Duchenne muscular dystrophy (DMD) is a genetic disorder that results from deficiency of the dystrophin protein. In recent years, DMD pathological models have been created using induced pluripotent stem (iPS) cells derived from DMD patients. In addition, gene therapy using CRISPR-Cas9 technology to repair the dystrophin gene has been proposed as a new treatment method for DMD. However, it is not known whether the contractile function of myotubes derived from gene-repaired iPS cells can be restored. We therefore investigated the maturation of myotubes in electrical pulse stimulation culture and

INTRODUCTION: The purpose of this study was to propose a method that allows extraction of the current muscle state under electrically induced fatigue.
METHODS: The triceps surae muscle of 5 subjects paralyzed by spinal cord injury was fatigued by intermittent electrical stimulation (5 × 5 trains at 30 Hz). Classical fatigue indices representing muscle contractile properties [peak twitch (Pt) and half-relaxation time (HRT)] were assessed before and after each 5-train series and were used to identify 2 relevant parameters (Fm , Ur ) of a previously developed mathematical model using the Sigma-P

Dystonia is a neurological disorder characterized by sustained or repetitive involuntary muscle contractions and abnormal postures. In the present article, we will introduce our recent electrophysiological studies in hyperkinetic transgenic mice generated as a model of DYT1 dystonia and in a human cervical dystonia patient, and discuss the pathophysiology of dystonia on the basis of these electrophysiological findings. Recording of neuronal activity in the awake state of DYT1 dystonia model mice revealed reduced spontaneous activity with bursts and pauses in both internal (GPi) and external (G

Neuroanatomical studies demonstrated the existence of direct descending projections from the insular cortex to the area of the solitary tract nucleus responsible for mediating the vagovagal reflexes of the proximal part of the gastrointestinal tract. These studies provided grounds for suggesting that one of the mechanisms mediating the influences of the insular cortex on stomach function may be modulation of its vagovagal reflex reactions, one of which is the antrofundal inhibitory reflex. Experiments on conscious dogs were performed to study the effects of electrical stimulation of the insula

Electron microscopy with negative contrast showed that direct interaction of one of the peptide fractions of scorpion venom with monomeric chromatographically pure actin led to polymerization of actin, transforming it from the globular form to the fibrillar form. The effects of prolonged orthodromic stimulation on the evoked electrical activity and ultrastructure of Mauthner neurons (MN) were studied in incubated slices of goldfish medulla oblongata in the presence of this actin-polymerizing venom fraction. Peptides in this fraction were found to stabilize the amplitude of the electrical respo

The estimation of the maximum comfort loudness levels (MCL) by measurements of the electrically elicited stapedius reflex was examined in six experienced cochlear implant users supplied with the COMBI 40 implant system. The stapedius reflex was tested and loudness scaling was performed simultaneously using an up/down stimulation protocol close to the reflex threshold with automated recording of both test procedures. The electrical stapedius reflex threshold (ESRT) and loudness scaling were evaluated separately. Scaling at the reflex threshold ranged between normal and loud. The range of stimul

The topographical organization of SI and SII somatosensory areas was investigated using fMRI at 1.5 T and electrical sensory stimulation. Electrical stimuli were delivered unilaterally to the median nerve at the wrist and to the tibial nerve at the medial malleolus, during a block paradigm study. In all subjects, activation was observed, contralaterally to the stimulated side, in the post-central gyrus, in the posterior parietal cortex, in the mesial pre-frontal region and, bilaterally, in the supratemporal region at the level of the Sylvian fissure. The latter region, corresponding presumably

An alternative technique for the continuous monitoring of the facial nerve, monopolar recording of facial nerve antidromic potentials (FNAPs), on 10 subjects undergoing retrosigmoid vestibular neurectomy for Meniere's disease is described. To elicit FNAPs bipolar electrical stimulation of the marginalis mandibulae was performed. Stimulus intensity ranged from 0 to 10 mA with a delivery rate of 7/second. Antidromic potentials were recorded with a silver wire monopolar electrode positioned intracranially on the proximal portion (root entry zone) of the acoustic-facial bundle. Bipolar recordings

