirmed that segmental/whole-genome duplications, supplemented by localized tandem duplications, drove family expansion. Members clustered within the same phylogenetic clades shared conserved motif organiza

Ultrastructural and Proteomic Signatures of Mechanoadaptive Fibroblast Remodeling across Microphysiological and Mesoscale Shear Platforms.

Mechanobiological cues in the tissue microenvironment increasingly drive pathological fibroblast activation in inflammatory bowel disease (IBD), yet engineered platforms modeling this transition remain limited. Here, a microfluidic gut-on-a-chip microphysiological system and a mesofluidic rotary shaker are used to demonstrate that sustained fluid shear stress alone is necessary and sufficient to drive an irreversible, profibrotic phenotypic switch in primary normal human intestinal fibroblasts. Across both platforms, normal fibroblasts from small and large intestine reproducibly self-organize

Multi-Omics Analysis Reveals Coordinated Adaptations in Genes, Metabolism, and Gut Microbiota Underpinning Herbivory in Lordiphosa Flies.

Herbivorous insects are among the most ecologically successful animal groups. However, the adaptive mechanisms that allow them to exploit plant hosts, which are often nutrient-poor (low in simple sugars, high in structural carbohydrates) and defended by toxic secondary metabolites, are not fully resolved. Here, we investigated the evolutionary basis of herbivory in Lordiphosa clarofinis, a drosophilid species feeding on living plant tissues, using multi-omics approaches. Behavioral experiments revealed a strong oviposition preference for Galinsoga parviflora (a host rich in secondary metabolit

BCL9 inhibition promotes fibroblast lipogenesis by regulating macrophage-fibroblast interactions to attenuate pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal interstitial lung disease with an urgent need for novel therapeutic strategies. M2 macrophage-derived TGF-β1 promotes fibroblast myogenesis, contributing to IPF pathogenesis. Targeting macrophage polarization and fibroblast function thus represents an effective therapeutic approach for treating IPF. Here, we identify B-cell lymphoma 9 (BCL9) as a key upstream regulator implicated in IPF pathogenesis. We demonstrate that BCL9 drives the macrophage M2 program through the MerTK-ERK-SPP1 axis. Notably, pharmacological inhibition of BCL

Histiocytosis development and clinical variation through the lens of genomics.

Histiocytic neoplasms are rare haematologic diseases characterised by clonal expansions of cells with a monocyte, macrophage or dendritic cell phenotype. Their clinical manifestations are diverse, ranging from indolent lesions to aggressive systemic disease. Over recent decades, advances in genomic profiling have transformed the biological understanding of these conditions. The discovery of recurrent oncogenic mutations has reframed histiocytoses from primary inflammatory disorders to myeloid neoplasms, with a notable dependence on aberrant mitogen-activated protein kinase (MAPK) signalling. Humanized hiPSC Platforms for I/R Injury: Advancing Toward Precision Cardioprotection.

Myocardial ischemia-reperfusion (I/R) injury remains a major contributor to infarct expansion, adverse remodeling, and heart failure despite timely coronary revascularization. The repeated failure of cardioprotective interventions that were effective in animal models has exposed a persistent translational gap. Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) and engineered cardiac platforms preserve donor-specific human genetic backgrounds, provide scalable cell sources, and offer experimentally tractable systems for studying selected mechanisms of I/R injury. Snail immunity to schistosomes: insights from omics studies.

Schistosomiasis is a serious public health concern, with transmission facilitated by a small number of freshwater snail intermediate host species. Infection outcomes vary greatly across the primary vector genera, Biomphalaria (for Schistosoma mansoni), Bulinus (for S. haematobium), and Oncomelania (for S. japonicum), even within species, ranging from full resistance to high compatibility. Omics methods have altered this field by correlating host genotype, baseline immunological status, and time-resolved responses to whether invading miracidia are eliminated or develop sporocysts. Evidence from
