Evaluation and comparison of three different separation techniques for analysis of retroamide enantiomers and their biological evaluation against h-P2X7 receptor.

The P2X receptors are seven-transmembrane domain G protein-coupled receptors and the 7 subtypes of P2X receptors identified in humans, and named P2X1 to P2X7, are channel receptors whose endogenous ligand is ATP. New antagonists of the P2X7 receptor were developed, since this purinergic receptor was highlighted to be involved in many diseases such as different types of pain, cancer, ischemia, neurodegenerative diseases (including Parkinson's and Alzheimer's diseases) characterized by inflammatory processes. With the aim of evaluate the impact of chirality on the pharmacological activity of a n

A targeted lipidomics approach to the study of eicosanoid release in synovial joints.

INTRODUCTION: Articular tissues are capable of producing a range of eicosanoid mediators, each of which has individual biological effects and may be affected by anti-inflammatory treatment. We set out to develop and evaluate a high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) approach for the simultaneous analysis of multiple eicosanoid lipid mediators in equine synovial fluid (SF), and to illustrate its use for investigation of the in vivo effects of non-steroidal anti-inflammatory drug (NSAID) treatment. METHODS: Synovial fluid samples were obtained from normal jo

Risk of new-incident mental health diagnoses following SARS-CoV-2 infection amongst children and young adults in Singapore: a retrospective cohort study.

BACKGROUND: Mental-health-disorders in children/young adults increased post-pandemic; however, the absence of linkage to COVID-19 registries made direct attribution of psychological sequelae to acute infection challenging. METHODS: Healthcare-claims databases in Singapore were utilised to evaluate rates of new-incident mental-health-disorders in children/young adults aged 6-25 years from January 1st, 2012 to 29th February, 2024. Influence on monthly observed new-incident mental-health-disorders across various pandemic phases was modelled using negative binomial regression, accounting for seas

Normative data for rotational chair considering motion susceptibility.

OBJECTIVE: Rotational Chair Test (RCT) is considered one of the most critical measures for vestibular functionality, which generally includes the sinusoidal harmonic acceleration test (SHAT), velocity step test (VST), and visual suppression (VS). The purpose of this study was to establish normal values for different age groups on the RCT and investigate whether motion susceptibility, such as with a history of motion sickness or migraine, has any effects on test metrics. METHODS: One hundred and nine subjects aged from 20 to 59 years who were free from neurotological and vestibular disorders w

Migration patterns of sympathetic preganglionic neurons in embryonic rat spinal cord.

The displacement of immature neurons from their place of origin in the germinal epithelium toward their adult positions in the nervous system appears to involve migratory pathways or guides. While the importance of radial glial fibers in this process has long been recognized, data from recent investigations have suggested that other mechanisms might also play a role in directing the movement of young neurons. We have labeled autonomic preganglionic cells by microinjections of horseradish peroxidase (HRP) into the sympathetic chain ganglia of embryonic rats in order to study the migration and d

Comparative Wound Healing Processes in Plants and Animals: Bioinspired Strategies for Advancing Regenerative Medicine.

Wound healing is a fundamental biological process essential to maintaining structural integrity and survival across both plant and animal life. Despite the profound evolutionary distance separating these kingdoms, wound healing provides one of those momentous occasions when these biological universes collide, revealing significant evolutionary parallels in the core mechanisms of healing, despite clear molecular and physiological differences. However, two challenges have hindered systematic cross-kingdom comparisons. First, unlike animal wound healing, the major phases of plant wound healing ha

Coupling between sterol and sphingolipid structure in ordered membrane domains.

