Plasmablast Storms: Microbial Drivers of Acute and Chronic Autoimmune Flares.

Autoimmune flares are often accompanied by abrupt surges of circulating plasmablasts-short-lived, high-output antibody-secreting cells generated through extrafollicular B-cell activation in response to microbial cues. Three categories of microbial input appear to repeatedly trigger these "plasmablast storms": latent herpesvirus reactivations (Epstein-Barr virus, cytomegalovirus, human herpesvirus-6, varicella-zoster virus), acute respiratory or gastrointestinal infections including SARS-CoV-2, and chronic oral or gut dysbiosis. Although biologically distinct, these stimuli converge on innate s

Type I interferon signaling promotes early innate control of Borrelia burgdorferi infection.

Borrelia burgdorferi, the causative agent of Lyme disease, elicits a robust type I interferon (IFN-I) response that has been strongly associated with chronic inflammatory manifestations such as Lyme arthritis. Although IFN-I is induced early after infection, its functional contribution during the initial stages of B. burgdorferi infection has remained unclear. Here, we identify a critical protective role for IFN-I signaling in the early control of B. burgdorferi. At 5 days post-infection, mice lacking the IFN-I interferon receptor (IFNAR1) exhibited markedly elevated spirochetal burdens at the

The tuxedo sea urchin Mespilia globulus: A fast-developing and tractable model for genomic and developmental biology.

Sea urchins are pivotal models in cellular and developmental biology, but their biphasic life cycle with an extended larval life limits the ability to study post-metamorphosis and adult characters. Here, we introduce a genomically-enabled model system, the tuxedo urchin Mespilia globulus, which has rapid access to late life stages in sea urchins. We describe how we cultured M. globulus in a landlocked marine facility, raised larvae under artificial conditions and closed their life cycle. We established the experimental tractability of M. globulus: we labelled transcripts by hybridization chain

Aggressive vascularized odontogenic myxoma. Case report and literature review.

INTRODUCTION AND IMPORTANCE: Odontogenic myxoma (OM) is an infrequent benign mesenchymal odontogenic tumor of the jaws composed of rounded and angular cells dispersed in an abundant mucoid stroma. An aggressive and vascularized presentation is unusual. CASE PRESENTATION: A 68-year-old woman was evaluated for a mass located under the palate and extended to the cheek. A large flattened mass covered the entire palate, except the left lateral dental arch, and extended to the cheek as a firmed 6 × 6 × 5 cm mass. An enhancement computed tomography and a magnetic resonance imaging revealed a large,

Evolutionary Dynamics of Transposable Element Activity and Regulation in the Apennine Yellow-Bellied Toad (Bombina pachypus).

Transposable elements (TEs) expansion and accumulation represent one of the main drivers of genomic gigantism. Different host genome silencing mechanisms have evolved to counteract TE amplification, leading to a genomic arms race between them. Nevertheless, the evolutionary relationship between TEs and host genome silencing pathways remains poorly understood. Here, we investigate the activity of TEs and TE-silencing mechanisms in somatic and germline tissues of Bombina pachypus, a 10 Gb anuran genome. Our findings reveal a higher activity of TEs in the gonads compared to the brain, with retrot

Alpha-Lipoic Acid Preserves Testicular Integrity Under 2.45 GHz Electromagnetic Radiation by Restoring Redox and Inflammatory Balance.

Background/Objective: Electromagnetic radiation (EMR) from wireless technologies has raised concerns about male reproductive health. We aimed to evaluate the protective role of alpha-lipoic acid (ALA), a potent antioxidant, against testicular alterations induced by 2.45 GHz EMR. Methods: Twenty-eight adult male rats were randomly divided into four groups: control, EMR, ALA, and ALA+EMR. Animals in the EMR and ALA+EMR groups were exposed to EMR for 2 h/day for 1 month. Testicular tissues were examined histologically, stereologically, and immunohistochemically, while serum samples were analysed

Two illustrative cases of adult Lhermitte-Duclos disease and a systematic review of literature related to surgical management.

