Conventional and genetic association between migraine and stroke with druggable genome-wide Mendelian randomization.

The genetic relationship between migraine and stroke remains underexplored, particularly in the context of druggable targets. Previous studies have been limited by small sample sizes and a lack of focus on genetic-targeted therapies for these conditions. We analyzed the association and causality between migraine and stroke using multivariable logistic regression in the UK Biobank cohort and Mendelian randomization (MR) analyses based on genome-wide association study (GWAS) data. Integrating expression quantitative trait loci (eQTLs) data from blood and brain regions, we explored the phenotypic…

Bidirectional Mendelian randomization study reveals interplay between multisite chronic pain and Post-traumatic stress disorder.

To determine the causal relationship between multisite chronic pain (MCP) and post-traumatic stress disorder (PTSD) using Mendelian Randomization (MR) analysis. Genome-wide summary statistics for MCP and PTSD were obtained. Linkage disequilibrium score regression (LDSC) analysis was used to assess genetic correlation. Independent SNPs associated with MCP and PTSD were used as instrumental variables for forward and reverse MR analyses. The inverse variance weighted (IVW) method was the primary analysis, with additional sensitivity tests to ensure robustness. LDSC identified a significant geneti

Exploring the genetic correlation and causal relationships between breast cancer and meningioma using bidirectional Mendelian randomization.

Previous studies have indicated a significantly higher prevalence of breast cancer (BC) among female patients with meningioma compared to the general female population. Therefore, this study aimed to assess the causal relationship between BC and meningioma at the genetic level. Genetic instrumental variables (IVs) for BC were identified from the Breast Cancer Association Consortium (BCAC), the Discovery Biology and Risk of Inherited Variants in Breast Cancer Consortium (DRIVE), the Collaborative Oncological Gene-environment Study (iCOGS), and 11 other BC genome-wide association studies (GWAS).

Gut microbiota and diabetic neuropathy/peripheral artery disease: A two-sample Mendelian randomization study investigating risk factors for diabetic foot ulcers.

Diabetic foot ulcers (DFUs) represent a significant complication affecting the lower extremities in individuals with diabetes. The development and progression of DFUs are primarily influenced by diabetic neuropathy (DN) and diabetic peripheral artery disease (DPAD). Recent studies indicate a novel link between the gut microbiota and these risk factors. However, it remains unclear whether a causal relationship exists between them. We obtained data on gut microbiota, derived from publicly available genome-wide association studies. Additionally, we included data on DN, diabetic polyneuropathy (DP

Exploring the early drivers of pain in Parkinson's disease.

Pain is a common and complex non-motor symptom in people with Parkinson's disease (PWP). Little is known about the genetic drivers of pain in PWP, and progress in its study has been challenging. Here, we conducted two genome-wide association studies (GWAS) to identify genetic variants associated with pain experienced during the earliest stages of Parkinson's disease. The study population consisted of 4,159 PWP of European ancestry who were mapped to five previously-described, longitudinal pain trajectories. In the first GWAS, the extreme pain trajectories (highest burden versus no significant

Integration of Dose Surface Maps and Genetic Data Identifies the Lower Posterior Rectum as a Key Region for Toxicity after Prostate Cancer Radiotherapy.

PURPOSE: Genome-wide association studies are the gold standard for identifying SNP associated with rectal toxicity after prostate cancer radiotherapy. However, they often neglect the radiotherapy dose distribution, which is a key contributor to toxicity risk. Here, we combined rectal dose surface maps with genetic data to identify rectal regions in which variants influence dose-toxicity relationships. EXPERIMENTAL DESIGN: Data were analyzed from 1,293 patients with prostate cancer from the REQUITE study. Deep learning rectum contouring ensured consistent segmentation, and rectum lengths were