A hallmark of eukaryotic membranes is the pairing of lineage-specific sterols with characteristic sphingolipid species. Mammalian cell membranes are enriched in both cholesterol and long-chain sphingolipids like sphingomyelin, whereas fungi synthesize ergosterol and very long-chain sphingolipids with sugar-containing head groups. It has been proposed that these two lipid classes co-evolved to support membrane structure and organization. Here we investigated how sterol structure and sphingolipid chain length together control membrane order and phase behavior. In the yeast Saccharomyces cerevisi

Thermal Properties and Ion Redistribution in Nanostructured Single-Ion Polymer Electrolyte Blends Composed of Polyanionic Miktoarm Stars.

Nanostructured single-ion polymer electrolytes offer a promising strategy to decouple ionic transport from mechanical constraints, yet how nanoscale morphology governs ion partitioning and transport remains poorly understood. Here, we investigate single-ion polymer blend electrolytes composed of polyanionic miktoarm star particles dispersed in a low-molecular-weight poly(ethylene oxide) (PEO) matrix. Differential scanning calorimetry reveals composition-dependent crystallization behavior that provides insight into the distribution of Li+ ions between the polyanionic particles and the oligomeri

What makes a safety control "enough" for high-energy hazards? Defining and testing acceptable alternative controls when a Direct Control is not feasible.

INTRODUCTION: Serious injuries and fatalities (SIFs) in construction have remained stubbornly high despite substantial safety efforts. Prior research suggests that these events frequently arise in contexts where controls are absent, ineffective, poorly defined, or inconsistently implemented. Prevention appears to depend less on adding more controls and more on designing controls that are explicitly intended for SIF prevention, usable in the field, and precise enough to support consistent decisions at the point of risk. Within the energy-based safety framework, the concept of Direct Control has

Adaptation of emergency surgical care during successive COVID-19 waves: a single-center analysis of surgically treated acute appendicitis.

BACKGROUND: During the first wave of the COVID-19 pandemic, several studies - including our own - demonstrated a significant decrease in emergency surgical presentations, including acute appendicitis (AA). While these early findings are well documented, little is known about how presentation patterns and clinical outcomes evolved across subsequent pandemic waves. This study examines longer-term trends in AA care across three distinct pandemic phases, highlighting how healthcare systems and patient responses adapted over time within a tertiary surgical setting. METHODS: We conducted a retrospe

Improving Chronological Age Estimation in Children Using the Demirjian Method Enhanced with Transformer and Regression Models.

This study presents a two-phase methodology for estimating chronological age in children using panoramic dental images and deep learning-based feature extraction. The dataset comprised 626 panoramic radiographs from children aged 6.0 to 13.8 years (320 males, 306 females; mean age = 9.88 years). Two expert dentists annotated each radiograph according to the Demirjian stages of seven mandibular teeth. In the first phase, three architectures-ResNet-18, EfficientNetV2-M, and Swin V2 Base-were trained separately for males and females to extract high-dimensional feature representations. Images were

Does resource recovery and utilization improve environmental and economic performance of rural wastewater treatment plants? A case study in Xiamen, China.

Wastewater treatment (WWT) has been recognized as a major source of greenhouse gas emissions. Rural WWT often lags behind urban areas due to insufficient infrastructure construction and lack of funding to maintain operation. Selection of optimal WWT technologies in rural areas requires that environmental impacts and costs are traded-off across the entire lifecycle, including the construction, operation, and demolition phases. This study compared the lifecycle environmental impacts of different rural WWT technologies, incorporating both positive and negative externalities into the cost-benefit

Loop diuretics in anuric hemodialysis patients for the clearance of protein-bound uremic toxins.

BACKGROUND: In chronic kidney disease, the accumulation of protein-bound uremic toxins (PBUTs), such as hippuric acid (HA), p-cresyl sulfate (PCS) and indoxyl sulfate (IS), contributes to systemic toxicity and organ dysfunction. These toxins bind to plasma proteins, primarily albumin, rendering them resistant to clearance by conventional dialysis. This study hypothesizes that loop diuretics, particularly torasemide and furosemide, can displace PBUTs from their albumin-binding sites, increasing their free fraction and enhancing their removal during hemodialysis. METHODS: This pilot multicenter
