Nitric oxide signaling through three receptors regulates virulence, biofilm formation, and phenotypic heterogeneity of Legionella pneumophila.

UNLABELLED: The causative agent of Legionnaires' disease, Legionella pneumophila, is an environmental bacterium, that replicates in macrophages, parasitizes amoeba, and forms biofilms. L. pneumophila employs the Legionella quorum sensing (Lqs) system and the transcription factor LvbR to control various bacterial traits, including virulence and biofilm architecture. LvbR negatively regulates the nitric oxide (NO) receptor Hnox1, linking quorum sensing to NO signaling. Here, we assessed the response of L. pneumophila to NO and investigated bacterial receptors underlying this process. Chemical NO…

CryoEM analysis of the essential native UDP-glucose pyrophosphorylase from Aspergillus nidulans reveals key conformations for activity regulation and function.

Invasive aspergillosis is one of the most serious clinical invasive fungal infections, resulting in a high case fatality rate among immunoco … ed subunit and 4 Å for the octameric complex) of a native AnUGP. The structure reveals an octameric architecture with each subunit comprising an N-terminal α-helical domain, a central catalytic glycosyltransferase A-like (GT-A-like) domain, and a C-terminal (CT) left-handed β-helix oligomerization domain. AnUGP displays unprecedented conformational variability …

Bacterial TLR2/6 Ligands Block Ciliogenesis, Derepress Hedgehog Signaling, and Expand the Neocortex.

Microbial components have a range of direct effects on the fetal brain. However, little is known about the cellular targets and molecular mechanisms that mediate these effects. Neural progenitor cells (NPCs) control the size and architecture of the brain and understanding the mechanisms regulating NPCs is crucial to understanding brain developmental disorders. We identify ventricular radial glia (vRG), the primary NPC, as the target of bacterial cell wall (BCW) generated during the antibiotic treatment of maternal pneumonia. BCW enhanced proliferative potential of vRGs by shortening the cell c…

Membrane anchoring of Epstein-Barr virus gp42 inhibits fusion with B cells even with increased flexibility allowed by engineered spacers.

UNLABELLED: We recently described the architecture of the Epstein-Barr virus (EBV) fusion-triggering complex consisting of the EBV B cell receptor human leukocyte antigen (HLA) class II and the EBV-encoded proteins gp42 and gH/gL. The architecture of this structure positioned the main body of gp42, comprising the C-type lectin domain (CTLD), away from the membrane and distant from where the membrane-bound form of gp42 might be tethered. gp42 is a type II membrane glycoprotein, with functional gp42 formed by cleavage near the gp42 amino-terminal transmembrane domain. This cleavage results in an…

Identification of human B and T lymphocytes by scanning electron microscopy.

In this study a variety of human lymphocytes of known B or T cell type, obtained from multiple sources, were prepared for scanning electron microscopy (SEM) by the critical point drying method. Distinction between normal B and T lymphocytes was relatively easy in most instances, on the basis of their surface architecture. Using immunological methods, between 20 and 30% of normal peripheral blood lymphocytes (PBL) were identified as B cells and from 69 to 82% as T cells. SEM results showed that 20% of the PBL had a complex villous surface and approximately 80% of cells were smaller and had a re…

Having the landscape in the background: What can metabolic constraints explain about memory?

Haueis and Colaço claim that metabolic considerations function as explanatory constraints. I argue the explanatory contribution of metabolic constraints for synaptic models of memory is not well delineated. While metabolic constraints determine which synaptic architectures are biologically possible, they do not structure the outcome of synaptic consolidation. Metabolic constraints are explanatory since they account for the background conditions of memory formation.

Synthesis and Thorough Investigation of Discrete Organotin Telluride Clusters.

Systematic experimental and theoretical investigations of reactions of R(1)SnCl3 (R(1) = CMe2CH2C(Me)O) with (Me3Si)2Te allowed for the stepwise formation and single-crystalline isolation of the first tin sesquitelluride clusters with functional organic ligands. Subsequent derivatization of the latter took place under reorganization of the inorganic core, affording clusters with complex hybrid architectures.

Iron-Catalyzed Defluorinative Alkylation for Accessing Cyclic Acetal-gem-Difluoroalkenes.

Cyclic acetals and gem-difluoroalkenes are proven structural motifs in medicinal chemistry, yet their modular incorporation into a densely functionalized hybrid architecture remains an uncharted frontier. Herein, we disclose an Fe-catalyzed reductive defluorinative cross-coupling between trifluoromethyl alkenes and 1,3-dioxolanyl- or 1,3-dioxepanyl-substituted alkyl iodides, enabling the rapid assembly of five- or seven-membered cyclic acetal-embodied fluorinated scaffolds, respectively. This protocol enriches defluorinative coupling versatility and offers streamlined access to structurally di…

Sleep-circadian modulation of autophagy and glymphatic function: failure of coordinated brain clearance in Parkinson's disease.

Parkinson's disease (PD) involves α-synuclein accumulation and progressive neurodegeneration. This review proposes that PD reflects failure of sleep-circadian regulated brain clearance network integrating intracellular autophagy and extracellular glymphatic transport across neurons, glia, and blood-brain-barrier. Disruption of sleep-architecture and circadian-timing desynchronizes these processes, impairing protein clearance and promoting pathology. This framework positions sleep-circadian dysfunction as an upstream mechanism, suggesting that restoring sleep-circadian regulation may improve pr…

A hollow PtPdRhRuCoNi high-entropy alloy as an effective electrocatalyst for ethanol, ethylene glycol and glycerol oxidation.