BACKGROUND: Lhermitte-Duclos disease is a rare subtype of gangliocytoma, a benign tumor growth in the cerebellum often associated with Cowden syndrome, a sporadic genetic pleomorphic disorder that is inherited in an autosomal dominant manner and caused by a harmful mutation in the PTEN gene. Such a mutation can originate malignant and benign tumors, including dysplastic gangliocytoma of the posterior cranial fossa. METHODS: We present two illustrative cases of Lhermitte-Duclos disease that we encountered and surgically treated during the last few years. We also performed a systematic literatu

Sculpting Solutions: A Case Report of Resection and Reconstruction in an Aggressive Mandibular Juvenile Ossifying Fibroma and Review of Literature.

Juvenile ossifying fibroma (JOF) is a rare, benign, yet locally aggressive fibro-osseous lesion that primarily affects children and adolescents. This case report describes a 10-year-old female patient presenting with a three-month history of progressive swelling on the right lower third of her face, accompanied by trismus. Clinical examination revealed a firm, non-tender swelling extending from the right corner of the mouth to the angle of the mandible, with buccal cortical expansion and vestibular tenderness near the mandibular right second molar. Radiographic imaging demonstrated ill-defined

PD-1 prelimits both the cytotoxic and exhaustion potential in thymic CD8+ T cells and impacts the maintenance of peripheral tumor immunity.

Durable T cell immunity against cancer depends on the continual replenishment of effector CD8+ T cells. Thymic output has been correlated with favorable prognosis in cancer patients across a range of ages, suggesting that the thymus is an important source for replenishing T cells capable of controlling cancer progression. However, the effector potential of thymic mature CD8+ T cells and their regulation have not been clearly defined. In this study, we identified the ability of thymic single positive CD8+ T cells to gain effector potential after thymic selection, but they are subject to the reg

Phase 1/2 study of high-dose palifermin for GVHD prophylaxis in patients undergoing HLA-matched unrelated donor HCT.

AbstractGraft-versus-host disease (GVHD) is a major complication of allogeneic hematopoietic cell transplantation (HCT). Palifermin, a recombinant N-truncated keratinocyte growth factor (KGF), protects epithelial tissues, including the thymus and gut. Although high-dose KGF prevents GVHD in preclinical models, lower doses of palifermin were ineffective in humans. We conducted a phase 1/2 trial evaluating high-dose palifermin for preventing severe chronic GVHD (CGVHD) in matched unrelated donor T-cell replete peripheral blood HCT after reduced-intensity conditioning (RIC). Using a 3+3 design, w

Embolization of Extracranial Arteriovenous Malformations: Interventional Approaches according to the Yakes Classification System.

Extracranial arteriovenous malformations (AVMs) are high-flow congenital malformations characterized by direct communication between arteries and veins. These shunts, depending on their size, can cause severe complications such as ischemic skin ulceration, bleeding, bone destruction, and even cardiac failure. AVMs present the greatest challenge in treatment of vascular malformations, requiring collaboration among interventional radiologists, surgeons, dermatologists, internal medicine specialists, and geneticists. This is due to the complex architecture of AVMs, which are characterized by mult

An abnormal increase in CD26(-)CD28(-) cytotoxic effector CD4 and CD8 T cell populations in patients with systemic lupus erythematosus.

CD26 is a human T cell costimulatory molecule as well as a T cell subset marker, and the increase of CD26+ T cells in inflamed tissues and peripheral blood has been reported in diverse autoimmune diseases. In contrast, our group has previously shown that levels of circulating CD26+ T cells are decreased in patients with systemic lupus erythematosus (SLE), although the role of reduced CD26 T cell surface expression in SLE pathology remains to be elucidated. In the present study, we conducted CD26-based T cell subset analyses utilizing peripheral blood mononuclear cells from 57 SLE patients and

T follicular helper 1 cells in blood potentially mirror salivary gland-infiltrating T cells in Sjögren's disease.

