In vivo 1H-spectroscopy of human intracranial tumors at 1.5 tesla. Preliminary experience at a clinical installation.

Magnetic resonance spectroscopy (MRS) may contribute to the characterization of intracranial tumors in vivo. Volume selective water suppressed proton spectroscopy offers the possibility to study a number of metabolites in the brain including choline (CHO), creatinine/phosphocreatinine (CR/PCR), N-acetylaspartate (NAA), and lactate. Using the stimulated echo technique we have studied 17 patients with intracranial tumors. In all cases the tumors were classified based on histologic evaluation. The tumor spectra differed considerably from those obtained in healthy brain tissue. The results indicat

PET-modulated fluorescence enhancement of cabergoline: A DFT and time-resolved fluorescence guided analytical strategy.

A novel, green, and highly sensitive spectrofluorimetric approach for the determination of cabergoline (CG) is presented. The method relied on the strategic modulation of the photoinduced electron transfer (PET) process. The native fluorescence of CG is inherently decreased by its tertiary amine moiety. The addition of hydrochloric acid efficiently diminishes this PET pathway ("PET-off"), resulting in a considerable, acid-induced improvement of the intrinsic fluorescence signal at 330 nm (excitation at 280 nm). The suggested mechanism is verified by density functional theory (DFT) calculations

Beyond acid-base dyshomeostasis: Dynamic instability of neuronal lysosomal pH as a pathogenic mechanism and therapeutic target in neurological diseases.

The precise maintenance of the intra-lysosomal acidic microenvironment is vital for neuronal functions, including protein degradation, metabolic regulation, and organelle quality control. This review systematically elucidates the dynamic regulatory network governing neuronal lysosomal pH homeostasis, transcending the classical proton pump-leak model. We highlight a multifaceted system integrating vacuolar-type H+-ATPase (V-ATPase)-driven active proton pumping, proton leakage mediated by channels such as transmembrane protein 175 (TMEM175) and solute carrier family 7 member 11 (SLC7A11), membra

No Evidence for Depletion of Circulating Lymphocyte Populations in Primary Brain Tumor Patients Receiving Radiation Therapy Alone.

PURPOSE: To investigate the impact of radiation therapy (RT) for brain tumors on circulating lymphocyte populations and understand this from a dosimetric perspective. METHODS AND MATERIALS: This prospective study enrolled patients with primary brain tumors treated with either photon or proton RT. To specifically study the effect of RT, only patients without concurrent chemotherapy were eligible. For each patient, blood samples were collected before, during, and after RT, as well as at the first follow-up, for flow cytometry analysis of blood cells. We used a stochastic model to compute blood

Fatigue-Resistant Dithienylarene Photoswitches with Acid-Regulated Thermal Ring Opening.

Dithienylarene photoswitches are an emerging class of diarylethenes, in which photochemical ring closure is accompanied by pronounced changes in aromaticity. This distinctive feature enables new opportunities for applications, including molecular solar thermal energy (MOST) storage, a promising approach within renewable energy technologies. Molecular switches used as MOST systems are designed to absorb solar energy, store it in chemical bonds, and release it as heat. However, fast and efficient on-demand release of energy poses a significant challenge. Herein, we present the synthesis of a pyr

Oxygen-dependent regulation of ion fluxes and intracellular pH in Saccharomyces cerevisiae exposed to acetic acid.

The study of acetic acid (AA) stress in yeast is crucial for understanding the mechanisms of acid stress tolerance and enhancing production efficiency. Although AA toxicity has been extensively studied, the mechanism by which oxygen availability modulates ion flux balance and intracellular proton handling under AA stress remains poorly understood. Here, we investigated the combined effects of AA concentration, acidic pH, and oxygen availability on plasma membrane Na+, K+, and H+ fluxes, intracellular pH, and growth in two Saccharomyces cerevisiae strains with distinct AA tolerance. We show tha

Magnetic sphincter augmentation versus conversion to Roux-en-Y gastric bypass for gastroesophageal reflux disease following sleeve gastrectomy in patients with body mass index <30: A single-center comparative study.

BACKGROUND: Gastroesophageal reflux disease (GERD) affects 20%-30% of patients after sleeve gastrectomy (SG). In patients with a body mass index (BMI) <30 kg/m2, the optimal surgical approach-magnetic sphincter augmentation (MSA) or conversion to Roux-en-Y gastric bypass (RYGB)-remains unclear. METHODS: We retrospectively compared outcomes of MSA (n = 20) and RYGB (n = 25) in patients with BMI <30 kg/m2 and refractory post-SG GERD at King Abdulaziz University Hospital (2019-2024). Inclusion required objective evidence of GERD despite ≥6 months of optimized proton pump inhibitor therapy. The p

Acidic extracellular pH-induced PTH secretion in male mouse parathyroids is OGR1-dependent.