The hollow PtPdRhRuCoNi high-entropy alloy was synthesized via an oil-bath and subsequent etching. With a specific surface area 1.7 times larger than Pt black, it exhibits superior activity and stability in 1 M KOH electrolytes containing ethanol, ethylene glycol, or glycerol. These improvements originate from the synergistic effect between its unique hollow architecture, which provides abundant accessible active sites, and its multi-element composition, which optimizes the electronic structure and facilitates efficient electron transfer.

Human-centric preclinical models for CAR T cell therapy in solid tumors: Where patient-derived organoids fit.

Chimeric antigen receptor (CAR) T cell therapy has shown promising activity in solid tumors, yet translational progress remains limited by preclinical models that incompletely capture human tumor biology. Patient-derived organoids (PDOs) preserve key features of human tumors, including cellular heterogeneity and tissue architecture, thereby enabling functional evaluation of CAR T cell targets, next-generation approaches, resistance mechanisms, and selected safety considerations. We discuss how PDOs complement existing preclinical platforms to support CAR T cell development for solid tumors.

Case report of canine discoid lupus erythematosus in Guatemala.

An 11-year-old male Golden Retriever was presented for consultation due to a chronic progressive lesion on the nose that had started a year before. The majority of the nasal mucosa was affected, with the disruption of the normal architecture, pigment atrophy and abundant peeling on the rostral plane. Histopathology revealed a band of lichenoid infiltrate at the interface and vacuolation of the cells in the basal layer consistent with a diagnosis of canine discoid lupus erythematosus.

Esthetic Correction of Orthodontically Transposed Teeth with Veneers and Laser Periodontal Modification.

Missing teeth in the esthetic zone, whether congenital or as a result of other factors, present difficult choices in clinical management. The missing teeth can be replaced by surgical or restorative intervention but are often treated orthodontically. These repositioned teeth often lead to an unaesthetic result because of differences in morphology, color, and particularly in gingival architecture. This article describes the use of multiple lasers for periodontal modification and feldspathic porcelain veneers to achieve a highly esthetic result.

Structure and allosteric regulation of eukaryotic 6-phosphofructokinases.

Although the crystal structures of prokaryotic 6-phosphofructokinase, a key enzyme of glycolysis, have been available for almost 25 years now, structural information about the more complex and highly regulated eukaryotic enzymes is still lacking until now. This review provides an overview of the current knowledge of eukaryotic 6-phosphofructokinase based on recent crystal structures, kinetic analyses and site-directed mutagenesis data with special focus on the molecular architecture and the structural basis of allosteric regulation.

Concise review: personalized human bone grafts for reconstructing head and face.

Regeneration of normal shape, architecture, and function of craniofacial tissues following congenital abnormality, trauma, or surgical treatment presents special problems to tissue engineering. Because of the great variations in properties of these tissues, currently available treatment options fall short of adequate care. We propose that the engineering of personalized bone graft customized to the patient and the specific clinical condition would revolutionize the way we currently treat craniofacial defects and discuss some of the current and emerging treatment modalities.

Traumatic brain injury-associated delayed sleep phase syndrome.

A 15-year-old girl developed a prominent delayed sleep phase syndrome (DSPS) following traumatic brain injury. Several physiological markers of the sleep-wake rhythm: plasma melatonin, body temperature, wrist activity and sleep architecture (EEG) were delayed almost half a day, returning to normal after treatment with 5 mg melatonin. This report suggests an association between traumatic brain injury and DSPS. Awareness of this phenomenon may result in better possibilities for treatment of patients with brain injury.

Effect of testicular torsion on the contralateral testis.

It has been previously shown that unilateral testicular torsion can cause disruptive anatomic changes in the contralateral testis in rats. In this experimental study plasma and urine prostaglandin E2 levels were studied correlatively with testicular histopathology in acute testicular torsion cases. As a result of this study, necrobiotic morphologic alterations causing testis necrosis and significant increase in plasma prostaglandin E2 levels were observed. Contralateral testicular histology was analyzed in all dogs. None of them showed abnormal tubular architecture.

Endogenous lipid pneumonia in opossums from Louisiana.

Endogenous lipid pneumonia was present in 19 of 27 opossums (Didelphis virginiana) trapped in the vicinity of Baton Rouge, Louisiana. The severity of lesions varied from small pleural and subpleural aggregates of foamy macrophages with minimal disruption of pulmonary architecture to large nodular accumulations of foam cells with cholesterol clefts and localized emphysema. The cause of the lipid pneumonia may be related to pulmonary nematode parasite infections, which were evident in 13 of the affected animals.

[Modification of the tissue thickness of the bladder in obstruction by urethral stenosis].

In experimental obstructed bladder by urethral stenosis, the trigone and the detrusor walls thicken in a very important but different way. The thickened mucosa and submucosa of the total bladder show oedema and hypertrophy of the connective fibro-elastic tissue. Related to the different axis of the pressure inside the bladder and the specific muscular architecture of both regions, the adventitial and muscular layers show no modification in the trigone but they become thinner in the detrusor.

A Macrophage-Integrated 3D Spheroid Model Inoculated with Bacteria for Simultaneous Evaluation of Antibacterial Efficacy, Cytotoxicity, and Immunomodulation.

Conventional antimicrobial-material testing typically relies on separate bacterial assays and two-dimensional (2D) mammalian cell cultures. Although essential as first-line screening tools, these approaches do not capture the three-dimensional (3D) tissue architecture, diffusion constraints, extracellular-matrix remodeling, and innate immune interactions encountered by host-contacting antimicrobial materials. Herein, this study introduces a macrophage-containing 3D spheroid model designed for the simultaneous evaluation of antimicrobial efficacy, host-cell cytotoxicity, and immune responses. T…
