The safety profiles of avacopan on microscopic polyangiitis and granulomatosis with polyangiitis: a real-world pharmacovigilance analysis.

BACKGROUND: Avacopan is an oral selective C5a receptor inhibitor. It received FDA approval in October 2021 for use with standard glucocorticoid therapy to treat antineutrophil cytoplasmic antibody-associated vasculitis. However, there are limitations to understanding adverse events (AEs) in clinical practice. In this study, we analyzed AEs related to avacopan by mining the FDA Adverse Event Reporting System (FAERS). METHODS: Adverse event reports associated with avacopan were retrieved from the FAERS database covering Q1-2022 to Q4 2024. After data standardization, reports exclusively involvi

Automatic Blob Detection Method for Cancerous Lesions in Unsupervised Breast Histology Images.

The early detection of cancerous lesions is a challenging task given the cancer biology and the variability in tissue characteristics, thus rendering medical image analysis tedious and time-inefficient. In the past, conventional computer-aided diagnosis (CAD) and detection methods have heavily relied on the visual inspection of medical images, which is ineffective, particularly for large and visible cancerous lesions in such images. Additionally, conventional methods face challenges in analyzing objects in large images due to overlapping/intersecting objects and the inability to resolve their

Frequency-dependent modulation of evoked potentials during GPi-DBS in pediatric dystonia.

Deep brain stimulation (DBS) of the globus pallidus internus (GPi) is an established treatment for dystonia, yet the neurophysiological mechanisms underlying frequency-dependent effects remain poorly understood. In this study, we investigate how different GPi-DBS frequencies modulate evoked potential (EP) characteristics within the basal ganglia-thalamo-cortical network in pediatric dystonia patients. We evaluated how increasing stimulation frequency alters EP morphology, focusing on peak-to-peak amplitude (PPa), peak-to-peak duration (PPd), and time to peak (TP). This analysis was motivated b

Depression in Premanifest Huntington's Disease: Aberrant Effective Connectivity of Striatum and Default Mode Network.

BACKGROUND: Depression frequently precedes motor symptoms in Huntington's disease gene expansion carriers (HDGECs), yet the neural mechanisms remain poorly characterized. OBJECTIVE: We investigated effective connectivity between the default mode network (DMN) and striatal regions in HDGECs. METHODS: We analyzed 3-T resting-state functional magnetic resonance imaging data from 98 HDGECs (48.98% females; mean age, 42.82 years). Spectral dynamic causal modeling estimated subject-level connectivity, whereas parametric empirical Bayes determined group-level effective connectivity differences betw

Comparative Analysis of Muscle Elasticity and Pathological Characteristics Between Affected and Unaffected Sides in Postherpetic Neuralgia Patients: Protocol for a Pilot Cohort Trail.

BACKGROUND: Postherpetic neuralgia (PHN) is the most common condition that can develop as a complication after herpes zoster (HZ) infection, characterized by pain that persists for more than 3 months after the initial rash has resolved. In most patients with HZ, the rash appears unilaterally. While the treatment of PHN is primarily focused on neural mechanisms due to HZ's neurotropism nature, recent evidence suggests that muscle tissues within the affected regions may also experience pathological changes that contribute to the pain. These changes could reveal novel therapeutic targets and enha

Numerical approximation of fractional order HIV/AIDS model with treatment using Levenberg-Marquardt backpropagation approach.

HIV specifically targets the immune system's T cells, mainly the CD4+ T cells, leading to a chronic and severe illness characterized by a long incubation period. In this study, we propose a novel nonlinear fractional-order model for analyzing and controlling HIV proliferation dynamics. The model categorizes the HIV-infected population into four distinct compartments: susceptible individuals (S), acutely infected (I1), asymptomatically infected (I2), and individuals with AIDS (A). Mathematical properties like positivity and boundedness provide for model validation. The analytical strategy blend

Cell type-specific and subcellular expression of phospholipid phosphatase-related proteins to modulate lyso-phosphatidic acid synaptic signaling in the developing and adult CNS.

Lysophosphatidic acid (LPA) is a bioactive phospholipid that participates in critical processes in neural development and adult brain function and is implicated in various pathophysiological conditions. Along with its six well-characterized receptors, atypical regulators of LPA signaling have also been suggested, including phospholipid phosphatase-related proteins (PLPPRs). PLPPRs have been mostly studied in the developing brain where they control LPA-dependent axon guidance, cortical network hyperexcitability, and glutamatergic neurotransmission. PLPPR4 and PLPPR3 represent two closely relate