The role of extracellular acidity in regulating PTH secretion in cultured mouse parathyroid glands (PTGs) has not been studied to date, largely because of the technical difficulty of isolating mouse PTGs. We hypothesized that acidic extracellular pH directly stimulates PTH secretion through activation of a proton-sensing receptor, specifically ovarian cancer G protein-coupled receptor 1 (OGR1, also known as GPR68). To test this, we developed a method to reliably identify and isolate PTGs from male mice by administering 5-aminolevulinic acid, which induced selective fluorescence in these glands

Synthesis, crystal structure, computational and solution studies of a new phosphotetradecavanadate salt. Assessment of its effect on U87 glioblastoma cells.

The new benzylammonium (C7H10N) salt of the phosphotetradecavanadate (PV14) anion PV14O429-, (C7H10N)6[H3PV14O42]∙7H2O (1), is synthesized under mild conditions and characterized by a combination of physicochemical techniques such as Fourier transform infrared spectroscopy, powder X-ray diffraction, elemental analyses and cyclic voltammetry. As evaluated by 51V NMR spectroscopy, at milimolar concentrations and pH ∼2.5 the PV14 anions decompose slowly, thus demonstrating kinetic stability, but at pH ∼7 this process takes place much faster. However, in the presence of human serum albumin, the 51

Molecular-level insight into ciprofloxacin adsorption on goethite: I. Approach and non-specific binding.

The fate of the antibiotic ciprofloxacin (CIP) in natural waters can be strongly affected by interactions with nanominerals, such as goethite (GT; α-FeOOH). Using classical molecular dynamics, this study resolved the early stages of CIP adsorption on the four main crystallographic faces of GT nanoparticles, which is otherwise difficult to study experimentally. These early stages are driven by (i) electrostatic attraction, and (ii) the establishment of non-specific bonds between CIP (carboxyl, keto, amine) and GT (surface OH) functional groups. Simulations revealed that the medium-range (<1.5 n

Functionality of the V-type ATPase during asexual growth and development of Plasmodium falciparum.

Vacuolar type ATPases (V-type ATPases) are highly conserved hetero-multisubunit proton pumping machineries found in all eukaryotes. They utilize ATP hydrolysis to pump protons, acidifying intracellular or extracellular compartments, and are thus crucial for various biological processes. Despite their evolutionary conservation in malaria parasites, this proton pump remains understudied. To understand the localization and biological functions of Plasmodium falciparum V-type ATPase, we employed CRISPR/Cas9 to endogenously tag the subunit A of the V1 domain. V1A (PF3D7_1311900) was tagged with a t…

Acid-Resistant and Physiological pH-Responsive DNA Hydrogel Composed of A-Motif and i-Motif toward Oral Insulin Delivery.

An acid-resistant DNA hydrogel that is stable in an extremely acidic environment with pH as low as 1.2 has not been reported before, largely due to the instability of DNA-hybridized structures. To achieve this, adenine (A)-rich and cytosine (C)-rich oligonucleotides are rationally designed and integrated to form copolymers with acrylamide monomers via free-radical polymerization. In an acidic environment (pH 1.2-6.0), the generated copolymers form a hydrogel state, which is cross-linked by parallel A-motif duplex configurations (pH 1.2-3.0) and quadruplex i-motif structures (pH 4.0-6.0) due to

Tale of the Breslow intermediate, a central player in N-heterocyclic carbene organocatalysis: then and now.

N-Heterocyclic carbenes (NHCs) belong to the popular family of organocatalysts used in a wide range of reactions, including that for the synthesis of complex natural products and biologically active compounds. In their organocatalytic manifestation, NHCs are known to impart umpolung reactivity to aldehydes and ketones, which are then exploited in the generation of homoenolate, acyl anion, and enolate equivalents suitable for a plethora of reactions such as annulation, benzoin, Stetter, Claisen rearrangement, cycloaddition, and C-C and C-H bond functionalization reactions and so on. Deprescribing medications that may increase the risk of hepatic encephalopathy: A qualitative study of patients with cirrhosis and their doctors.

BACKGROUND AND AIMS: Multiple medications are associated with an increased risk of incident hepatic encephalopathy. Despite this known risk, medications such as opioids, benzodiazepines, gabapentin/pregabalin, and/or proton pump inhibitors are increasingly prescribed to persons with cirrhosis. Deprescribing is a promising intervention to reduce the burden of hepatic encephalopathy. Given that deprescribing has not been trialed in cirrhosis, we evaluated the barriers and facilitators to safe and successful deprescribing in cirrhosis. METHODS: We conducted, transcribed, and analyzed semi-struct
