On the convergent evolution of animal toxins. Conservation of a diad of functional residues in potassium channel-blocking toxins with unrelated structures.

BgK is a K+ channel-blocking toxin from the sea anemone Bunodosoma granulifera. It is a 37-residue protein that adopts a novel fold, as determined by NMR and modeling. An alanine-scanning-based analysis revealed the functional importance of five residues, which include a critical lysine and an aromatic residue separated by 6.6 +/- 1.0 A. The same diad is found in the three known homologous toxins from sea anemones. More strikingly, a similar functional diad is present in all K+ channel-blocking toxins from scorpions, although these toxins adopt a distinct scaffold. Moreover, the functional dia

Reversibility of stunting: epidemiological findings in children from developing countries.

The growth literature from developing countries is reviewed to assess the extent to which stunting, a phenomenon of early childhood, can be … growth later in life. Improvements in living conditions, as through food supplementation or through adoption, trigger catch-up growth but do so more effectively in the very young. One study cautions that in older adopted subjects, accelerated growth may accelerate maturation, shorten the growth period and lead to short adult stature.

A family study of Gilles de la Tourette syndrome.

Previous studies have demonstrated that Gilles de la Tourette syndrome (TS) is a familial disorder and that chronic tics (CT) and obsessive compulsive disorder (OCD) appear to be etiologically related to the syndrome. In the present study we report the results from a study of 338 biological relatives of 86 TS probands, 21 biologically unrelated relatives of adopted TS probands, and 22 relatives of normal subjects. The 43 first-degree relatives of the adopted TS and normal probands constituted a control sample. The rates of TS, CT, and OCD in the total sample of biological relatives of TS proba

Oral-dependence and gender: factors in help-seeking response set and self-reported psychopathology in psychiatric inpatients.

Psychoanalytic theory suggests that orality and being female are positively related to adopting a help-seeking stance in the world. This may result in inflated scores on self-report psychological tests. These hypotheses were investigated using Rorschach-based, oral-dependence scores (Masling, 1986), the Minnesota Multiphasic Personality Inventory (MMPI) help-seeking F minus K index (Gough, 1950; Meehl, 1951), and the MMPI clinical and supplementary scales. High orals are found to be more likely than low orals to adopt a help-seeking response set which in turn results in inflated scores on a nu

Induction of immunity with avirulent Listeria monocytogenes 19113 depends on bacterial replication.

Events necessary for triggering the cell-mediated response to intracellular parasites are poorly understood. Here we show that extremely high doses of avirulent Listeria monocytogenes 19113 (greater than 10(9] induce a modest and short-lived state of resistance in BALB/c mice. Induction of this protective state could not be achieved with nonviable bacteria and was blocked by inhibiting replication of viable L. monocytogenes 19113 through antibiotic treatment. The immune response was antigen specific and could be adoptively transferred with lymphoid cells. However, unlike the prototypic acquire

Conformational effects of ether bridge substitution in thyroid hormone analogues.

It has been suggested that the oxygen bridge linking the two iodophenyl rings of the thyroid hormones at an angle of 120 degrees promotes electron transfer between the rings or establishes the proper stereochemical relationship between the rings. To determine the effects of oxygen bridge substitution on conformation, an analysis of the X-ray crystal structures of a number of oxygen bridge analogues (NH, CO, CHOH, S) was undertaken and the results compared with the thyroid hormone structures. The 3,5-iodine substituents cause the two phenyl rings to adopt a skewed or twist-skewed conformation.

Transfusion Medicine: New Clinical Applications of Cellular Immunotherapy.

There is now clear clinical evidence that adoptive cellular immunotherapy can eradicate hematologic malignancy and cure otherwise lethal viral infections. With this knowledge comes the challenge of improving the effectiveness and safety of the approach and of simplifying the methodologies required whilst still meeting appropriate federal regulatory guidelines. This review provides an overview of the current status of cellular immunotherapies and addresses how they may be implemented and the future directions they are likely to take. In Section I, Dr. Brenner with Drs. Rossig and Sili reviews t

Cell to cell interaction in the immune response. 3. Chromosomal marker analysis of single antibody-forming cells in reconstituted, irradiated, or thymectomized mice.

