Brunner's glands are located in the submucosa of the proximal duodenum and are unique to mammalian species. The North American opossum (Didelphis virginiana) is generally regarded as a prototype marsupial that closely resembles fossil didelphids which can be placed at the beginning of mammalian evolution. The current investigation provided an opportunity for the analysis of secretory products from these glands in a species thought to be more closely related to earlier evolutionary forms. Extracts of Brunner's glands were subjected to SDS-PAGE and Western blotting. The results indicate the pres

Most nonmammalian tetrapods exhibit more than three phalanges in their posterior digits (III-V), whereas mammals are restricted to no more than three. So far, the molecular mechanism underlying this evolutionary reduction and the functions of Lhx2 and Lhx9 in digit development remain unclear. Here we show divergent evolution of Lhx2 and Lhx9 expression patterns in embryonic limbs across tetrapods. We further demonstrate that although ectopic overexpression of each gene in chick embryos delayed limb skeletal development, only Lhx9 reduced the phalanx number in the posterior digits. Transcriptom

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies, with limited therapeutic success and a persistently low 5-year survival rate. Despite significant advances in genomics and tumor biology, a fundamental challenge persists: to identify the elusive transformation from common benign pancreatic lesions to occasional malignant cellular identity. This review addresses a critical missing link in PDAC pathogenesis, focusing on when and where the switch to malignancy occurs, and why surgical intervention is often insufficient. We explore the biological and spatial-temp

Progression in understanding the relationships among cardiovascular, kidney, and metabolic diseases necessitates reappraising these concepts. Here, this narrative review explains the evolution of the ideas behind cardiovascular-kidney-metabolic syndrome (CKMS), focusing both on the impact of kidney disease on the cardiovascular system and metabolic syndrome and, conversely, on the effects of metabolic syndrome on cardiovascular and kidney diseases. Merging these concepts has resulted in a holistic approach more pertinent to managing the increased pressure from civilization diseases. In light o

Marine ferromanganese (Fe-Mn) oxides serve as both reactive interfaces and long-term archives for oceanography and environmental changes on the Earth. These oxides are ubiquitous in the ocean and provide reactive mineral surfaces that control elemental abundances and isotope ratios in seawater. Recent synchrotron-based spectroscopy and high-precision isotope analyses have significantly improved the molecular-scale understanding of elemental partitioning and isotope fractionation, which establishes a basis for understanding the relationships between minerals and seawater. The economic importance of the sea urchin Glyptocidaris crenularis drives the need to elucidate the genetic mechanisms controlling its sex differentiation and development, a key step for enabling targeted genetic improvement. This study presents a chromosome-scale genome assembly for G. crenularis characterized by 22 chromosome-length scaffolds and a total length of 787.45 Mb. The phylogenetic tree revealed that G. crenularis diverged early among sea urchins. Synteny analysis among species suggested that superscaffold 10 might be the sex chromosome. Subsequent transcriptomic profiling further

The influence of plate tectonics on microbial distributions remains poorly understood. Here, we demonstrate that the global biogeography of hydrothermal vent-endemic chemoautotrophic microbiota is structured by the tectonic history of the global major oceans. These microbiota, particularly anaerobic ones, are significantly more abundant in early-origin Pacific, Arctic, and Mediterranean oceans, whereas they are notably scarce in late-formed Atlantic and Indian Oceans. This pattern was attributed to the timing of ocean formation and its interplay with global redox evolution. Fully oxygenated co

Antimicrobial resistance (AMR) is a systemic global threat, frequently described as a silent pandemic due to the challenges in attributing mortality to drug-resistant infections. Addressing this crisis necessitates a paradigm shift from viewing AMR as a linear consequence of antibiotic exposure to understanding it as an emergent property of ecological systems and metabolic plasticity. The ASM-IISc Symposium on the One Health Approach to AMR brought together a multidisciplinary cohort to deliberate on the drivers and solutions for resistance at the intersection of human, animal, and environment

Chemotherapy remains a key component of the arsenal of tools to fight malaria. Specifically, new drugs with diverse mechanism(s) of action are required to combat existing drug resistance. Here, we describe comprehensive studies to determine the molecular target(s) of gamhépathiopine, a thienopyrimidinone showing promise for the treatment of malaria. In vitro evolution of gamhépathiopine resistance and whole genome analyses identified mutations within the QO site of Plasmodium falciparum cytochrome b, part of complex III of the electron transport chain. Subsequent biochemical assays demonstrate

Endolysosomes are dynamic organelles that undergo movement along the cytoskeleton, fusion, fission, and tubulation during their lifetime. These processes are regulated by complex molecular machineries, including the structurally related hetero-octameric complexes BLOC-1 and BORC. BLOC-1 associates with early endosomes to mediate the biogenesis of lysosome-related organelles (LROs), such as melanosomes and platelet dense bodies. Accordingly, mutations in BLOC-1 subunits cause Hermansky-Pudlak syndrome (HPS), a disorder characterized by pigmentation defects and bleeding abnormalities. In contras

Despite the power conversion efficiency (PCE) of organic photovoltaics (OPVs) exceeding 20%, their commercialization remains hindered by inadequate operational stability. The spontaneous evolution of the phase-separated nanostructures of donor-acceptor blends in the active layer toward equilibrium causes aging of the devices. Here, we report that confining the structures in an ultrathin active layer creates a higher energy barrier for the structural fluctuations, thereby resisting device aging. Specifically, nanoconfinement elevates the glass transition temperature (Tg) by 12 °C and the energy

To unveil the molecular players that maintain neural stem cell (NSC) homeostasis, we conducted a genetic screen in Drosophila and isolated an uncharacterized gene that we named Inseparable (Insep). Insep is the Drosophila homologue of human IER3IP1, a gene associated with Microcephaly, Epilepsy, and Neonatal Diabetes Syndrome-1 (MEDS-1). We show that Insep loss leads to early larval lethality with small brains and these phenotypes can be rescued by expressing IER3IP1 indicating that their biological function is conserved through evolution. The Insep deficient neuroblasts fail to complete cytok

The bilaterian through gut with an anal opening is a major evolutionary innovation in animals. It facilitates effective food processing, which allows animals to grow to a larger body size. However, because non-bilaterian animals (such as cnidarians) lack a through gut, the evolution of the anus in bilaterians (such as insects and humans) remains unresolved. The formation of the bilaterian hindgut is driven by the spatial expression of several transcription factors (for example, Caudal and Brachyury) under the control of Wnt signalling. Here we show that this bilaterian 'hindgut' molecular sign

Cerebellar Purkinje cells (PCs), the sole output neurons of the cerebellar cortex, are essential for motor coordination, learning and circuit formation. While functionally diverse, the extent of PC heterogeneity and the molecular drivers of this diversity remain unclear. Using single-cell RNA sequencing, we identified at least 11 molecularly distinct PC subtypes in the embryonic mouse cerebellum. Spatial reconstruction revealed that these subtypes are organized in embryonic patterns that predict key features of adult cerebellar architecture, including longitudinal stripes and lobular specifici

Human cytomegalovirus (HCMV) establishes lifelong latency in humans largely through the evolution of sophisticated immune evasion strategies that allow it to escape recognition and clearance by the host immune system. While significant progress has been made in elucidating the mechanisms underlying HCMV immune evasion, the precise regulation of host immune responses that enables viral persistence remains incompletely understood. In this review, we provide a comprehensive overview of the current knowledge on HCMV immune evasion, with a particular emphasis on the functions of HCMV-encoded genes.