Understanding the intricate relationship between peripheral blood immune profiles and the inflammatory environment within affected tissues is pivotal for uncovering mechanisms driving autoimmune diseases. This study aimed to characterize CD4+ T cell subsets in peripheral blood that mirror the immunological activation of labial salivary glands (LSG) infiltrating T cells in primary Sjögren's disease (pSjD). Using multicolor flow cytometry and T cell receptor (TCR) sequencing, we identified CXCR3+CXCR5+ T follicular helper 1 (Tfh1) cells as significantly elevated in the circulation of pSjD patien

Cellular landscape of adrenocortical carcinoma at single-nuclei resolution.

Adrenocortical carcinoma (ACC) is a rare yet devastating tumour of the adrenal gland with a molecular pathology that remains incompletely understood. To gain novel insights into the cellular landscape of ACC, we generated single-nuclei RNA sequencing (snRNA-seq) data sets from twelve ACC tumour samples and analysed these alongside snRNA-seq data sets from normal adrenal glands (NAGs). We find the ACC tumour microenvironment to be relatively devoid of immune cells compared to NAG tissues, consistent with known high tumour purity values for ACC as an immunologically "cold" tumour. Our analysis i

Autonomic neuronal modulations in cardiac arrhythmias: Current concepts and emerging therapies.

The pathophysiology of atrial fibrillation and ventricular tachycardia that result in cardiac arrhythmias is related to the sustained complicated mechanisms of the autonomic nervous system. Atrial fibrillation is when the heart beats irregularly, and ventricular arrhythmias are rapid and inconsistent heart rhythms, which involves many factors including the autonomic nervous system. It's a complex topic that requires careful exploration. Cultivation of speculative knowledge on atrial fibrillation; the irregular rhythm of the heart and ventricular arrhythmias; rapid oscillating waves resulting f

Extant and extinct bilby genomes combined with Indigenous knowledge improve conservation of a unique Australian marsupial.

Ninu (greater bilby, Macrotis lagotis) are desert-dwelling, culturally and ecologically important marsupials. In collaboration with Indigenous rangers and conservation managers, we generated the Ninu chromosome-level genome assembly (3.66 Gbp) and genome sequences for the extinct Yallara (lesser bilby, Macrotis leucura). We developed and tested a scat single-nucleotide polymorphism panel to inform current and future conservation actions, undertake ecological assessments and improve our understanding of Ninu genetic diversity in managed and wild populations. We also assessed the beneficial impa

Modulation of Carnitine Palmitoyl Transferase 1b Expression and Activity in Muscle Pathophysiology in Osteoarthritis and Osteoporosis.

In the pathophysiology of osteoarthritis and osteoporosis, articular cartilage and bone represent the target tissues, respectively, but muscle is also involved. Since many changes in energy metabolism occur in muscle with aging, the aim of the present work was to investigate the involvement of carnitine palmitoyl transferase 1b (Cpt1b) in the muscle pathophysiology of the two diseases. Healthy subjects (CTR, n = 5), osteoarthritic (OA, n = 10), and osteoporotic (OP, n = 10) patients were enrolled. Gene expression analysis conducted on muscle and myoblasts showed up-regulation of CPT1B in OA pa

Endoneurosurgical Resection of Parenchymal and Intraventricular Lesions Using Tubular Retraction System.

OBJECTIVE: Endoscopic surgery has emerged in the recent years as an alternative to the conventional microsurgical approaches for removal of the deep-seated brain and intraventricular tumors. Endoport has enhanced the tumor access and visualization without any significant brain retraction. In this chapter, we describe the surgical technique of the endoscopic excision of the deep-seated intra-axial brain tumors using tubular retraction system with review of the literature. METHODS: The endoscopic endoport technique that we use at our institution for the surgical management of intraventricular a