The effects of local spinal applications of the substance P antagonist, CP-96,345, on the electrically-evoked discharges of nociceptive neurons were examined in the superficial spinal dorsal horn of adult rats anesthetized with sodium pentobarbital. Extracellular single-unit recordings were made from lumbar spinal dorsal horn neurons which were excited by noxious mechanical stimulation and had early and late discharges evoked by A- and C-fiber inputs, respectively, following transcutaneous electrical stimulation. CP-96,345 was applied directly onto the surface of the spinal cord close to the r

Physiological properties of guinea-pig ureteral segments were studied by measuring the internal pressure responses to electrical stimulation in iso-volume conditions and by recording the contraction elicited by local electrical stimulation in iso-pressure conditions with a video-system. In iso-volume conditions, the phasic pressure response of the ureter increased in magnitude with increasing resting intra-luminal pressure and its threshold was lower in the proximal end, i.e., the part nearest to the renal pelvis, and higher in the distal end. In iso-pressure conditions, the average conduction

Gastroparesis or gastric stasis is the delayed transit of the ingested contents through the stomach in the absence of mechanical obstruction. It can have multiple etiologies, most commonly idiopathic (ID) and diabetic (DM). Gastroparesis can cause significant distress to patients as it leads to symptoms like intractable nausea and vomiting, weight loss, abdominal bloating, early satiety, etc. The pathogenesis is mainly thought to be due to the dysfunction of the gastric pacemaker cells, i.e., interstitial cells of Cajal (ICC), and their interaction with the other gastric motor function regulat

Reappearance of sympathetic skin response (SSR) below lesion is reported in a patient with a complete thoracic-9 spinal cord injury 6 months following injury. SSR was elicited by electrical stimulation of supraorbital nerve (SON) and pudendal nerve (PN). SON stimulation induced SSRs only in the hand. SSRs were initially absent below the level of SCI but reappeared only with PN stimulation. This case suggests that 6 months following a complete lesion, the isolated spinal cord can generate a SSR. Possible underlying mechanisms and implications for autonomic plasticity below spinal lesion are dis

Babies and young children with early spinal cord injury (SCI) have evidence of an improved level of recovery over an extended time period. Enhanced neuroplasticity is well recognized in neonatal animal models. In the young human, developmental apraxia and learned early habitual movements mask expression of residual or recovered motor function. Techniques providing sensorimotor stimulation with threshold electrical stimulation (TES) and EMG triggered stimulation (ETS) act to increase awareness and useful function. Small cohort size and prolonged developmental maturation argue for the use of sin

In chloralosed cats, usually with ligated adrenals and paralysed with gallamine, the thoracic dorsal roots were electrically stimulated and gastric motor responses were recorded. Thoracic dorsal root stimulation regularly elicited gastric contractions after pretreatment with guanethidine and/or hexamethonium which were readily blocked by atropine. It was established that these gastric motor responses were due to peripheral stimulation of the dorsal root and not to spread to the ventral roots. Together with other results (Delbro & Lisander 1980) the findings suggest an antidromic activation of

Brattleboro rats, homozygous for diabetes insipidus, and Long-Evans rats were anaesthetized with urethane, and antidromically identified neurons were recorded from the supraoptic nucleus. Phasically firing neurones were studied during repeated electrical stimulation of the neural stalk, whereby most supraoptic neurones, but not the recorded neurone, were activated antidromically. Such stimulation consistently modified the discharge pattern of phasic neurones in Long-Evans rats, but was relatively ineffective in Brattleboro rats. These results suggest that the effects of neural stalk stimulatio

Electrically induced bulbocavernosus (BC) reflexes were investigated in 47 patients with benign prostatic hypertrophy and the possible harmful effects of prostatectomy on the pudendal nerve branches were followed up in 30 of the patients using the same method. BC reflex obtained by electrical stimulation of the glans penis did not differ significantly from the normal value in bulbocavernosus and external urethral sphincter muscles whether or not patients had been operated on. It was concluded that there is no reason to believe in the existence of somatic involvement of the pudendal nerve branc