Two new methods are described for making chromosomal spreads of single antibody-forming cells. The first depends on the controlled rupture of cells in small microdroplets through the use of a mild detergent and application of a mechanical stress on the cell. The second is a microadaptation of the conventional Ford technique. Both methods have a success rate of over 50%, though the quality of chromosomal spreads obtained is generally not as good as with conventional methods. These techniques have been applied to an analysis of cell to cell interaction in adoptive immune responses, using the ful

Efficacy and safety of autologous CD5-KO anti-CD5 CAR-T cells in relapsed/refractory CD5+ hematological malignancies.

Chimeric antigen receptor (CAR)-T cell therapy targeting antigens shared with normal T cells requires genetic modifications to prevent fratricide. This phase 1 trial evaluates autologous CD5-targeting CAR-T cells with CD5 gene deletion (CT125A) in seven patients with relapsed/refractory CD5+ hematologic malignancies. The overall response rate is 85.7%, including four complete responses. All patients experience cytokine release syndrome (six grade 1-2, one grade 3), and two patients develop immune effector cell-associated neurotoxicity syndrome. The most common grade ≥3 adverse events are cytop

Mathematical modeling of immune counter-regulation predicts efficacy of fractionated CD8+ T cell dosing strategies.

Adoptive cell therapy (ACT) with tumor-specific CD8+ T cells (TSTs) induces tumor regression but rarely achieves durable responses in solid tumors. This limitation stems from secondary counter-regulatory mechanisms that are also induced by ACT, notably the recruitment of tumor-infiltrating myeloid cells (TIMs). However, the precise dynamics of these responses and the optimal TST dosing strategies to overcome their immunosuppression remain largely unclear. Here, we developed a mathematical model of ACT incorporating TIM-driven counter-regulation and simulated alternative TST dosing strategies b

Overcoming T cell exhaustion and senescence in CAR T cell therapy for solid tumors.

Chimeric antigen receptor (CAR) T cell therapies have demonstrated remarkable efficacy in hematologic malignancies; however, their clinical benefit in solid tumors remains limited. A major barrier is T cell dysfunction, particularly exhaustion and senescence, which impair persistence, effector function, and durable tumor control. Targeting these dysfunctional states is therefore essential in order to improve CAR T cell efficacy in solid tumors. This review summarizes recent preclinical strategies aimed at preventing or reversing CAR T cell exhaustion and senescence in solid malignancies. While

A novel Chimeric Antigen Receptor (CAR) - strategy to target EGFRVIII-mutated glioblastoma cells via macrophages.

BACKGROUND: Autologous chimeric antigen receptor (CAR) expressing T-Cells (CAR-T) have been efficiently used in hematological malignancies but their efficacy in solid tumors remains limited. CAR therapies via the use of macrophages, offer a promising avenue due to their unique ability to infiltrate tumors and to initiate phagocytosis. METHODS: To generate a preclinical model of CAR-Macs, we have designed a novel CAR-construct to target EGFRVIII-expressing glioblastoma cells (DK-MG) using THP-1 monocytic cell line able to differentiate towards macrophages. The CAR structure comprises a ScFv re

In-house manufacture of plasma-derived human AB serum for use in cellular therapeutics.

Successful manufacture of cellular therapeutic products requires a medium containing key nutrients to support cell expansion. Some of the most common supplements to cell culture media include serum products such as fetal bovine serum, human serum albumin, and human AB serum. Medium supplementation with human AB serum provides critical growth factors, lipids, proteins, and hormones without the risk of cross-species antigenicity or anti-AB antibodies. In this report, we describe the use of pooled defibrinated plasma, sourced from a US blood bank, to produce serum in-house that complies with bloo

Reprogramming CAR T cells across genetic, epigenetic, metabolic and microenvironmental axes to improve efficacy and safety in cancer and autoimmune disease.

CAR T-cell therapy has delivered durable remissions in several hematologic cancers, yet activity in solid tumors and extension to immune-mediated diseases remain constrained by recurring failure modes: imperfect antigen specificity, inadequate trafficking, progressive dysfunction under chronic stimulation, and toxic inflammatory syndromes. Early reports of CAR-based immune "resets" in refractory autoimmune disease amplify both promise and stakes, because acceptable risk is lower than in cancer and "on-target" effects may still be clinically unacceptable if they create long-term immunodeficienc
